The relation of collagen synthesis to experimentally induced arteriosclerosis was studied by measuring proline hydroxylase activity. Gross aortic plaques were produced in rabbits by daily injection of epinephrine (intravenous) and thyroxine (intraperitoneal) for 4, 9, or 14 days. Activity of proline hydroxylase was significantly increased after 4 days of treatment and reached a peak, five- to sixfold increase, after 14 days of treatment. The increase in enzyme activity was correlated with the severity of observed arteriosclerosis. Increase in proline hydroxylase activity may be a possible biochemical defect in the aortas of rabbits with arteriosclerosis induced by injury. ...

Induction of cancer specific apoptosis mechanisms. Dear Sirs and Madams,. Sustainable cancer therapy requires new solutions based on new thinking that is fundamentally different from traditional practices.. With these new methods of thinking, the author can postulate a promising route to sustainable cancer therapy. Hereby, cancer-specific membrane receptors are used for the therapeutic application. It is assumed, and there is much to support this contention, that these cancer-specific membrane receptors have structures that are common to all types of cancer.. In this respect, the treatment concept is not only applicable for all types of cancer but also for all stages of development and grading. And both primary tumors and metastases appear equally effectively treatable.. In an effort to understand apoptosis in principle and to reconcile it with the control mechanisms of cell division, the author has succeeded in postulating a therapy concept of cancer via the induction of cancer cell specific ...

TY - JOUR. T1 - The transition metal binding properties of a 3RD generation bleomycin analogue, tallysomycin. AU - Greenaway, Frederick T.. AU - Dabrowiak, James C.. AU - Van Husen, Mark. AU - Grulich, Robert. AU - Crooke, Stanley T.. PY - 1978/12/29. Y1 - 1978/12/29. N2 - Employing a number of physical techniques the transition metal binding site of the bleomycin (BLM) related antibiotic tallysomycin (TLM) has been determined. The new antibiotic was shown to have two metal binding sites. One site is similar to that of bleomycin and involves the pyrimidine-imidazole portion of the molecule. The second binding site, which is thermodynamically less stable than the first site, utilizes the amino group of the L-talose moiety and the amino groups located in the β lysine-spermidine portion of the antibiotic. The presence of two metal binding sites and its implication on the mechanism of action of TLM is also discussed.. AB - Employing a number of physical techniques the transition metal binding site ...

The mode of cell death of two strains of mouse lymphoma LSI78Y cells was studied following photodynamic therapy (PDT) sensitized by chloroaluminum phthalocyanine. Strains LY-R and LY-S differ in their relative sensitivities to UVC radiation, X-radiation, and PDT; both responded to PDT by undergoing apoptosis. The DNA was degraded into fragments with lengths which are multiples of approximately 180-190 base pairs (i.e., oligonucleosome size), a biochemical marker of apoptosis. The DNA fragmentation was dose and time dependent which indicates this response to be an enzymic process related to cell killing. Cycloheximide, a protein synthesis inhibitor, and actinomycin D, an RNA synthesis inhibitor, enhanced the endonucleolytic DNA fragmentation. Transmis sion electron microscopy revealed chromatin condensation around the periphery of the nucleus, which is also characteristic of apoptosis. The induction of apoptosis in L5178Y cells by PDT was rapid, with marked degradation of DNA occurring in as ...

Moreover, the computational studies were carried out for all new compounds. The results indicated that five compounds showed promising binding affinities (10a: − 3.4 kcal/mol, 6d: − 3.0 kcal/mol, 15a: − 2.2 kcal/mol, 3a: − 1.6 kcal/mol, and 21: − 1.3 kcal/mol) against the active site of the epidermal growth factor receptor kinase (EGFR). The cytotoxicity of the potent products 3a, 6d, 10a, 15a, and 21 was examined against human liver carcinoma cell line (HepG-2) and revealed activities close to Doxorubicin standard drug.
E2F is a complex family of transcription factors implicated in different cell fates, including proliferation and apoptosis (Stevens and La Thangue, 2003; Frolov and Dyson, 2004; Polager and Ginsberg, 2008; van den Heuvel and Dyson, 2008). The first family member identified, E2F‐1, physically interacts with the retinoblastoma tumour suppressor protein pRb, which negatively regulates E2F‐1 activity (Bandara and La Thangue, 1991; Zamanian and La Thangue, 1992; Weinberg, 1995; Stevens and La Thangue, 2003; Frolov and Dyson, 2004). While it has been established that E2F‐1 promotes proliferation and therefore is potentially oncogenic, it has also become clear that E2F‐1 is endowed with apoptotic activity (Polager and Ginsberg, 2008; van den Heuvel and Dyson, 2008). Thus, many E2F target genes are connected with apoptosis (Ren et al, 2002), and in Rb−/− mice the enhanced level of tissue‐specific apoptosis reflects deregulated E2F‐1 activity (Tsai et al, 1998; Iaquinta and Lees, 2007). ...

