TY - JOUR. T1 - Immunohistochemical distribution of pro-somatostatin-related peptides in cerebral cortex. AU - Morrison, John H.. AU - Benoit, Robert. AU - Magistretti, Pierre. AU - Bloom, Floyd E.. PY - 1983/3/7. Y1 - 1983/3/7. N2 - Mammalian brain contains 3 peptides related to the pro-somatostatin molecule: somatostatin-14 (SS14), the form originally identified from hypothalamic extracts, somatostatin 28 (SS28) and somatostaton 28 (1-12) (SS28 (1-12)). By using antibodies which selectively recognize one or more of these 3 somatostatin-related peptides, we have characterized their immunohistochemical distribution in neocortex. These somatostatin-related peptides have a specific laminar distribution in cortex and are differentially distributed such that SS28 is largely restricted to cell bodies, whereas SS28 (1-12) is preferentially localized in neuroal processes and terminals in a density which far exceeds that revealed by SS-14 immunoreactivity. These data suggest that there may be an ...

MANAGING SUSPECTED TEMPORAL ARTERITIS WITHOUT A TEMPORAL ARTERY BIOPSY Keith Ong, Ophthalmologist, University of Sydney, RNSH, Ryde Hospital, Dalcross Adventist Hospital. Lawrence Tsng Chze Ong, Immunology Registrar, Westmead Hospital. Lillian Beatrice Ong, JMO, Prince of Wales Hospital. We would like to report 2 cases of suspected temporal arteritis which were managed without routine temporal artery biopsy.. The gold standard diagnostic test for temporal arteritis, the temporal artery biopsy (TAB), is highly specific for a diagnosis of temporal arteritis, but is only positive in up to 82% of temporal arteritis cases (with sensitivities reported as low as 20%). This figure has been shown to decrease even further (12%) following corticosteroid therapy.. Although it is a relatively minor procedure, the risks of TAB are not trivial and include facial nerve injury, infection, haemorrhage, incisional alopecia, scar widening and foreign body reaction to entrapped hairs. Furthermore, the need for ...
Full Text - Purpose: This study aimed to validate the newly proposed American Joint Committee on Cancer (AJCC) pathological prognostic staging system for young breast cancer patients (aged ≤40 years). Results: We included 12811 women in this study. Overall, 52.8% of patients in the 7th AJCC stages were restaged to the 8th AJCC pathological staging system, including 10.7% upstaged and 42.1% downstaged. The receiver operating characteristics analysis showed that the new staging system had a better role in predicting breast cancer-specific survival (BCSS) compared with 7th edition staging (P<0.001). The results of the multivariate prognostic analysis showed that the hazard ratio of BCSS increased with the 8th AJCC stages, while the 7th anatomic stages had no significant difference in BCSS.Conclusions: The novel pathological staging system could provide more accurate prognostic stratification for young women with breast cancer because of the high proportion of stage migration.Patients and Methods:

