Snake venom has been used for centuries as a traditional Chinese medicine. Hydrostatin-SN1 (H-SN1), a bioactive peptide extracted from the Hydrophis cyanocinctus venom gland T7 phage display library, was reported to have the ability to reduce inflammation in a dextran sulfate sodium-induced murine colitis model. In this study, we sought to investigate the inhibitory potential of H-SN1 on inflammation in a murine model of lipopolysaccharide (LPS)-induced acute lung injury (ALI), and elucidate the anti-inflammatory mechanism in LPS-stimulated RAW 264.7 cells. In vivo, C57BL/6 male mice were intratracheally instilled with LPS or physiological saline with concurrent intraperitoneal injection of H-SN1 or saline alone. Lung histopathologic changes, lung wet-to-dry weight ratio, and myeloperoxidase activity in lung tissues were assessed. Total cell number, the protein concentration, and cytokine levels were determined in the bronchial alveolar lavage (BAL) fluid. In vitro, RAW 264.7 cells were treated with

RATIONALE: Drugs used in chemotherapy use different ways to stop tumor cells from dividing so they stop growing or die. Giving chemotherapy drugs before surgery may shrink the tumor so that it can be removed during surgery. Radiation therapy uses high-energy x-rays to kill tumor cells. Combining radiation therapy with chemotherapy may kill more tumor cells. It is not yet known whether chemotherapy is more effective followed by surgery or combined with radiation therapy in treating cervical cancer.. PURPOSE: Randomized phase III trial to compare the effectiveness of chemotherapy followed by radical hysterectomy with that of chemotherapy plus radiation therapy in treating patients who have stage IB or stage II cervical cancer. ...

BACKGROUND AND RATIONALE:. Carcinoma Cervix is the commonest malignancy and a leading cause of cancer mortality seen in Indian women. At Tata Memorial Hospital, Carcinoma Cervix constitutes approximately 10% of all cancers (1). Nearly 85% of the patients present with advanced stages (FIGO Stage II/III). The main stay of treatment has traditionally been radical radiation therapy with 80-90% of patients requiring radiation in their lifetime and over decades the survival rates have achieved a plateau of 30 - 55% at 5 years. In patients with advanced stages (stages IIB to IVA), 15 - 38% have para-aortic lymph nodal metastases (2). Identification of para-aortic nodal status allows modification of radiation therapy fields to include this nodal disease, which, because of intestinal morbidity, is not routinely included in the treatment field by most of the Radiation Oncologists. Extended field radiation therapy that includes the para-aortic nodes is associated with a 31% to 50% 5 year-survival, ...
The yolk sac and the para-aortic splanchnopleura/aorta-genital ridges-mesonephros (P-Sp/AGM) region are the main sites of haematopoietic activity in the mouse embryo at the pre-liver stage of development. By day 11.5 of gestation, the AGM region is capable of autonomous initiation and expansion of definitive haematopoietic stem cells (HSCs). By day 12.5, HSC activity in the AGM region is reduced whilst a second wave of HSCs begins to emerge in the yolk sac. We show here that HSCs emerging in both locations are marked by co-expression of the endothelial-specific marker VE-cadherin and the pan-leukocyte antigen CD45. Phenotypic characterisation using CD31, TIE2, FLK1, Ac-LDL receptors, and CD34 markers demonstrated significant similarities between this VE-cadherin+CD45+ ;double-positive population and endothelial cells suggesting a common origin for these cells. The double-positive fraction also expressed the stem cell markers Kit, Sca1 and AA4.1. Long-term transplantation experiments ...

Comparative genomics has revealed extensive horizontal gene transfer among prokaryotes, a development that is often considered to undermine the tree of life concept. However, the possibility remains that a statistical central trend still exists in the phylogenetic forest of life. A comprehensive comparative analysis of a forest of 6,901 phylogenetic trees for prokaryotic genes revealed a consistent phylogenetic signal, particularly among 102 nearly universal trees, despite high levels of topological inconsistency, probably due to horizontal gene transfer. Horizontal transfers seemed to be distributed randomly and did not obscure the central trend. The nearly universal trees were topologically similar to numerous other trees. Thus, the nearly universal trees might reflect a significant central tendency, although they cannot represent the forest completely. However, topological consistency was seen mostly at shallow tree depths and abruptly dropped at the level of the radiation of archaeal and

What theory were you expecting early molecular evolutionists to use? They had to develop their own theory of evolution, because what little the MS had to offer them was wrong. Anfinsen, Dayhoff, Vogel, Zuckerkandl, and others compared molecules, and inferred differences that they interpreted as individual evolutionary changes. For them, an accepted mutation was the unit step in evolution. They referred to these discrete changes as mutations that were said to be (individually) accepted or rejected by selection (which was the editor, not the composer of the genetic message, in the famous words of King & Jukes). They reasoned that if part of a protein remains constant between two species, while another part changes, the latter part is less important for the function of the protein. Over time, this mode of reasoning became central to molecular studies, and today it is a cornerstone of genomics ...

