Studies were undertaken to determine the extent and rate of binding of Hg-203 labeled phenylmercuric acetate and mercuric ace tate in rat kidney and liver slices and their subcellular fractions after 37°C incubation of the slices in Krebs-Ringer -phosphate solutions containing the mercurials at 10⁻⁴ molar. A fast and parallel rate of uptake of both mercurials in kidney slices between 1 to 3 hour periods was observed. The uptake of the two mercurials into liver slices was much less than that found in kidney slices, The binding rate of phenylmercuric acetate was almost double the rate of inorganic mercury. The subcellular fractions (nuclear, mitochondrial, microsomal, and soluble) of the tissue slices were prepared by homogenizing in 0.25 molar sucrose with subsequent differential centrifugation. Tissue inhibitor of metalloproteinases (TIMP)-2 forms a noncovalent complex with the precursor of matrix metalloproteinase 2 (proMMP-2, progelatinase A) through interaction of the C-terminal domain of each molecule. We have isolated the proMMP-2-TIMP-2 complex from the medium of human uterine cervical fibroblasts and investigated the processes involved in its activation by 4-aminophenylmercuric acetate (APMA). The treatment of the complex with APMA-activated proMMP-2 by disrupting the Cys73-Zn2+ interaction of the zymogen. This is triggered by perturbation of the proMMP-2 molecule, but not by the reaction of the SH group of Cys73 with APMA. The activated proMMP-2 (proMMP-2*) formed a new complex with TIMP-2 by binding to the N-terminal inhibitory domain of the inhibitor without processing the propeptide. Thus the APMA-treated proMMP-2*-TIMP-2 complex exhibited no gelatinolytic activity. Phenylmercuric acetate is an organomercury compound. This compound was formerly used as a preservative in paints, and as a disinfectant. When applied to the leaves of plants, it is an antitranspirant. Phenylmercuric acetate (PMA) has been used against antifungal activity in agriculture and in leather processing and has known to work well. PMA has also been used as an additive in eye-drops and paint in order to preserve them, as a disinfectant, and as a catalyst in polyurethane systems. In the 1950s-1970s, PMA was used as a catalyst in the 3M Tartan brand polyurethane flexible floors that were commonly in school gymnasiums. School officials worried that mercury vapor was being released into air above the surface of these floors. They had to be carefully removed since mercury is such a toxic heavy metal. Our research encompasses basic cell biology of tumor cell invasion, focusing on two tyrosine kinases, c-Src and Syk, with opposing roles in cancer metastasis. A major focus is invadopodia, the cell surface membrane protrusions formed during tumor cell invasion, and their role in invasion and metastasis. We use cortactin, a Src substrate, and MT1-MMP, an extracellular matrix-degrading, membrane protein, to identify and study the formation of invadopodia and acquisition of proteolytic activity. High resolution light and electron microscopy techniques are used to identify these structures and examine the membrane-associated signal transduction events that occur in conjunction with the formation of invadopodia. We have used an antisense oligonucleotide (ASO) approach to delineate the role of ADAM10 and tumour necrosis factor-α converting enzyme (TACE; ADAM17) in the ectodomain shedding of ACE and APP from human SH-SY5Y cells. Dr. Haltermans primary research interest is to improve the delivery of preventive care for underserved children with asthma. Asthma is the most common chronic illness of childhood. While national guidelines recommend daily preventive medications for all children with persistent asthma, studies indicate that many children in the U.S. who should receive these medications are not receiving them. Although young urban children suffer the largest burden from asthma, they are the least likely group to receive adequate preventive care. Thus, there is a substantial amount of suffering that could be prevented with improvements in care. The most common resistance mechanism relies on the H+-assisted cleavage of the Hg-C bond of methylmercury by the organomercurial lyase MerB. Although the initial reaction steps which lead to the loss of methane from methylmercury have already been studied experimentally and computationally, the reaction steps leading to the removal of Hg2+ from MerB and regeneration of the active site for a new round of catalysis have not yet been elucidated.