Canine adenovirus (CAV) type 1 and 2, respectively, cause infectious canine hepatitis and infectious canine laryngotracheitis in members of the families Canidae and Ursidae worldwide. Both of these infections are acute diseases, especially in young dogs. The aim of this study was to conduct a serological investigation of canine adenovirus infection. For this purpose, serum samples were collected from native pure-bred Kangal (n = 11), and Akbash dogs (n = 17) and Turkish Greyhounds (n = 15) in Eskisehir and Konya provinces. None of the dogs were previously vaccinated against CAV types. Indirect ELISA detected 88.2 %, 93.3 % and 100 % prevalences in Akbash, Greyhound and Kangal dogs, respectively. The remainder of the samples (n = 51) were collected at the Afyonkarahisar Municipality Shelter. Fourty-two of these ...
Percutaneous needle core biopsy may be the definitive procedure where important diagnostic and prognostic information in acute and persistent renal allograft dysfunction is normally obtained. the promise of improving diagnostic precision and developing fresh, processed molecular pathways for restorative treatment will also be offered. Intro Renal biopsy remains the platinum standard by which essential diagnostic and prognostic info is definitely acquired after kidney transplantation.1,2 Biopsy methodologies have been devised to assess the acceptability of an organ before transplantation and to assess and forecast renal allograft overall performance after implantation. Renal transplant biopsy samples are analyzed from the same traditional and modern techniques as are used to assess samples from native kidneys (Package 1). With this Review, we describe the practical part D-106669 of renal biopsy in the management of renal allograft recipients and spotlight the changes that take place in renal ...
Subcutaneous oedema as a presenting feature of polymyositis/dermatomyositis: a poor prognostic indicator? - Grand Rounds Subcutaneous oedema as a presenting feature of polymyositis/dermatomyositis: a poor prognostic indicator? - Widespread subcutaneous oedema is a rare presenting feature of polymyositis (PM)/dermatomyositis (DM). It was reported in the initial description of the disease by Wagner in 1877 but only nine cases have since been reported in the literature and it is not listed in standard textbooks of rheumatology. We present a further case of subcutaneous oedema as a presenting feature of dermatomyositis, briefly review the existing literature and postulate that this presentation represents a subset of the disease with a poorer prognosis.
BACKGROUNDThe neutrophil-to-lymphocyte ratio (NLR), a marker of inflammation, has been reported to be a poor prognostic indicator in prostate cancer. Here we explore the use of the NLR to establish a simple prognostic score for men with metastatic castration-resistant prostate cancer (mCRPC) treated with docetaxel. METHODS: In the training cohort, the NLR and other known prognostic variables were evaluated among a cohort of chemotherapy-naive patients treated with thrice-weekly docetaxel at the Princess Margaret Cancer Centre. Significant prognostic variables identified by univariable Cox regression were evaluated by the area under the receiver operating characteristic curves. Multivariable Cox regression was then used to derive a prognostic score where 1 risk point was assigned for each significant variable. The model was externally validated in a cohort of patients treated at the Royal Marsden. RESULTS: Three hudred fifty-seven patients were analyzed in the training cohort. Median age was 71 ...

Nasal-type extranodal natural killer/T-cell lymphoma (ENKTCL) originates primarily in the nasal cavity or extra-nasal sites within the upper aerodigestive tract. However, it is unclear whether the primary site can serve as an independent prognostic factor or whether the varying clinical outcomes observed with different primary sites can be attributed merely to their propensities of regional lymph node involvement. The aim of this study was to investigate the prognostic implications of the primary site and regional lymph node involvement in patients with early-stage nasal-type ENKTCL. To develop a nomogram, we reviewed the clinical data of 215 consecutively diagnosed patients with early-stage nasal-type ENKTCL who were treated in Sun Yat-sen University Cancer Center with chemotherapy and radiotherapy between 2000 and 2011. The predictive accuracy and discriminative ability of the nomogram were determined using a concordance index (C-index) and calibration curve. The 5-year overall survival (OS) and

The incidence of oropharyngeal squamous cell carcinoma (OPSCC) has been steadily increasing in the United States and Western Europe, accompanied by a rise in the number of patients with human papillomavirus (HPV)-related OPSCC [1,2]. Patients with HPV-related OPSCC tend to be relatively young and less exposed to tobacco smoking and alcohol [3]. HPV-related OPSCC is now well recognized as a distinct entity presenting with more extensive nodal disease but a better prognosis than HPV-unrelated OPSCC [4-6]. As the 7th edition of the American Joint Committee on Cancer/Union for International Cancer Control (AJCC/UICC) TNM staging system for traditional OPSCC does not accurately reflect the prognosis of HPV-related OPSCC, there have been several attempts to propose a new prognostic classification. The International Collaboration on Oropharyngeal cancer Network for Staging (ICON-S) study group proposed a novel prognostic classification for HPV-related OPSCC based on the outcome of over 1,900 patients ...

