Family-wide molecular diagnostic assays are valuable tools for initial identification of viruses during outbreaks and to limit costs of surveillance studies. Recent discoveries of paramyxoviruses have called for such assay that is able to detect all known and unknown paramyxoviruses in one round of PCR amplification. We have developed a RT-PCR assay consisting of a single degenerate primer set, able to detect all members of the Paramyxoviridae family including all virus genera within the subfamilies Paramyxovirinae and Pneumovirinae. Conflicting reports exist regarding the requirement for virus replication in interferon (IFN) induction by paramyxoviruses. Our previous work has demonstrated that pathogen-associated molecular patterns capable of activating the IFN-induction cascade are not normally generated during virus replication, but are associated instead with the presence of defective interfering (DI) viruses. We demonstrate here that DIs of paramyxoviruses, including parainfluenza virus 5, mumps virus and Sendai virus, can activate the IFN-induction cascade and the IFN-β promoter in the absence of virus protein synthesis. As virus protein synthesis is an absolute requirement for paramyxovirus genome replication, our results indicate that these DI viruses do not require replication to activate the IFN-induction cascade.

A comprehensive study using virological and serological approaches was carried out to determine the occurrence of avian paramyxoviruses (APMVs) and avian influenza viruses (AIVs) in live healthy mallard ducks (Anas platyrhynchos) in addition to caged birds, wild birds (other than waterfowl), and poultry. Thirty-three viruses were isolated from 321 tracheal and cloacal swabs from mallard ducks and were characterised as: 6 AIV (two H5N2 and four H4N6), 10 APMV-1, and 17 APMV-4. Of 335 serum samples tested for AIV antibodies, 109 (32.5%) sera were positive by nucleoprotein-blocking ELISA (NP-B-ELISA). Pigeon paramyxovirus type 1 (PPMV-1) is an antigenic variant of avian paramyxovirus type 1, which mainly infects pigeons. Here, we characterized ten PPMV-1 viruses isolated from pigeons in China during 1996-2019. Phylogenetic analysis of available complete genomes, F and HN genes of PPMV-1 from China showed that multiple PPMV-1 genotypes (I, II, VI, and VII) exist in pigeons in China. Ten PPMV-1 viruses isolated in this study belonged to genotypes VI.1.2.2.2, VI.2.1.1.2.1, VI.2.1.1.2.2 and VII respectively. Genotype VI is predominant in pigeons. This project is concerned with how the recently discovered paramyxovirus, human metapneumovirus (HMPV), interacts with host cells to initiate entry. In this dissertation I explore whether the HMPV fusion protein interacts with cellular proteins during virus attachment and whether receptor binding is sufficient to initiate virus-mediated fusion at the cell surface. In this research I developed novel techniques to study HMPV fusion and entry. I learned that the HMPV fusion protein uses RGD-binding integrins to mediate attachment and virus entry, binding to integrins is not sufficient to trigger HMPV fusion, and HMPV fuses with intracellular membranes instead of at the cell surface. We examined the sites of phosphorylation and the role that phosphorylation plays in the function of the M2-1 protein in transcription and interaction with viral RNA. The M2-1 protein was found by proteolytic digestion and site-directed mutagenesis to be phosphorylated at serine 58 and serine 61. Serines 58 and 61 lie in the consensus sequence for phosphorylation by CKI and are conserved in human, bovine, and ovine RS virus and in turkey rhinotracheitis virus (1, 8, 26, 29, 35, 37). The conservation of these residues suggests a functional role for M2-1 phosphorylation. Phosphorylation of serine 58 creates a new CKI site at serine 61. Resumo: O metapneumovírus aviário (AMPV) é o agente primário da rinotraqueíte dos perus (TRT). O AMPV pertence à família Paramyxoviridae, subfamília Pneumovirinae, gênero Metapneumovirus. Também está associado à síndrome da cabeça inchada (SHS) em galinhas e é responsável por significativas perdas econômicas em sua produção. O presente estudo foi dividido em três partes. A primeira parte do trabalho consistiu em avaliar a beta-actina, gene utilizado como controle interno das técnicas moleculares de detecção viral, das células chicken embryo related (CER). Para isso, foi realizado o sequenciamento dos amplicons gerados pelo PCR do gene da beta-actina. A beta-actina das células BHK21 e CER foram detectadas utilizando oligonucleotídeos hamster-específicos. Além disso, pela análise filogenética as células CER e BHK21 apresentaram uma alta similaridade genética (p,0.996).
