In the present study, we demonstrate that the LGG-derived soluble protein p40 activates EGFR in vivo and in vitro in colon epithelial cells. Activation of EGFR by p40 is required for inhibition of cytokine-induced apoptosis and disruption of barrier integrity in intestinal epithelial cells in vitro and ex vivo. Importantly, p40 prevents and treats DSS-induced colon epithelial injury and inflammation and ameliorates oxazolone-induced colitis, and it mediates reductions of intestinal epithelial apoptosis and disruption of barrier function in these two mouse models, in an EGFR activation-dependent manner. Therefore, our findings provide a rationale for conducting new hypothesis-driven studies to define the clinical efficacy of probiotic-derived proteins in preventive, adjunctive, or alternative treatments for intestinal inflammatory disorders.. Understanding p40s structure-function relationship is critical to elucidate the biological effects of this protein. Sequence analysis of p40 predicts that ...
Although previous studies have suggested that appendix seems to be involved in the colitis, the role of this in the pathogenesis remains unclear. In this study, we assessed the importance of appendiceal lymphoid follicles, specifically the cecal patches (CP) in mice, using an experimental colitis model. Treatment with oxazolone resulted in ulcerations particularly at CP with follicular expansion as well as colitis. The colitis was attenuated by either appendectomy or the absence of mature B cells. We therefore established an intravital imaging system accompanied by the fluorescence resonance energy transfer technology to analyze the dynamic immune response of CP B cells ...

Animal models mimicking human diseases have been used extensively to study the pathogenesis of autoimmune diseases and the efficacy of potential therapeutics. They are, however, limited with regard to their similarity to the human disease and cannot be used if the antagonist and its cognate receptor require high similarity in structure or binding. Here, we examine the induction of oxazolone-mediated features of atopic dermatitis (AD) in NOD-scid IL2R \(γ^{null}\) mice engrafted with human peripheral blood mononuclear cells (PBMC). The mice developed the same symptoms as immunocompetent BALB/c mice. Histological alterations induced by oxazolone were characterized by keratosis, epithelial hyperplasia and influx of inflammatory cells into the dermis and epidermis. The cellular infiltrate was identified as human leukocytes, with T cells being the major constituent. In addition, oxazolone increased human serum IgE levels. The response, however, required the engraftment of PBMC derived from patients ...

The presented examples demonstrate that 1,3-oxazol-5(4H)-ones are attractive intermediates not only for intermolecular but also for intramolecular reactions with nucleophiles. With a series of examples, the potential of these reactions was exemplified. The reactions of 1,3-oxazol-5(4H)-ones, as starting materials or as intermediates, bearing the nucleophilic group in ω-postion of the substituent at C(2), belong to real ring enlargement reactions as all atoms of the oxazolone ring are included in the new ring. They proceed under acid catalysis, e.g. under the conditions of the direct amide cyclization. Together with the azirine/oxazolone method for the introduction of α,α-disubstituted α-amino acids into peptides, these reactions are ideally suited for the synthesis of cyclodepsipeptides and cyclopeptides, especially of sterically congested ones, which contain α-aminoisobutyric acid (Aib) or other α,α-disubstituted glycines. On the other hand, 4,4-disubstituted 1,3-oxazol-5(4H)-ones ...
Performing PCR direct from crude samples can be highly beneficial due to greatly reduced experimental time, cost and sample loss, particularly for high volume genotyping, transgene detection, knockout analysis or PCR-based clinical and forensic tests. A major problem however can be the presence of inhibitors to PCR which may lead to low sensitivity reactions, low yields or false positives. PCRBIO HS Taq DNA Polymerase and PCRBIO Ultra Polymerase are designed to perform in demanding applications with high efficiency and improved tolerance to common inhibitors. Our advanced buffer and antibody-mediated hot start technology allows robust performance and high sensitivity across a wide range templates, including GC and AT-rich, together with easy reaction setup at room temperature. Furthermore, the convenient ready mix format, including the option of a red dye for direct gel loading, increases the speed and ease-of-use for successful PCR from crude samples.. For rapid DNA extraction combined with ...
Ear thickness measurements: The appearance of the ears, which were the application sites, appeared hard and reduced in size on Day 6 in the group treated with 25%. The ears in the group treated with 25% could not be measured on Day 6. Therefore, a second irritation screen was run with the substance at concentrations of 2.5, 5 and 10% (v/v). In the second irritation screen, the ears in the group treated with the test article at 10% had a hard feel to them on Day 6. The ear measurements indicated little or no edema in the ears of the mice treated with the test article at 2.5 and 5%, but treatment with 10% caused a 22.7% increase in ear thickness on day 3 and a 67.5% increase on Day 6, indicating the presence of edema ...