Despite shortening vasopressor use in shock, hydrocortisone administration remains controversial, with potential harm to the immune system. Few studies have assessed the impact of hydrocortisone on the transcriptional response in shock, and we are lacking data on burn shock. Our objective was to assess the hydrocortisone-induced transcriptional modulation in severe burn shock, particularly modulation of the immune response. We collected whole blood samples during a randomized controlled trial assessing the efficacy of hydrocortisone administration in burn shock. Using whole genome microarrays, we first compared burn patients (n = 32) from the placebo group to healthy volunteers to describe the transcriptional modulation induced by burn shock over the first week. Then we compared burn patients randomized for either hydrocortisone administration or placebo, to assess hydrocortisone-induced modulation. Study groups were similar in terms of severity and major outcomes, but shock duration was significantly

An emerging class of natural surfactants, named alkylpolyglucosides, which can form both, the thermotropic and the lyotropic, liquid crystalline phases, were focused. The aim of the study was to integrate some physicochemical properties (characterised through the polarization and transmission electron microscopy, wide-angle X-ray diffraction, thermal analysis and rheology) of the three formulations based on cetearyl glucoside and cetearyl alcohol, with the in vitro (the artificial skin constructs) and in vivo bioavailability of hydrocortisone (HQ, in comparison with a standard pharmacopoeial vehicle. The parameters measured in vivo were erythema index (an instrumental human skin blanching assay), transepidermal water loss (TEWL) and stratum corneum hydration. A complex colloidal structure of lamellar liquid crystalline and lamellar gel crystalline type was deduced for sugar surfactant-based vehicles. In dependence on surfactant/water/oil ratio, several thermodinamically variable fractions of ...
The aorta-gonad-mesonephros (AGM) is a region of embryonic mesoderm that develops during embryonic development from the para-aortic splanchnopleura in chick, mouse and human embryos. It has been suggested that this area, in particular the ventral wall of the dorsal aorta, is one of the primary origins of the definitive haematopoietic stem cell. The aorta-gonad-mesonephros (AGM) region is an area derived from splanchnopleura mesoderm identified in embryonic humans, mice, and non-mammalian vertebrates such as birds and zebrafish. It contains the dorsal aorta, genital ridges and mesonephros and lies between the notochord and the somatic mesoderm, extending from the umbilicus to the anterior limb bud of the embryo. The AGM region plays an important role in embryonic development, being the first autonomous intra-embryonic site for definitive haematopoiesis. Definitive haematopoiesis produces hematopoietic stem cells that have the capacity to differentiate any blood cell lineage in the adult ...