Our knowledge of the fine structure of the Human Spiral Ganglion (HSG) is still inadequate and new treatment techniques for deafness using electric stimulation, call for further information and studies on the neuronal elements of the human cochlea. This thesis presents results of analyses of human cochlear tissue and specimens obtained during neurosurgical transpetrosal removal of life-threatening meningeomas. The use of surgical biopsies produced a well-preserved material suitable for ultrastructural and immunohistochemical studies on the human inner ear. The SG was studied with respect to fine structure, using TEM technique and the immunostaining pattern of synaptophysin, which is an integral membrane protein of neuronal synaptic vesicles. The immunostaining patterns of the tight junctional protein ZO-1 and the gap junctional proteins Cx26 and Cx43 in the human cochlea were also studied. The ultrastructural morphology revealed an absence of myelination pattern in the HSG, thus differing from ...

Treatment success was defined as either cardioversion to sinus rhythm or a reduction in the ventricular rate to ≤90 b.p.m. at 24h after the start ...
Fetal Alcohol Spectrum Disorder (FASD) is a prevalent developmental disease that is caused by excess in utero exposure to ethanol. The importance of timing and dosage of ethanol has been elucidated by previous investigations conducted with humans, rodents, and other model organisms, but challenges associated with determining functional deficiencies have necessitated the introduction of a novel animal model for investigating the dynamics of FASD. In this thesis, we propose that the freshwater flatworm planarian species, Schmidtea mediterranea, has the capability to fulfill this need. Planaria have the ability to completely regenerate fragments into intact, functional animals, and in these studies, we exploited this property to characterize nervous system development in the presence of ethanol. Functional testing of head-regenerating planaria included planarian locomotor velocity testing to assess locomotor capabilities and light avoidance testing to assess development of innate negative phototaxic

Background: In this study, we aimed to investigate frequency, patterns, etiologies, and costs of unplanned readmissions after left ventricular assist device implantation.. Methods: Between April 2012 and September 2016, 99 unplanned readmissions of a total of 50 consecutive bridge-to-transplant patients (45 males, 5 females; mean age 46.9±10.3 years; range, 19 to 67 years) who were successfully discharged after left ventricular assist device implantation were retrospectively analyzed. Patient demographic data, hemodynamic measurements before implantation, and readmissions after discharge were recorded. Hospitalizations due to major problems which were unable to be managed in routine outpatient clinic were accepted as unplanned readmissions. Survival analysis was performed.. Results: The readmission rate was 1.7 per year after discharge. Survival of patients who were readmitted within the first 90 days was found to be significantly lower than those without early readmission. The most common ...

Atorvastatin protects GABAergic and dopaminergic neurons in the nigrostriatal system in an experimental rat model of transient focal cerebral ischemia. Biomédica [online]. 2014, vol.34, n.2, pp.207-217. ISSN 0120-4157. http://dx.doi.org/10.7705/biomedica.v34i2.1851.. Introduction: Cerebral ischemia is the third leading cause of death and the primary cause of permanent disability worldwide. Atorvastatin is a promising drug with neuroprotective effects that may be useful for the treatment of stroke. However, the effects of atorvastatin on specific neuronal populations within the nigrostriatal system following cerebral ischemia are unknown. Objective: To evaluate the effects of atorvastatin on dopaminergic and GABAergic neuronal populations in exofocal brain regions in a model of transient occlusion of the middle cerebral artery. Materials and methods: Twenty-eight male eight-week-old Wistar ...
BACKGROUND/AIMS Fascin is an actin-bundling protein that is important in cell motility. Fascin expression has been shown to have a potential role in tumor progression for some epithelial tumors. However, there are only a few studies related to its expression in mesenchymal tumors. We investigated fascin expression in gastrointestinal stromal tumors. METHODS Thirty gastrointestinal stromal tumors, which were very low (n=6), low (n=2), moderate (n=4), and high (n=18) risk, constituted our series. Immunohistochemical expression of fascin was studied in all cases. RESULTS Immunoreactivity was observed in only five cases, all of which were in the high-risk group. The remaining cases (25/30) showed no immunoreactivity, and the difference did not seem statistically important (p=0.261). Fascin expression was stronger in epithelioid cells than spindle-shaped cells (p=0.003). In addition, gastrointestinal stromal tumors in the small bowel showed higher fascin expression than those in the other localizations

