Studies were undertaken to determine the extent and rate of binding of Hg-203 labeled phenylmercuric acetate and mercuric ace tate in rat kidney and liver slices and their subcellular fractions after 37°C incubation of the slices in Krebs-Ringer -phosphate solutions containing the mercurials at 10⁻⁴ molar. A fast and parallel rate of uptake of both mercurials in kidney slices between 1 to 3 hour periods was observed. The uptake of the two mercurials into liver slices was much less than that found in kidney slices, The binding rate of phenylmercuric acetate was almost double the rate of inorganic mercury. The subcellular fractions (nuclear, mitochondrial, microsomal, and soluble) of the tissue slices were prepared by homogenizing in 0.25 molar sucrose with subsequent differential centrifugation. Even though the two mercurials showed similar binding in kidney slices, it was found that phenylmercuric acetate was bound to almost twice the extent that of inorganic mercury in the mitochondria, ...

Tissue inhibitor of metalloproteinases (TIMP)-2 forms a noncovalent complex with the precursor of matrix metalloproteinase 2 (proMMP-2, progelatinase A) through interaction of the C-terminal domain of each molecule. We have isolated the proMMP-2-TIMP-2 complex from the medium of human uterine cervical fibroblasts and investigated the processes involved in its activation by 4-aminophenylmercuric acetate (APMA). The treatment of the complex with APMA-activated proMMP-2 by disrupting the Cys73-Zn2+ interaction of the zymogen. This is triggered by perturbation of the proMMP-2 molecule, but not by the reaction of the SH group of Cys73 with APMA. The activated proMMP-2 (proMMP-2*) formed a new complex with TIMP-2 by binding to the N-terminal inhibitory domain of the inhibitor without processing the propeptide. Thus the APMA-treated proMMP-2*-TIMP-2 complex exhibited no gelatinolytic activity. In the presence of a small amount of free MMP-2, however, proMMP-2* in the complex was converted into the 65 kDa MMP

Phenylmercuric acetate is an organomercury compound. This compound was formerly used as a preservative in paints, and as a disinfectant. When applied to the leaves of plants, it is an antitranspirant. Phenylmercuric acetate (PMA) has been used against antifungal activity in agriculture and in leather processing and has known to work well. PMA has also been used as an additive in eye-drops and paint in order to preserve them, as a disinfectant, and as a catalyst in polyurethane systems. In the 1950s-1970s, PMA was used as a catalyst in the 3M Tartan brand polyurethane flexible floors that were commonly in school gymnasiums. School officials worried that mercury vapor was being released into air above the surface of these floors. They had to be carefully removed since mercury is such a toxic heavy metal. The study on PMA as an anti-fungal is the most studied topic for this compound. It has been found that plants treated with PMA transpired more due to the retarded stomatal closure in the plants. ...

Migration of lymphocytes into inflammatory sites requires their adhesion to the vascular endothelium and subendothelial extracellular matrix (ECM). The ensuing penetration of the ECM is associated with the expression of ECM-degrading enzymes, such as endo-beta-D glucuronidase (heparanase), which cleaves heparan sulfate (HS) proteoglycans. We now report that, depending on the local pH, a mammalian heparanase can function either as an enzyme or as an adhesion molecule. At relatively acidified pH conditions, heparanase performs as an enzyme, degrading HS. In contrast, at the hydrogen ion concentration of a quiescent tissue, heparanase binds specifically to HS molecules without degrading them, and thereby anchors CD4+ human T lymphocytes. Thus, the local state of a tissue can regulate the activities of heparanase and can determine whether the molecule will function as an enzyme or as a proadhesive molecule. ...

With the additional insights I gained at a pain management clinic, I learnt to manage my pain and be unafraid of re-injury. Using a combination of medication, physiotherapy and a new attitude to pain, I gradually resumed normal function and quality of life.. However, my employer resisted my efforts to return to work because I couldnt return to my former position, and my superannuation fund deemed me to be totally and permanently incapacitated for full-time employment.. In 2009 I established a consumer health charity for people suffering from persistent pain, the Australian Pain Management Association (APMA).. Prior to APMA, there was no or very limited community-based support for people living with chronic pain.. In the past four years, APMA has grown into a powerful support network and lobby group, and is now an Australia-wide organisation.. As CEO of APMA, I work a 50-hour week, including nights and weekends, in order to help others regain their lives, despite the pain (theirs and ...