Evolution is defined as adapted set of positively favorable changes across successive generations in heritable characteristics of different biological species [1]. It often imposes divergence in the organisms at every evolutionary level, be it genetic or epigenetic [2]. Canalization thus study the genotypic variants contributing to the similar phenotype [3]. Hence irrespective of genotypes, genetic alterations assisting the individuals phenotype solely guides the evolutionary adaptation. In this regard, the molecular clock hypothesis proposed by Zuckerkandl & Pauling [4] seems to be correct. Sarich & Wilson [5,6] also demonstrated the divergence of humans and apes approximately 5 million years ago through the same hypothesis. Evolutionary factors Evolution being highly dimensional phenomenon is almost impossible to explain simply. Even the mathematical analysis fails to correctly track the evolutionary rate in a population [7,8]. While diversions exist for every evolved factor among different ...
There are just two questions to be asked in evolution: how are things related, and what makes them differ? Lamarck was the first biologist-he invented the word-to address both. In his Philosophie Zoologique (1809) he suggested that the relationships among species are better described by branching trees than by a simple ladder, that new species arise gradually by descent with modification and that they adapt to changing environments through the inheritance of acquired characteristics. Much that Lamarck imagined has since been superseded. Following Wallace and Darwin, we now envision that species belong to a single highly branched tree and that natural selection is the mechanism of adaptation. Nonetheless, to Lamarck we owe the insight that pattern is produced by process and that both need mechanistic explanation.. Questions of pattern, process and mechanism pervade the modern discipline of molecular evolution. The field was established when Zuckerkandl & Pauling (1965) noted that haemoglobins ...
The general idea of a molecular clock was developed by Linus Pauling and Emile Zuckerkandl shortly after the prerequisite developments in protein chemistry during the late 1950s.3 At that point it was known that each protein is constructed from a sequence of amino acids that fold into a distinctive shape required by the proteins function. Each of the 20 possible amino acid molecules consists of a carboxyl group (-COOH) opposite an amino group (-NH2) at the other end of the molecule. In between these extremities is a so-called alpha carbon atom from which an additional side chain is attached that gives each amino acid its distinctive structure. These side chains vary considerably in size and complexity and thus can be expected to be a factor in explaining why only certain amino acids are found at crucial locations in the operative protein. When linked together in the polypeptide chain that constitutes a protein, the carboxyl group of one amino acid binds to the amino group of another with the ...

Molecular phylogenetics (/məˈlɛkjʊlər ˌfaɪloʊdʒəˈnɛtɪks, mɒ-, moʊ-/) is the branch of phylogeny that analyses hereditary molecular differences, mainly in DNA sequences, to gain information on an organisms evolutionary relationships. The result of a molecular phylogenetic analysis is expressed in a phylogenetic tree. Molecular phylogenetics is one aspect of molecular systematics, a broader term that also includes the use of molecular data in taxonomy and biogeography. The theoretical frameworks for molecular systematics were laid in the 1960s in the works of Emile Zuckerkandl, Emanuel Margoliash, Linus Pauling, and Walter M. Fitch. Applications of molecular systematics were pioneered by Charles G. Sibley (birds), Herbert C. Dessauer (herpetology), and Morris Goodman (primates), followed by Allan C. Wilson, Robert K. Selander, and John C. Avise (who studied various groups). Work with protein electrophoresis began around 1956. Although the results were not quantitative and did not ...

Of the 81 patients with imaging data, 19 (23%) had isolated adrenal involvement (iPAL), while 62 (77%) had additional extra-adrenal involvement (PAL+). Among patients who had both CT and PET scans, 18FDG-PET revealed extra-adrenal involvement not detected by CT scan in 9/18 cases (50%). The most common clinical manifestations were B symptoms (55%), fatigue (45%), and abdominal pain (35%). Endocrinological assessment was often inadequate. With a median follow-up of 41.6 months, 3-year progression-free (PFS) and overall (OS) survival rates in the entire cohort were 35.5% and 39.4%, respectively. The hazard ratios of iPAL for PFS and OS were 40.1 (95% CI: 2.63-613.7, P = 0.008) and 2.69 (95% CI: 0.61-11.89, P = 0.191), respectively. PFS was much shorter in iPAL vs PAL+ (median 4 months vs not reached, P = 0.006), and OS also appeared to be shorter (median 16 months vs not reached), but the difference did not reach statistical significance (P = 0.16). Isolated PAL was more frequent in females (OR = ...