Previous research suggests the therapeutic cancer vaccine L-BLP25 potentially provides a survival benefit in patients with locally advanced unresectable stage III non-small cell lung carcinoma (NSCLC). These promising findings prompted the phase III study, INSPIRE, in patients of East-Asian ethnicity. East-Asian ethnicity is an independent favourable prognostic factor for survival in NSCLC. The favourable prognosis is most likely due to a higher incidence of EGFR mutations among this patient population. The primary objective of the INSPIRE study is to assess the treatment effect of L-BLP25 plus best supportive care (BSC), as compared to placebo plus BSC, on overall survival time in East-Asian patients with unresectable stage III NSCLC and either documented stable disease or an objective response according to the Response Evaluation Criteria in Solid Tumors (RECIST) criteria following primary chemoradiotherapy. Those in the L-BLP25 arm will receive a single intravenous infusion of cyclophosphamide (300
Sentinel lymph node metastasis is a common and early event in the metastatic process of head and neck squamous cell carcinoma (HNSCC) and is the most powerful prognostic factor for survival of HNSCC patients. 3-O-acetyloleanolic acid (3AOA), a pentacyclic triterpenoid compound isolated from seeds of Vigna sinensis K., has been reported to have potent anti-angiogenesis and anti-tumor activities. However, its effects on tumor-related lymphangiogenesis and lymph node metastasis are not yet understood. The in vitro inhibitory effects of 3AOA on VEGF-A-induced lymphangiogenesis were investigated via in vitro experiments using mouse oral squamous cell carcinoma (SCCVII) cells and human lymphatic microvascular endothelial cells (HLMECs). The in vivo inhibitory effects of 3AOA on VEGF-A-induced lymphangiogenesis and sentinel lymph node metastasis were investigated in an oral cancer sentinel lymph node (OCSLN) animal model. 3AOA inhibited tumor-induced lymphangiogenesis and sentinel lymph node metastasis in an
Various muscular dystrophies are associated with the defective glycosylation of alpha-dystroglycan and are known to result from mutations in genes encoding glycosyltransferases. Fukutin-related protein (FKRP) was identified as a homolog of fukutin, the defective protein in Fukuyama-type congenital muscular dystrophy (FCMD), that is thought to function as a glycosyltransferase. Mutations in FKRP have been linked to a variety of phenotypes including Walker-Warburg syndrome (WWS), limb girdle muscular dystrophy (LGMD) 2I and congenital muscular dystrophy 1C (MDC1C). Zebrafish are a useful animal model to reveal the mechanism of these diseases caused by mutations in FKRP gene. Downregulating FKRP expression in zebrafish by two different morpholinos resulted in embryos which had developmental defects similar to those observed in human muscular dystrophies associated with mutations in FKRP. The FKRP morphants showed phenotypes involving alterations in somitic structure and muscle fiber organization, ...
STUDY DESIGN: The progression of intervertebral disc degeneration (IDD) follows anterolateral "stab" of adult rabbit lumbar discs by 16-gauge hypodermic needle to a limited (5-mm) depth was studied for up to 24 weeks using magnetic resonance imaging (MRI).OBJECTIVES: To develop a slowly progressive, reproducible rabbit model of IDD suitable for studying pathogenesis and pathopysiology of intervertebral disc degeneration. Moreover, to improve the MRI methods and achieve improved MRIs a custom quad coil tuned and matched for 3.0T was developed. Higher SNR achievable with this coil allowed the acquisition of higher resolution images.METHODS: Part1 - The L2-L3, L3-L4, and L4-L5 lumbar intervertebral discs of 18 skeletally mature female New Zealand White (NZW) rabbits were stabbed by 16-gauge hypodermic needle to a depth of 5-mm in the left anterolateral annulus fibrosis (AF). Serial MRI scans of the stabbed discs and intact L1-2 and L5-6 control discs were performed at 3, 6, 12, and 24 weeks post ...