Our research encompasses basic cell biology of tumor cell invasion, focusing on two tyrosine kinases, c-Src and Syk, with opposing roles in cancer metastasis. A major focus is invadopodia, the cell surface membrane protrusions formed during tumor cell invasion, and their role in invasion and metastasis. We use cortactin, a Src substrate, and MT1-MMP, an extracellular matrix-degrading, membrane protein, to identify and study the formation of invadopodia and acquisition of proteolytic activity. High resolution light and electron microscopy techniques are used to identify these structures and examine the membrane-associated signal transduction events that occur in conjunction with the formation of invadopodia. In vitro and in vivo assays are used to measure the consequent matrix adhesion, proteolysis and phagocytosis that are mediated by invadopodia. We also focus on determining the mechanism of breast cancer tumor suppression by the Syk tyrosine kinase. Utilizing mouse model systems, 3D culture ...
Numerous transmembrane proteins, including the blood pressure regulating angiotensin converting enzyme (ACE) and the Alzheimers disease amyloid precursor protein (APP), are proteolytically shed from the plasma membrane by metalloproteases. We have used an antisense oligonucleotide (ASO) approach to delineate the role of ADAM10 and tumour necrosis factor-α converting enzyme (TACE; ADAM17) in the ectodomain shedding of ACE and APP from human SH-SY5Y cells. Although the ADAM10 ASO and TACE ASO significantly reduced (, 81%) their respective mRNA levels and reduced the α-secretase shedding of APP by 60% and 30%, respectively, neither ASO reduced the shedding of ACE. The mercurial compound 4-aminophenylmercuric acetate (APMA) stimulated the shedding of ACE but not of APP. The APMA-stimulated secretase cleaved ACE at the same Arg-Ser bond in the juxtamembrane stalk as the constitutive secretase but was more sensitive to inhibition by a hydroxamate-based compound. The APMA-activated shedding of ACE ...
The test measures the activity of alfa-N-acetylgalactosaminidase (nagalase) in blood. Nagalase is an extracellular matrix-degrading enzyme that is (increased) secreted by cancerous cells in the process of tumour invasion. It also is an intrisic component of envelope protein of various virions, such as HIV and the influenza virus. It is secreted from virus-infected cells..1,3,4. Nagalase deglycosylates the vitamin D3-binding protein DBP (in humans better known as Gc-protein). Gc-protein is the precursor for the major macrophage-activating factor (MAF). Gc-protein carries one trisaccharide consisting of N-acetylgalactosamine with dibranched galactose and sialic acid termini. By the deglycosylation the (complete) trisaccharide is removed from the Gc-protein. The glycosylated Gc-protein cannot be converted to MAF anymore.. Normally MAF is produced from Gc-protein by sequential removal of the galactose and sialic acid termini from this protein by respectively beta-galactosidase of inflammation- ...

Dr. Haltermans primary research interest is to improve the delivery of preventive care for underserved children with asthma. Asthma is the most common chronic illness of childhood. While national guidelines recommend daily preventive medications for all children with persistent asthma, studies indicate that many children in the U.S. who should receive these medications are not receiving them. Although young urban children suffer the largest burden from asthma, they are the least likely group to receive adequate preventive care. Thus, there is a substantial amount of suffering that could be prevented with improvements in care. The goal of the Preventive Care Program for Urban Children with Asthma is to develop and test sustainable models for identifying and treating children and adolescents with this all too common disease. ...
Bacterial populations present in Hg-rich environments have evolved biological mechanisms to detoxify methylmercury and other organometallic mercury compounds. The most common resistance mechanism relies on the H+-assisted cleavage of the Hg-C bond of methylmercury by the organomercurial lyase MerB. Although the initial reaction steps which lead to the loss of methane from methylmercury have already been studied experimentally and computationally, the reaction steps leading to the removal of Hg2+ from MerB and regeneration of the active site for a new round of catalysis have not yet been elucidated. In this paper, we have studied the final steps of the reaction catalyzed by MerB through quantum chemical computations at the combined MP2/CBS//B3PW91/6-31G(d) level of theory. While conceptually simple, these reaction steps occur in a complex potential energy surface where several distinct pathways are accessible and may operate concurrently. The only pathway which clearly emerges as forbidden in our

As early as birth, foot complications can present themselves in children. Some complications, such as flatfoot and ingrown toenails, can be inherited or caused in the womb. Others can occur as children begin their walk through life, from the occasional fall to an injury suffered playing a sport. No matter the cause or severity of the problem, the health of a childs feet should not be taken for granted. However, according to an American Podiatric Medical Association (APMA) survey of parents with children under the age of 18, only 35 percent said that they would not be motivated to take their child to a doctors office for foot or leg pain treatment. Furthermore, only 25 percent of those surveyed would take their child to see a podiatrist-a foot and specialist. Member podiatrists from the APMA can help make sure your child stays on his/her feet by offering expert medical diagnosis and treatment options for all issues associated with the feet. From the time a child begins to walk, he/she should be ...