The coexistence of hematological malignancy with endometrial cancer is a rare phenomenon. We report a case of coexistence of endometrial cancer with follicular lymphoma which we suspected preoperatively and diagnosed during surgery by a multidisciplinary intraoperative assessment. A 67-year-old woman was referred to our hospital due to a suspicion of an endometrial cancer. Endometrial biopsy revealed grade 1 endometrioid adenocarcinoma. MRI showed invasion of the tumor into the outer half of the myometrium, and abdominal CT showed para-aortic and atypical mesentery lymphadenopathy which was suspected to be metastasis of endometrial cancer or malignant lymphoma. Abdominal hysterectomy with bilateral salpingo-oophorectomy, pelvic and para-aortic lymphadenectomy, partial omentectomy, and mesentery lymph node biopsy for endometrial cancer were performed. The mesentery and para-aortic lymph nodes that were sent for frozen section analysis showed no metastasis of the endometrial cancer. We simultaneously
Hematopoietic stem cells (HSCs) are the cells that can give rise to all different lineages of differentiated/mature blood cells. HSCs are capable of self-renewal and maintaining the homeostasis of differentiated cells for specific functions. The adult type HSCs were first identified in the para-aortic splanchnopleura/Aorta-Gonad-Mesonephros (PSp/AGM) region, the vitelline and umbilical arteries, yolk sac, and placenta. The current hypothesis is that HSCs differentiate from a hemogenic endothelium. Core binding factors (CBFs), Runx1 and CBFβ, are essential for the generation of definitive hematopoiesis. We hypothesized that Runx1-CBFβ function is specifically required in the hemogenic endothelium per se for HSC emergence. To test this hypothesis, we selectively disrupted or supplied Runx1-CBFβ function in endothelial cells. Specifically, we conditionally deleted Runx1 in endothelial cells with Cre recombinases expressed in the endothelium (Tek-Cre and VE-cadherin-Cre) combined with a floxed ...
If you or a loved one have been diagnosed with endometrial cancer, talk to your doctor about your options for treatment. Since most uterine cancers are diagnosed in their early stages most doctors recommend surgical treatment as it provides an excellent chance for cure. Surgically removing the uterus - also known as a hysterectomy is the second most common surgical procedure amongst women in the United States. With endometrial cancer, comprehensive surgical staging that includes a hysterectomy with pelvic and para-aortic lymphadenectomy is recommended. Here at the CARES Center our gynecologic oncology surgeons are leading the southeast in a minimally invasive procedure known as the da Vinci® Hysterectomy for Endometrial Cancer. By using the da Vinci® Surgical System surgeons are able to perform the procedure through smaller incisions and with increased visualization of the surgical field. This minimally invasive approach has shown numerous benefits when compared to a traditional hysterectomy. ...
Ruptured pseudoaneurysm following pelvic surgery is a rare and fatal complication. Because of its rarity, existing evidence is limited to a small case series. The role of nongenomic action of estrogens on elicited catecholamine secretion and exocytosis kinetics was studied in perfused rat adrenals and in cultured bovine chromaffin cells. 17β-Estradiol as well as the estrogen receptor modulators raloxifene and LY117018, but not 17α-estradiol, inhibited at the micromolar range the catecholamine output elicited by acetylcholine or high potassium. However, these agents failed to modify the secretion elicited by high Ca2+ in glands treated with the ionophore A-23187 (calcimycin), suggesting that estrogens did not directly act on the secretory machinery. At the single cell level, estrogens modified the kinetics of exocytosis at nanomolar range. All of the drugs tested except 17α-estradiol produced a profound slowing down of the exocytosis as measured by amperometry. LY117018 also reduced the granule content of catecholamines. 17β-Estradiol reduced the intracellular free Ca2+ but only at micromolar concentrations, whereas nanomolar concentrations ...
Abstract: Small cell lung cancer (SCLC) is one of the most lethal human malignancies, due to its high metastatic potential and chemo-resistance upon relapse. Using the Rbf/f;p53f/f mouse model for SCLC, we found that the tumors are often composed of phenotypically different cells, characterized by mesenchymal and neuroendocrine markers. These cells often share a common origin. Crosstalk between these cells can endow the neuroendocrine component with metastatic capacity, illustrating the potential relevance of tumor cell heterogeneity in dictating functional tumor properties. Also specific genetic lesions appear to be associated with metastatic potential. Interestingly, the two cell types can also interconvert raising the question of their cell-of-origin. To investigate this in more detail, we inactivated Trp53 and Rb1 in distinct cell types in the adult lung by targeting Cre-recombinase expression to Clara, neuroendocrine, and alveolar type II cells using adenoviral vectors. We could show that ...
Despite the decades long demonstration of lengthy CNS axon regeneration, our knowledge of the glial factors that support the migration of these axons after injury is rather limited. We do know that, depending on the types of regenerative stimuli, some axons appear to be able to migrate successfully toward their targets. In many cases, however, axons take circuitous routes back toward the cell body (Luo et al., 2013; Pernet and Schwab, 2014; Yang et al., 2020). As functional regeneration requires innervation of axons to their correct targets, we are becoming increasingly aware of the importance of defining the cellular and molecular factors that regulate this process in adulthood.. Axons cannot simply elongate in space. Rather, they normally bind to substrates. Several studies have demonstrated that regenerating axons follow astrocytes. In the mature CNS, there are two main types of astrocytes, the protoplasmic and the fibrous. The former resides mainly in the gray matter, whereas the latter ...
