Considerable insight has been gained on Periplasmic Binding proteins from E.coli as novel candidates to develop sensors for metabolites like glucose, glutamine and lactate. However, these proteins are still used as assays and for them to become sensors and used repeatedly, require immobilization. Additionally, there is not much information on the application of the sensors in combination with innovative sampling technologies. The first part of the research deals with constructing C- terminal Poly histag Glucose/Galactose binding protein and C-terminal Poly histag lactate binding protein and evaluating their sensing properties. Histag serve the purpose of purification and immobilization of these proteins. In Escherichia coli L-arginine is taken up by three periplasmic binding protein-dependent transport systems that are encoded by two genetic loci: the artPIQM-artJ and argT-hisJQMP gene clusters. The transcription of the artJ, artPIQM and hisJQMP genes and operons is repressed by liganded ArgR, whereas argT, encoding the LAO (lysine, arginine, ornithine) periplasmic binding protein, is insensitive to the repressor. Here we characterize the repressible Eσ 70 P artJ, P artP and P hisJ promoters and demonstrate that the cognate operators consist of two 18 bp ARG boxes separated by 3 bp. Determination of the energy landscape of the ArgR-operator contacts by missing contact probing and mutant studies indicated that each box of a pair contributes to complex formation in vitro and to the repressibility in vivo, but to a different extent. Im trying to recall Amirs funniest lines. Some dont translate, some require too much background. But I thought of a few that may illustrate Amirs special humor. When a student of his was called to military reserve duty whenever his oral exam for the completion of his MSc was scheduled, (temporal logic) until he found a note from Amir saying "you had your oral exam in absentia and you passed with flying colors." Or when I interchanged N and M in a paper, on which, upon proof reading, Amir wrote a note "even for N=M" when I claimed I proved something for any N. Or... after long arguments with me that the past operators in temporal logic are not needed, he gave a lecture in which he basically admitted they were useful, with slides whose titles were "Admitting the Past" and "Why Should We be Ashamed of the Past" that I felt were for me. Cation-π interaction is a non-covalent binding force that plays a significant role in protein stability and drug-receptor interactions. In this work, we have investigated the structural role of cation-π interactions in sugar-binding proteins (SBPs). We observed 212 cation-π interactions in 53 proteins out of 59 SBPs in dataset. There is an average one energetically significant cation-π interaction for every 66 residues in SBPs. In addition, Arg is highly preferred to form cation-π interactions, and the average energy of Arg-Trp is high among six pairs. Long-range interactions are predominant in the analyzed cation-π interactions. Comparatively, all interaction pairs favor to accommodate in strand conformations. The analysis of solvent accessible area indicates that most of the aromatic residues are found on buried or partially buried whereas cationic residues were found mostly on the exposed regions of protein. It turns out the Miss Lola had a small hole in her heart (likely a *Patent Ductus Arteriosus...very common in premature babies) and that caused problems with pulmonary hypertension. Where has the time gone? I feel so very blessed to say that Lola is 18 months and doing great! She is a bit behind on some things, but thats to be expected after such a rough start. She will be evaluated in a few days to see if she would benefit from some therapy. At her 18 month well being check, she was not walking and had no words. But about a week after her apt., she finally started walking! I am so proud. Still no words, but that will come. She is sweet and she is sassy! She loves playing with her big brother these days.
