The chito-oligosaccharide-specific lectin from pumpkin (Cucurbita maxima) phloem exudate has been purified to homogeneity by affinity chromatography on chitin. After SDS/PAGE in the presence of 2-mercaptoethanol, the pumpkin phloem lectin yielded a single band corresponding to a molecular mass of 23.7 kDa, whereas ESI-MS (electrospray ionization MS) gave the molecular masses of the subunit as 24645 Da. Analysis of the CD spectrum of the protein indicated that the secondary structure of the lectin consists of 9.7% α-helix, 35.8% β-sheet, 22.5% β-turn and 32.3% unordered structure. Saccharide binding did not significantly affect the secondary and tertiary structures of the protein. The haemagglutinating activity of pumpkin phloem lectin was mostly unaffected in the temperature range 4-70 °C, but a sharp decrease was seen between 75 and 85 °C. A flow-sensitive nuclear magnetic resonance (NMR) microimaging technique was applied to measure directly the in-vivo water flow in 6-d-old castor bean seedlings. The achieved in-plane resolution of the technique allowed discrimination between xylem and phloem water flow. Both the xylem- and the phloem-average flow velocities in the intact seedling could be quantified. Furthermore, the total conductive cross-sectional area of the xylem vessels and the phloem sieve elements could be determined using the non-invasive and non-destructive NMR microimaging technique. The pressure flow hypothesis introduced by Ernst Münch in 1930 describes a mechanism of osmotically generated pressure differentials that are supposed to drive the movement of sugars and other solutes in the phloem, but this hypothesis has long faced major challenges. This paper presents the basic principle of and initial results from a novel instrumentation system that uses a combination of electrostatic and digital imaging based sensors in order to achieve these goals. An inferential approach is adopted for the mass flow measurement of particles where velocity and volumetric concentration of particles are measured independently. The velocity of particles is determined by cross correlating two signals derived from a pair of electrostatic sensors, whilst the volumetric concentration of particles is obtained from a digital imaging sensor. APHIS is proposing to remove the domestic quarantine regulations for the emerald ash borer (EAB). Eliminating this regulation is in keeping with USDAs goal of reducing regulations that have outlived their usefulness. The proposal would end APHIS domestic regulatory activities, which includes actions such as issuing permits, certificates and compliance agreements, making site visits, and conducting investigations of suspected violations, and instead direct all available resources toward managing the pest. Using population genetic approaches, scientists determined that to optimize conservation of genetic diversity, insecticide application efforts should focus on treating many sites with at least 10 trees per site rather than treating many trees within a single site. The results also highlight how valuable it is to select the correct sites for treatment, with certain sites holding more genetic diversity than others. The effects of chronic exposure to ozone on leaf anatomy, cell ultrastructure and crop yield were studied. Severe cell damage was found, even at ambient ozone levels, mainly affecting the spongy parenchyma and areas near the stomata. Damage to the cell wall caused loss of cell contact, and loss of turgor pressure due to tonoplast disintegration, contributed to cell collapse. Phloem sieve plates were obstructed by callose accumulation, and damaged mesophyll cells increased their starch stores. Tuber yield fell sharply (24-44%), due to the biggest tubers becoming smaller, which affected commercial yield. The results of quantitative reverse transcription-polymerase chain reaction analysis revealed that NaKR3 was induced by high salinity and osmotic stresses, but not by Cu2+ stress.