Here, we analyzed the ability of C. vasculum-derived metabolites to inhibit proliferation, induce cytotoxicity, and trigger apoptotic signaling in NSCLC, SCLC, and ovarian carcinoma. It has previously been shown that compounds isolated from this sponge can kill tumor cells of different origin albeit through unknown mechanism of action (19). We show here that the two acetylenic compounds 1 and 2 as well as the parental mixture of these compounds from which they were separated, indeed cause tumor-specific cytotoxicity in NSCLC and SCLC. Importantly, we demonstrate that for both compounds this cytotoxicity is tumor specific, as all normal cells tested representing heart tissue, bronchial and retina epithelium, normal PBMCs, and foreskin or lung fibroblasts remained unaffected at concentrations that were toxic for NSCLC and SCLC. In ovarian carcinoma cells compound 1 but not compound 2 caused tumor-specific cell death. For compound 2, a cytotoxic effect was evident in NSCLC and SCLC whereas in ...
The major findings of the present study are that water-soluble fullerene directly affects vascular ECs to cause cytotoxic injury or cell death and inhibition of cell growth. To the best of our knowledge, this study provides the first demonstration of the direct effects of water-soluble fullerene on vascular endothelium. In other human cells, including dermal fibroblasts, liver carcinoma cells (HepG2), neuronal astrocytes, and T-lymphocytes (Jurkat cells), recent reports have noted that water-soluble fullerene shows cytotoxic effects, presumably via production of reactive oxygen species (22, 24). Notably, several types of water-soluble fullerene derivatives are available [e.g., hydroxyl fullerene used herein, dendritic C60 monoadduct (22), malonic acid C60 (22) and nano-C60 (24) (basically pristine C60)], and cytotoxicity to cells varies depending on the fullerene subtype used, presumably because of surfactant chemistry, including a balance between hydrophobicity and hydrophilicity (3).. An N-formamido pyrrole- and imidazole-containing triamide (f-PIP) has been shown by DNase I footprinting, SPR, and CD studies to bind as a stacked dimer to its cognate sequences: 5-TACGAT-3 (5-flank of the inverted CCAAT box-2 of the human topoisomerase IIalpha promoter) and 5-ATCGAT-3. A gel shift experiment provided evidence for f-PIP to inhibit protein-DNA interaction at the ICB2 site. Western blot studies showed that expression of the topoisomerase IIalpha gene in confluent NIH 3T3 cells was induced by treatment with f-PIP. The results suggested that the triamide was able to enter the nucleus, interacted with the target site within ICB2, inhibited NF-Y binding, and activated gene expression.. ...
TWENTY FIVE Aspergillus isolates were screened from Giza Governorate and Saint Catherine Protectorate soils in Egypt. The antimicrobial activity of the crude extracts was tested against two Gram positive bacteria (Bacillus subtilis NRRL-B-4219, Staphylococcus aureus ATCC29213), four Gram negative bacteria (Alcaligenes faecalis B-170, Escherichia coli ATCC25922, Klebsiella pneumoniae ATCC10131, Pseudomonas aeruginosa ATCC27953), and one yeast (Candida albicans ATCC10231). The antioxidant activity using free radical scavenging model was assayed for the crude extracts. The antitumor activity for all of crude extracts was determined against HCT116 (Colon carcinoma cell line), HEPG2 (Liver carcinoma cell line), and MCF-7 (Breast carcinoma cell line). Aspergillus nomius was the most potent fungal species accordingly, it was chosen for bioactivity assay. Identification of this species was further confirmed at the molecular level based on nuclear ribosomal DNA 18s identities. An accession number, LC199488, was

TY - JOUR. T1 - DNA methylation reduces binding and cleavage by bleomycin. AU - Roy, Basab. AU - Tang, Chenhong. AU - Alam, Mohammad P.. AU - Hecht, Sidney. PY - 2014/9/30. Y1 - 2014/9/30. N2 - In a recent study, we described the enhanced double-strand cleavage of hairpin DNAs by Fe·bleomycin (Fe·BLM) that accompanies increasingly strong binding of this antitumor agent and suggested that this effect may be relevant to the mechanism by which BLM mediates its antitumor effects. Because the DNA in tumor cells is known to be hypomethylated on cytidine relative to that in normal cells, it seemed of interest to study the possible effects of methylation status on BLM-induced double-strand DNA cleavage. Three hairpin DNAs found to bind strongly to bleomycin, and their methylated counterparts, were used to study the effect of methylation on bleomycin-induced DNA degradation. Under conditions of limited DNA cleavage, there was a significant overall decrease in the cleavage of methylated hairpin DNAs. ...