Guanine is the most readily oxidized of the four DNA bases, and guanine oxidation products cause G:C-T:A and G:C-C:G transversions through DNA replication. 8-Oxo-7,8-dihydroguanine (8-oxoG) causes G:C-T:A transversions but not G:C-C:G transversions, and is more readily oxidized than guanine. This review covers four major findings. (i) 2,2,4-Triamino-5(2H)-oxazolone (Oz) is produced from guanine and 8-oxoG under various oxidative conditions. Guanine is incorporated opposite Oz by DNA polymerases, except REV1. (ii) Several enzymes exhibit incision activity towards Oz. (iii) Since the redox potential of GG is lower than that of G, contiguous GG sequences are more readily oxidized by a one-electron oxidant than a single guanine, and OzOz is produced from GG in double-stranded DNA. Unlike most DNA polymerases, DNA polymerase ζ efficiently extends the primer up to full-length across OzOz. (iv) In quadruplex DNA, 3′-guanine is mainly damaged by one-electron oxidation in quadruplex DNA, and this damage
The use of amino thio S-acids in the azirine/oxazolone method and a novel isomerization led to Aib-containing endothiopeptides. With the aim of generalizing this method, a variety of Aib-containing dipeptide thioanilides have been prepared. By their treatment with ZnCl2 in AcOH, followed by HCl-saturated AcOH, the C=S group was shifted from the last to the penultimate amino acid in high yield and without epimerization. As this methodology is very useful for the specific introduction of a thioamide group, it was extended to Aib-containing tripeptides. In addition, it could be shown that a mechanism via spirocyclic intermediates (cf. Scheme 4) is most likely for this isomerization. To establish the proposed neighboring-group participation of the N-acyl group, model dipeptide thioanilides containing no N-terminal C=O group were synthesized. These derivatives did not undergo rearrangement. ...

AU - Teeri, Tuula. N1 - Project code: BEL2026. PY - 1995. Y1 - 1995. N2 - Single-chain antibodies were constructed using six different linker peptides to join the VH and VL domains of an anti-2-phenyloxazolone (Ox) antibody. Four of the linker peptides originated from the interdomain linker region of the fungal cellulase CBHI and consisted of 28, 11, six and two amino acid residues. The two other linker peptides used were the (GGGGS)3 linker with 15 amino acid residues and a modified IgG2b hinge peptide with 22 residues. Proteolytic stability and Ox binding properties of the six different scFv derivatives produced in Escherichia coli were investigated and compared with those of the corresponding Fv fragment containing no joining peptide between the V domains. The hapten binding properties of different antibody ...
It has been suggested that reserpine blocks expression of delayed hypersensitivity (DH) by depleting tissue mast cells of serotonin (5-HT), thereby preventing a T cell-dependent release of mast cell 5-HT necessary to localize and to amplify the DH response. However, reserpine blocks expression of DH in mast cell-deficient mice. We therefore decided to reevaluate the mechanism by which reserpine abrogates expression of cellular immunity, and investigated whether the drug might interfere with T cell activity in vitro or in vivo. At concentrations as low as 4 microM, reserpine profoundly suppressed baseline or antigen-augmented levels of [3H]thymidine incorporation by immune lymph node cells obtained from mice sensitized to the contactant oxazolone [I-LNC(Ox)]. This effect was observed both with I-LNC derived from normal mice and with I-LNC derived from congenitally mast cell-deficient W/Wv mice, cell preparations that lacked detectable mast cells, histamine, and 5-HT. Furthermore, treatment of ...

