Most bands and DJs tend to get louder as the evening progresses destroying the original percussion balance. A master fader handles that balance problem making everything louder or softer. Dont let anyone move that fader. DJs usually do not have a feel for a musicians balance ending with ear-splitting decibels having the drums tuned out. Without a soundman, someone has to make balance decisions.. The EQ is control of different frequencies. The thump of a nightclub (very low end 80hz) is not the upper range of trumpets (2000k). I usually trim my very low frequencies up 4 or 5 decibels the, 80Hz up 5 to 7 decibels and vocals (1000 to 2000hz) up same. The very high end doesnt make much difference in EQ. I would estimate from the time I started in 1988 to when I left in 1995, the orchestra had acquired about $500,000 worth of equipment. I very strongly felt from the very beginning that it was necessary to create an inventory environment for percussionists at NWS that was professional, and the very best quality that could be achieved. My reasoning was simply that young players coming out of, often poorly equipped, conservatories to NWS should be able to experience being in a section of not only great players, but also the very best and most complete instrument inventory possible. My long-term hope was that they would take that experience to their final jobs and raise the level of awareness and interest in percussion, and not simply accept being in a situation that was unprofessional or inadequate. We are studying a number of TBI models including 1) Ellis Model of Mechanical Cell Injury: cortical neurons and astrocytes are cultured atop a silastic membrane. A mechanical strain is applied to those cells (akin to human TBI) creating a mild, moderate or severe injury. There is an injury dependent physiological response of cultured cells to injury including sodium and calcium influx which leads to cellular pathology and subsequent cell death. 2) Cortical Contusion Injury (CCI): CCI is a model of focal TBI. A small craniotomy is made and an impactor is driven into the cortex. Both depth and rate of impact are controlled to create a range of severities. Outcome measures including cell death and cognitive performance correlate with injury severity. 3) Fluid Percussion Injury (FPI): FPI is a model of diffuse TBI. Virally mediated RNA interference (RNAi) to knock down injury-induced genes could improve functional outcome after traumatic brain injury (TBI); however, little is known about the consequences of gene knockdown on downstream cell signaling pathways and how RNAi influences neurodegeneration and behavior. Here, we assessed the effects of adeno-associated virus (AAV) siRNA vectors that target two genes with opposing roles in TBI pathogenesis: the allegedly detrimental neuronal nitric oxide synthase (nNOS) and the potentially protective glutathione peroxidase 1 (GPx-1). In rat hippocampal progenitor cells, three siRNAs that target different regions of each gene (nNOS, GPx-1) effectively knocked down gene expression. However, in vivo, in our rat model of fluid percussion brain injury, the consequences of AAV-siRNA were variable.
