Classical research has suggested that early palate formation develops via epithelial-mesenchymal interactions, and in this study we reveal which signals control this process. Using Fgf10-/-, FGF receptor 2b-/- (Fgfr2b-/-), and Sonic hedgehog (Shh) mutant mice, which all exhibit cleft palate, we show that Shh is a downstream target of Fgf10/Fgfr2b signaling. Our results demonstrate that mesenchymal Fgf10 regulates the epithelial expression of Shh, which in turn signals back to the mesenchyme. This was confirmed by demonstrating that cell proliferation is decreased not only in the palatal epithelium but also in the mesenchyme of Fgfr2b-/- mice. These results reveal a new role for Fgf signaling in mammalian palate development. Cleft palate is a common congenital abnormality that results from defective secondary palate (SP) formation. The Sine oculis-related homeobox 2 (Six2) gene has been linked to abnormalities of craniofacial and kidney development. Our current study examined, for the first time, the specific role of Six2 in embryonic mouse SP development. Six2 mRNA and protein expression were identified in the palatal shelves from embryonic days (E)12.5 to E15.5, with peak levels during early stages of palatal shelf outgrowth. Immunohistochemical staining (IHC) showed that Six2 protein is abundant throughout the mesenchyme in the oral half of each palatal shelf, whereas there is a pronounced decline in Six2 expression by mesenchyme cells in the nasal half of the palatal shelf by stages E14.5-15.5. Palatal primordia of day-12.5 mouse fetuses were cultivated in a chemically-defined medium by a suspension culture technique, and their growth and differentiation in vitro were compared with those of the fetal palates in vivo at the corresponding gestational stages. The maxillary explants cultivated in vitro for up to 72 hours did not grow in size and remained almost in the same size as those in day-12.5 fetuses. However, the palatal shelves elevated and grew faster towards the midline until 24 hours in culture as compared with in vivo development. Disruption of signaling pathways such as those mediated by sonic hedgehog (Shh) or platelet-derived growth factor (Pdgf) causes craniofacial abnormalities, including cleft palate. The role that microRNAs play in modulating palatogenesis, however, is completely unknown. We show that, in zebrafish, the microRNA Mirn140 negatively regulates Pdgf signaling during palatal development, and we provide a mechanism for how disruption of Pdgf signaling causes palatal clefting. The pdgf receptor alpha (pdgfra) 3 UTR contained a Mirn140 binding site functioning in the negative regulation of Pdgfra protein levels in vivo. pdgfra mutants and Mirn140-injected embryos shared a range of facial defects, including clefting of the crest-derived cartilages that develop in the roof of the larval mouth. Scar formation after repair of the cleft palate leads to growth impairment of the upper jaw and midface. The implantation of a suitable scaffold during surgery may reduce this adverse effect. However, little is known about tissue reactions to scaffolds implanted in the oral cavity. Our goal was to analyze the tissue reactions to cross-linked type I collagen scaffolds after submucoperiosteal implantation in the palate of rats. Collagen type I scaffolds were implanted in the palate of 25 male Wistar rats. Groups of 5 rats were killed consecutively after 1, 2, 4, 8, and 16 weeks and were processed for histologic and immunohistochemical analyses. Women who have epilepsy and who take the anticonvulsant diphenylhydantoin (DPH) during pregnancy produce infants that have an abnor-high incidence of cleft palate and other anomalies. DPH will induce cleft palates in laboratory mice exposed to the drug while in utero. It has been suggested that DPH may exert its teratogenic effect by interfering with Vitamin D metabolism. We tested the hypothesis that decreased concentrations of vitamin D could be secondary to treatment with DPH and could be related to the induction of cleft palates in mice. To do this we treated groups of pregnant mice with DPH or the combination of DPH and 25 hydroxycholecalciferol (25 hydroxy Vitamin D3) during the critical period of palatogenesis. Treatment with 25 hydroxycholecalciferol did not decrease the incidence of DPH-induced cleft palate. MATERIALS AND METHODS Pre-treatment records of 1007 patients were reviewed by a single examiner. A total of 125 records fulfilled the inclusion criteria and were further investigated. Bone measurements were performed on cone-beam computed tomography scans, at a 90° angle to the bone surface, on 28 predetermined and standardized points on the hard palate. Bone thickness at various areas was associated to clinically identifiable areas on the hard palate by means of pre-treatment plaster models. RESULTS Bone thickness ranged between 1.51 and 13.86mm (total thickness) and 0.33 and 1.65mm (cortical bone thickness), respectively.