Almost one half of prospective drugs, including new chemical entities are not suitable for clinical use because of their metabolic unstability and for danger of drug interactions. This type of toxicity has been underlying several drug withdrawals in recent years and hence it is needed now to bring with any new compound proposed an information on its metabolism and on its interactions with major drug metabolizing enzyme systems namely with liver microsomal cytochromes P450 (CYP). Then, possibility of drug interactions with other drugs taken by the patient on the basis of drug metabolism may be evaluated to prevent significant losses in time, effort and money in drug development. The analysis of drug metabolism by individual CYPs, the possibility of drug interactions and of induction of individual CYP forms are performed with CYP in vitro systems with the respective substrates, inhibitors and inductors based on the recommendation of the U.S. Food and Drug Administration: ...
Rapid transportation, global trading and improvements in storage have eliminated seasonality from our table, making it possible to get almost any fruit or vegetable at any season. As an example of this, fresh white asparagus produced in Peru is in the stores this winter. Americans favor green asparagus, whereas white (blanched) asparagus seems more popular in Europe. Blanching dates back to at least the 16th century when asparagus forced in cellars became popular with the rich aristocracy. The word blanching is also used in food preservation when vegetables are immersed in boiling water to kill the tissue. But in the gardening sense, blanching is used to describe growing plants in the dark. Asparagus is a labor-intensive crop to produce and blanching requires more of it, so blanched spears cost about 15 percent more than the green form. Blanched plants have a slightly milder taste (some describe it as nuttier) than green asparagus. Chlorophyll forms only in the presence of light so blanched ...

A battery monitor circuit for rechargeable multicell batteries that provides a reliable indication of battery failure independent of temperature, load and age, by continuously comparing the impedance of the cells of the battery against each other. Comparison means are provided responsive to a first voltage at a tapped terminal of the battery that divides the battery cells into two groups, and to a second voltage that represents a fraction of the battery voltage corresponding to the ratio of the number of cells in each group, said comparison means generating an error signal when said first and second voltages change so as to experience a reversal in relative magnitude, which is indicative of cell deterioration.
Bleomycin is a broad-spectrum glycopeptide antitumor antibiotic produced by Streptomyces verticillus. Clinically, the mixture of bleomycin A2 and bleomycin B2 is widely used in combination with other drugs for the treatment of various cancers. As a secondary metabolite, the biosynthesis of bleomycin is precisely controlled by the complex extra-/intracellular regulation mechanisms, it is imperative to investigate the global metabolic and regulatory system involved in bleomycin biosynthesis for increasing bleomycin production. N-acetylglucosamine (GlcNAc), the vital signaling molecule controlling the onset of development and antibiotic synthesis in Streptomyces, was found to increase the yields of bleomycins significantly in chemically defined medium. To mine the gene information relevant to GlcNAc metabolism, the DNA sequences of dasR-dasA-dasBCD-nagB and nagKA in S. verticillus were determined by chromosome walking. From the results of Real time fluorescence quantitative PCR (RT-qPCR) and

Yale researchers have uncovered a new BET inhibitor-a class of drugs usually used to treat blood cancers-that offers promise against a wider variety of cancers, including solid tumors. The new substance, called NHWD-870, was shown in animal studies to be between three and fifty times more potent against cancer cells than existing BET inhibitors. Additionally, while existing inhibitors produce side effects, including fatigue, nausea, loss of taste, and low platelet counts, NHWD-870 showed lower toxicity (as measured through animals body-weight loss). Lead researcher Qin Yan and his colleagues are now pursuing human clinical trials, testing NHWD-870 on melanoma and small-cell lung cancer patients.. ...

A Full-Bridge MMC with External DC Link block is used to feed a 1800-HP induction motor. this block is programmed to model five H-bridges in series. Phase-shifting carrier-based PWM technique is used to control the H-bridges.. On the rectifier side, a phase-shifting transformer and a three-phase diode bridge is used to feed each individual DC link. Using phase-shifting transformer allows significantly reducing current harmonics injected into the distribution grid.. In order to cancel harmonics produced by the five six-pulse rectifier units in each phase, voltages applied to the AC side of individual rectifiers must be phase shifted by 60/5 = 12 degrees with each others. These phase shifts are specified in the PhaseShift parameter defined in the model pre-load function (PhaseShift = [12,24,36,48,60]). ...