However, a major limitation to the study of diet-induced insulin resistance is the lack of relevant animal models. Numerous studies, mostly in rodents, have demonstrated that diets high in fructose induce insulin resistance; however, important metabolic differences exist between rodents and primates. Thus, the results of metabolic studies performed in primates are substantively more translatable to human physiology, underscoring the importance of establishing nonhuman primate models of common metabolic conditions. In this report, we demonstrate that a high-fructose diet in rhesus monkeys produces insulin resistance and many features of the
E7 proteins are major oncoproteins of high-risk human papillomaviruses (HPVs), which play a key role in cervical carcinogenesis. These proteins have been shown to immortalize primary human cells. Due to the absence of antibodies with suitable sensitivity and specificity, little is known about expression of the E7 oncoproteins in naturally infected tissues. Recently, high-level expression of the E7 protein of HPV-16, the most prevalent oncogenic HPV type, was demonstrated in cervical carcinomas by immunohistochemistry; however, approximately 15 additional high-risk HPV types are known to be associated with cervical carcinoma. It is unknown whether the E7 oncoproteins of HPV-18 and -45, the second and third most prevalent HPV types, are expressed in cervical cancers. Using antibodies against HPV-18 and -45 E7 proteins, it is shown here for the first time that the HPV-18 and -45 E7 proteins can be detected in cervical carcinoma biopsies. Together with anti-HPV-16 E7 antibodies, this could create the

One such example is metaplastic breast carcinoma, a rare but aggressive cancer with a heterogeneous histology, including squamous, chondroid, and spindle cells. Metaplastic carcinomas often contain an admixed conventional ductal invasive or in situ mammary carcinoma component, and are typically triple-negative for estrogen receptor, ...
Abstract:. Follicular dendritic cell sarcoma (FDCS) is an uncommon tumor arising from follicular dendritic cells that normally form meshwork in lymphoid follicles , this case of ours is a 65yr old female , presented with one month history of cervical swelling along with abdominal discomfort . She has a past history of radical mastectomy for carcinoma breast treated with chemoradiation 12 years back. Endoscopy revealed polypoid lesion in the body of stomach. Both the tumors - Lymph node & Gastric lesion, were excised and submitted to routine histopathological examination. Lymphnode swelling was confirmed as FDCS on immunohistochemistry. Polypoidal lesion of stomach was confirmed as Benign Gastro intestinal stromal tumor(GIST).Cases of FDCS associated with other malignancies have been described in literature. However to our knowledge, this is the first report in literature, were FDCS is seen simultaneously with GIST - stomach. However further studies are needed for possible relationship between ...
Background: Previous work has implicated accumulation of Zn2+ as a contributor to ischemic brain injury, however the sources of toxic Zn2+ accumulation are not fully understood. Previous data report that ICV delivery of a Zn2+ chelator limited mild focal stroke (Neuroscience 115 (2002) 871-8). We have recently demonstrated substantial synaptic Zn2+ release following spreading depolarization (JCBFM 31 (2011) 1073-84), and thus it is possible that repetitive spreading depolarization_ known to occur following stroke_ could be a major source of toxic Zn2+. In the present study, we examined whether synaptic Zn2+ release contributes to neuronal injury in a murine focal stroke model.. Methods The effect of synaptic Zn2+ was assessed using mice lacking synaptic Zn2+, due to genetic deletion of the synaptic vesicle transporter ZnT3 (ZnT3 KO). Wild type C57Bl/6 mice were age and sex matched as controls. Mice were anesthetized with isofluorane and the cortical branch of the right middle cerebral artery was ...
