Purpose: : Retinal prosthesis provides a promising approach to restore vision following the photoreceptor loss in people affected by retinitis pigmentosa or macula degeneration. The concept has already been validated in patients, but the physiological state of the surviving cells and network remains partially unknown. We therefore examined retinal ganglion cells (RGCs) at physiological and morphological levels in P23H rats at different ages during and after photoreceptor degeneration. Methods: : Whole cell patch clamp recordings were obtained by RGCs of P23H rats and compared to age-matched (10-18 weeks) control Sprague Dawley rats. Immunostaining techniques were used to test the presence of morphological alterations. Results: : Whole cell patch clamp recordings showed that in P23H rats, RGCs, although still able to fire action potentials, are significantly depolarized compared to control animals. Pain signaling involves transmission of nociceptive stimuli in the spinal cord where a critical balance between excitatory and inhibitory inputs determines the response to noxious stimuli. The neuropeptide, substance P (SP), mediates transmission of pain in part by binding to the tachykinin receptor (NK-1R) in the dorsal horn (DH) of the spinal cord. One of SPs downstream effects is to modulate N-type Ca2+ (N-) channels. While phospholipid breakdown is a part of the inflammatory process that accompanies tissue damage, the role of this metabolic pathway has not been completely described with respect to N-channel modulation during pain signaling. Despite the incomplete understanding of this modulation, pharmacological antagonists of both NK-1R and N-channels have been used to treat pain. As far as we know, our experiments are the first to combine single-molecule imaging and tracking with simultaneous whole-cell voltage-clamp recordings to measure receptor mobility and function concomitantly. We found that receptor lateral mobility doubled when the channels were opened and that this required calcium flux. The evidence to suggest this is threefold. First, the ATP-evoked increase in mobility was blocked by recording in calcium-free extracellular buffers. Second, S340Y mutants that are known to abrogate calcium flux also lacked ATP-evoked increases in P2X2 mobility (Migita et al., 2001; Egan and Khakh, 2004). Third, ATP-evoked mobility increases were blocked by voltage clamping the cells at positive voltages (+70 mV), when the inward driving force on calcium ions was substantially reduced. However, ATP-evoked P2X2 receptor mobility increases were not blocked by intracellular dialysis with BAPTA, implying that calcium entry via P2X2 receptors acts locally to increase mobility. ...

This study provides the first direct evidence that ATP can pass through Cx43 hemichannels. Three alternative approaches were used to evaluate the permeability of ATP through Cx43 hemichannels. First, we used inside-out patches with a pipette solution containing 130 mm Na2ATP, whereas the intracellular side of the patch was exposed to a pure sucrose solution. Under these conditions, frequent 26 pS channel openings were detected as outward currents in patches obtained from C6-Cx43+ cells (Fig. 6A). The outward current is generated by efflux of ATP anions, because passage of Na+ will result in an inward current. The VRev were −15 mV in these experiments, suggesting that the permeability of ATP2− is higher than Na+ (PNa/PATP = 1:2.5), because Na+ concentration was double of ATP2− concentration and sucrose has no charge. At first I could handle the induction, then it got so overwhelming my contractions never ceased. I had one continual one for over 2 hours. They werent even recording on the monitor. Then the mag started and I couldnt move or open my eyes. It was as if I was locked inside my body and I thought I was going to die. I could kind of hear what was happening around me, but it was all very surreal. Then I pushed very hard and he came really really quickly. I guess my pressures were 160s over 130s. I know women here have had higher than that, and I am SO scared about seizures. I have been convinced that I will die this time ...

The present study was designed to investigate the electrophysiological properties of strial pericytes and the effect of aspirin on pericyte K+ channels. Pericytes were identified by determining their morphological characteristics and using pericyte‑associated immunofluorescence techniques. The electrophysiological properties of strial pericytes were observed with a whole‑cell patch‑clamp technique. Alterations in the outward current of cochlear pericytes in the stria vascularis of guinea pigs were examined following the application of K+ channel retardants. The effects of aspirin on pericyte K+ channels were also evaluated with the whole‑cell patch‑clamp technique. Tetrodotoxin (TTX) is believed to be one of the most selective inhibitors of voltage-gated fast Na+ channels in excitable tissues. Recently, however, TTX has been shown to block L-type Ca2+ current (ICa) in canine cardiac cells. In the present study, the TTX-sensitivity of ICa was studied in isolated canine ventricular myocytes as a function of (1) channel phosphorylation, (2) extracellular pH and (3) the redox potential of the bathing medium using the whole cell voltage clamp technique. Fifty-five micromoles of TTX (IC50 value obtained under physiological conditions) caused 60% ± 2% inhibition of ICa in acidic (pH = 6.4), while only a 26% ± 2% block in alkaline (pH = 8.4) milieu.
