Papain is also known as papaya proteinase-I which is an enzyme present in papaya. Papain is an important digestive enzyme which breakdown tough protein fibers. Papain is very stable also at very elevated temperatures. Papain latex is a rich source of cysteine endopeptidases including chymopapain, caricain and glycyl endopeptidase. Papain is traditionally used as a meat tenderizer and also used in skin burns, skin ulcers, wounds etc. Some of the common uses of papain are it boosts digestion, treats skin and wound, resistant to fungus, supports immune system and source of antioxidant etc. As papain is source of antioxidant, it is commonly used in food preservation to reduce bacterial infestations and spoilage of food due to oxidation.. Market Segmentation: Papain. The Papain market is segmented on the basis of its applications in different industries like food industry, cosmetic industry, pharmaceutical industry and chemical industry etc. Papain is used for food industry for food processing, as a ...
Limited papain hydrolysis of immunoglobulin M (IgM) produces a subunit-like proteolytic fragment designated IgMp (Inman & Hazen, 1968). In the presence of mercaptans, IgMp partially dissociated into Fcμ-like and Fabμ fragments. Treatment of residual IgM (that remaining after a papain digestion) with 2mm-mercaptoethylamine resulted in fragmentation of the same type that occurs in a routine limited digestion of IgM with papain, although exogenous enzyme was not added to the mixture. When IgM was hydrolysed with 14C-labelled papain, a small quantity of the enzyme was found to be associated with the residual IgM and IgMp fractions. IgM and IgM 7S subunit (IgMs) that had been exposed to papain in the absence of activating mercaptan and separated from the enzyme by gel filtration also fragmented when subsequently treated with 2mm-mercaptoethylamine. The fragments resembled those produced during a typical limited papain digestion of IgM. It was concluded that mercaptoethylamine induced fragmentation ...
Allergic diseases represent a significant burden in industrialized countries, but why and how the immune system responds to allergens remain largely unknown. Because many clinically significant allergens have proteolytic activity, and many helminths express proteases that are necessary for their life cycles, host mechanisms likely have evolved to detect the proteolytic activity of helminth proteases, which may be incidentally activated by protease allergens. A cysteine protease, papain, is a prototypic protease allergen that can directly activate basophils and mast cells, leading to the production of cytokines, including IL-4, characteristic of the type 2 immune response. The mechanism of papains immunogenic activity remains unknown. Here we have characterized the cellular response activated by papain in basophils. We find that papain-induced IL-4 production requires calcium flux and activation of PI3K and nuclear factor of activated T cells. Interestingly, papain-induced IL-4 production was dependent
Papain, also known as papaya proteinase I, is a cysteine protease (EC 3.4.22.2) enzyme present in papaya (Carica papaya) and mountain papaya (Vasconcellea cundinamarcensis). Papain belongs to a family of related proteins with a wide variety of activities, including endopeptidases, aminopeptidases, dipeptidyl peptidases and enzymes with both exo- and endo-peptidase activity. Members of the papain family are widespread, found in baculoviruses, eubacteria, yeast, and practically all protozoa, plants and mammals. The proteins are typically lysosomal or secreted, and proteolytic cleavage of the propeptide is required for enzyme activation, although bleomycin hydrolase is cytosolic in fungi and mammals. Papain-like cysteine proteinases are essentially synthesised as inactive proenzymes (zymogens) with N-terminal propeptide regions. The activation process of these enzymes includes the removal of propeptide regions, which serve a variety of functions in vivo and in vitro. The pro-region is required for ...

Our collective results have shown that LEKTI1-12 and LEKTI multidomains had a strong inhibitory effect on trypsin, plasmin, KLK5, KLK6, KLK13, KLK14, cathepsin G, HNE, and subtilisin A. They had no inhibitory effect on KLK1, chymase, chymotrypsin and cysteine proteinases papain or cathepsins K, L, or S (Table 1). To understand whether LEKTI behaves as a slow - or fast-binding inhibitor, we measured the time course of various proteinase activities in the presence of different concentrations of rLEKTI. We have observed that the product formation over the 60 min assay period in the absence and presence of inhibitor was linear with respect to time [19,31,49,54]. The linear shapes of these inhibition curves indicate that rLEKTI is not a time-dependent inhibitor, suggesting that LEKTI binds rapidly to these proteinases and inactivates them. To classify the type of inhibition, the kinetic constants (Km and Vmax) of plasmin, trypsin, subtilisin A, cathepsin G, HNE, KLK5, KLK6, KLK13 and KLK14 were ...
Four hydrophobic amino acids (Leu, Tyr, Phe, Trp) were oligomerized by the protease papain in homo-oligomerization, binary co-oligomerization and ternary co-oligomerization. After 24 h, solid polydisperse reaction products of the homo-oligomerization were obtained in yields ranging from 30-80% by weight. A DPavg was calculated based on MALDI-ToF MS results using the ion counts for the chains in the product. Based on the DPavg and the yield of the homo-oligomerization it was determined that the amino acids can be ranked according to reactivity in the order: Tyr | Leu | Phe | Trp. Thermal degradation of the homo-oligomers shows two degradation steps: at 178-239 °C and at 300-330 °C. All the products left a significant amount of char ranging from 18-57% by weight at 800 °C. Binary co-oligomers were obtained as a polydisperse precipitate with a compositional distribution of the chains. Both the compositional and chain length distribution are calculated from MALDI-ToF mass spectra. By comparing the amount

