The electrophysiological properties of any plant high-affinity Pi transporter have not been described yet. Here, we report the detailed characterization of electrophysiological properties of the barley Pi transporter, HvPHT1;1 in Xenopus laevis oocytes. A very low Km value (1.9 µm) for phosphate transport was observed in HvPHT1;1, which falls within the concentration range observed for barley roots. Inward currents at negative membrane potentials were identified as nH+:Pi- (n , 1) co-transport based on simultaneous Pi radiotracer uptake, oocyte voltage clamping and pH dependence. In this study, we cloned and characterized a bHLH transcription factor, OsPTF1, which is responsible for tolerance to Pi starvation in rice. Overexpression of OsPTF1 can enhance tolerance to Pi deficiency. It was reported that PHO4, which was also a bHLH protein, acts as a key regulator of PHO regulon in yeast. This study functionally characterizes the Arabidopsis (Arabidopsis thaliana) plastidial glycolytic isoforms of glyceraldehyde-3-phosphate dehydrogenase (GAPCp) in photosynthetic and heterotrophic cells. We expressed the enzyme in gapcp double mutants (gapcp1gapcp2) under the control of photosynthetic (Rubisco small subunit RBCS2B [RBCS]) or heterotrophic (phosphate transporter PHT1.2 [PHT]) cell-specific promoters. Mutations in the pyrophosphate (PPi) transporter ANKH cause the autosomal dominant form of craniometaphyseal dysplasia (CMD), a rare disorder characterized by hyperostosis of craniofacial bones and widened metaphyses in long bones. We have generated a knock-in (KI) mouse model expressing a deletion mutation (Phe377del) in the Ank gene, the most common mutation found in our CMD patients. AnkKI/KI mice replicate many CMD features including increased radiopacity of craniofacial bones; undertrabeculated metaphyses in club-shaped femurs; narrowing of cranial neural foramina; fusion of middle ear bones. In vivo skeletal analyses show that AnkKI/KI mice have increased osteoblast and ostseoclast numbers, increased bone turnover serum markers, however, no changes in bone formation and mineral apposition rate. Moreover, femoral bones from AnkKI/KI mice are hypomineralized. Consistently, AnkKI/KI osteoblast cultures show significantly reduced mineral nodules. Previous studies have found that some phosphate (Pi) starvation inducible transporter genes are downregulated and arbuscular mycorrhizal (AM) inducible Pi transporter genes are upregulated in maize roots associated with the fungus Glomus intraradices. However, little is known about the functional diversity of different AM fungal species in influencing the expression of Pi transporters in maize roots. Here, we studied the expression of two Pi transporter genes ZEAma:Pht1;3 (Pi starvation inducible) and ZEAma:Pht1;6 (AM inducible) in maize root colonized by different AM fungal inoculants. In order to study and compare the phosphate transporter gene codon usage and its respond to the traits like salt tolerance, day length, Pollination and temperature in different plants, 100 isoform from 10 plants are extracted from NCBI website and then analyzed with Gene Infinity and Minitab 16 software. The result shows that the highest codon usage similarity (81.95%) was for wheat and oryza (annual, self-polinated and Psychrophilic) from Poaceae family. The result for poaceae family shows that the highest mean abundance was for codons that have U or G at the end. In this study Cucurbita maxima (salt tolerance, annual and cross pollinated) have the lowest codon usage similarity (70.37%) in compare with other plants in this study. The highest difference between families was for Fabaceae (77.64%) but they are divided in one group at the cluster. The histone deacetylase Rpd3p functions as a transcriptional repressor of a diverse set of genes, including PHO5. Here we describe a novel role for RPD3 in the regulation of phosphate transporter Pho84p retention in the cytoplasmic membrane. We show that under repressing conditions (with P(i)), PHO5 expression is increased in a pho4Delta rpd3Delta strain, demonstrating PHO regulatory pathway independence. However, the effect of RPD3 disruption on PHO5 activation kinetics is dependent on the PHO regulatory pathway. Upon switching to activating conditions (without P(i)), PHO5 transcripts accumulated more rapidly in rpd3Delta cells. This more rapid response correlates with a defect in phosphate uptake due to premature recycling of Pho84p, the high-affinity H+/PO4(3-) symporter.