High enzyme foods are one of the four main foundations of the E-Factor Diet plan. Enzymes are important because they help to digest the food that enters your body. As John Rowley stated in his book, its not only about what you eat that matters, but also what your body can absorb.. When you eat nutrient dense foods, you need enzymes to digest and absorb those nutrients. Without enzymes, most of the nutrients wont be absorbed by your body. You may even eat a lot but still feel hungry because your body has not received the nutrients it needs.. Enzymes come from enzymatic foods, such as plants and animals. Most foods with good enzyme are fruits and vegetables. The picture above gives a short list of various foods that are high in enzymes.. Here are a few details about enzymatic foods you should be eating:. Papaya contains the enzyme papain that digests protein. When you are trying to lose weight, eating enough protein is important to maintain muscle mass. Eating papaya supplies you with the ...

Two antigenic types of naturally occurring proteins related to the papain produced Fc fragment of γG-globulin have been studied. Most normal and myeloma γG-globulins are related to one or the other of these proteins, based on antigenic and structural characteristics of their heavy chain. Molecules bearing the determinant "Cr" are approximately 10 times as abundant as those bearing the determinant termed "Zu" in pools of normal γG-globulin and among a large group of γG-myeloma globulins. Both populations have properties of typical γG-globulin antibodies. Splenic cells, stained for these two populations with specific fluorescent antibody preparations, are found to contain one or the other of the two populations of γG-globulin, but not both. Approximately 10 times as many cells contain the Cr determinant as contain the Zu determinant.. ...

The chemical composition and evaluation of Indian squid (Loligo duvauceli) mantle, epidermal connective tissue and tentacle is investigated in this current study. It is observed that squid mantle contains 22.2% total protein; 63.5% of the total protein is myofibrillar protein. The unique property of squid myofibrillar protein is its water solubility. Squid mantle contains 12.0% total collagen. Epidermal connective tissue has highest amounts of total collagen (17.8%). SDS-PAGE of total collagen identified high molecular weight α-, β- and γ- sub-chains. Amino acid profile analysis indicates that mantle and tentacle contain essential amino acids. Arginine forms a major portion of mantle collagen (272.5 g/100 g N). Isoleucine, glutamic acid and lysine are other amino acids that are found in significantly high amounts in the mantle. Sulphur containing cystine is deficit in mantle collagen. Papain digest of mantle and epidermal connective tissue is rich in uronic acid, while papain digest, ...
Glycyl endopeptidase is a cysteine endopeptidase of the papain family, characterized by specificity for cleavage C-terminal to glycyl residues only and by resistance to inhibition by members of the cystatin family of cysteine proteinase inhibitors. Glycyl endopeptidase has been crystallized from high salt with a substrate-like inhibitor covalently bound to the catalytic Cys 25. The structure has been solved by molecular replacement with the structure of papain and refined at 2.1 A to an R factor of 0.196 (Rfree = 0.258) with good geometry. The structure of the S1 substrate binding site of glycyl endopeptidase differs from that of papain by the substitution of glycines at residues 23 and 65 in papain, with glutamic acid and arginine, respectively, in glycyl endopeptidase. The side chains of these residues form a barrier across the binding pocket, effectively excluding substrate residues with large side chains from the S1 subsite. The constriction of this subsite in glycyl endopeptidase explains ...

Papain is a powerful enzyme from the papaya fruit. This enzyme plays a crucial role in digestive processes involving breaking down tough meat protein fibres.
As highlighted by the similarity of their fold and the superimposition of their active site, we have shown that NsPCS belongs to the papain superfamily of cysteine proteases. It is therefore expected that the peptide hydrolase activity of NsPCS, i.e., the deglycination of GSH, resembles the general and well known mechanism of papain-like cysteine proteases. By analogy with such a mechanism, Cys-70, His-183, and Asp-201 (equivalent to Cys-56, His-162, and Asp-180 in AtPCS1) correspond to the catalytic triad in NsPCS. Moreover, it is also highly probable that Cys-70 and His-183 form a thiolate-imidazolium ion pair, stabilized by Asp-201, and that the nucleophilic attack of Cys-70 on the Cys-Gly peptide bond is favored by the oxyanion hole, made up of Cys-70 and Gln-64. In a second step, the water molecule W153, whose nucleophilicity is enhanced by the proximity of His-183 and Asp-201, is then ideally placed to attack the thioester bond and liberate the γ-glutamylcysteine. We could trap the ...

Basophils have gained renewed interest as inducers and amplifiers of Th2 immunity through their potential to migrate to LNs, provide an early source of IL-4, and at the same time act as APCs in models of helminth infection and injection of the model protease papain, but have been largely ignored in the field of pulmonary mucosal immunity and development of allergic asthma to inhaled allergens (Sokol et al., 2008, 2009). Here, we found that basophils are recruited to the MLNs draining the site of i.n. administration of HDM allergen within 3 d of HDM injection. Several mechanisms could explain this increase. Like basophil expansion and recruitment to sites of helminth infection (Voehringer et al., 2006), we found that this process required adaptive immunity and CD4 T cells, as MHCII-deficient mice had severely reduced recruitment of basophils. The most likely explanation is T cell-derived production of IL-3 (Shen et al., 2008; Kim et al., 2010), which we found to be induced in CD4 T cells upon ...