How Do You Determine Your CORE food allergens?. It is a simple process that you can do at home no charge. The majority of food allergies/intolerances are delayed sensitivity reactions. Elimination is the most accurate way for you to determine the delayed sensitivity reactions. To do this, you must, eliminate the suspected food intolerance completely from the diet for 3 weeks then reintroduce and watch for reactions for 4 days (see below). Your core food allergen/intolerance complicates the healing process by increasing the overall body inflammation. Once you remove the core food allergen your bodys inflammation will reduce, and sensitivities to other foods will decrease as well.. The main 4 food allergens are: wheat (gluten), corn (zein), dairy (casein), and soy Next common after that are chocolate, peanuts, eggs, tomatoes and beef. THE TEST:. Eliminate the suspected food intolerance completely from the diet for 3 weeks then reintroduce and watch for reactions for 4 days (see below). Immediate ...
The effect of a single time exposure of SLS to the buccal mucosa of mice was compared to one application of the hapten OXA (oxazolone), evaluated by routine histology, immunohistochemistry and ELISA quantifications of cytokines. The SLS concentrations (2%, 4% and 8%) resulted in epithelial surface necrosis at 1-6 h, after 2-6 h accumulation of intra-epithelial neutrophils and at 24 h the main inflammatory cells were mononuclear. Increased concentrations of SLS gave more severe damage. CD4(+) T cells were found at 6 h and increased slightly up to 24 h and were most frequently seen at the lowest SLS dose. The CD8(+) T cells were kept at a low number during the whole 24 h observation period, but increased proportionally to the CD4(+) T cells. One application of 1% OXA did not raise the number of cells of either phenotype (2-24 h). Neither IL-2 nor IFN-γ demonstrated increased levels during the week of observation at any concentration of SLS, contrary to one application of OXA which caused ...

The Trouble with Wheat Part III, or How You Got Fat Silently. Reference: Your Hidden Food Allergies are Making You Fat, Rudy Rivera 2002. Date: Oct 5th, 2010. Our last letter was about how wheat has a protein called gliadin in it that is hard to digest (11 steps) and can sneak into your gut. How exactly it gets through your intestinal wall is a bit of a mystery, but get through it does, in part because you have sampling cells that are constantly nibbling at whats out in the gut to see if its friend or foe. But the gliadin sets up a brisk response, and loosens the tight junctions. Now, you have leaky gut syndrome. More gliadin floods in and your body starts to make antibodies against any more gluten/gliadin that might show up.. Remember, you have 70% of your immune system around your intestines. Think about having a sensitivity reaction to a very common food, like wheat. What percentage of your immune output is going to be directed to battling wheat? Its meant to be good for you, and ...
TY - JOUR. T1 - Analysis of dose response of trinitrochlorobenzene contact hypersensitivity induction in mice. T2 - Pretreatment with cyclophosphamide reveals an optimal sensitizing dose. AU - Sullivan, Sabra. AU - Bergstresser, Paul R.. AU - Streilein, J. Wayne. PY - 1990/5. Y1 - 1990/5. N2 - A detailed dose-response curve has been established for induction of contact hypersensitivity (CH) in mice with trinitrochlorobenzene (TNCB). It was determined that in BALB/c, CBA/J, and C57BL/6 mice, the dose required to sensitize via epicutaneous application was between 1 and 10,μg TNCB. When doses of hapten of 200,μg or greater were painted on abdominal skin, CH responses were induced which were only marginally greater than responses induced by sensitizing doses of hapten in the 10-50-μg range, implying that no further dose-response relationship exists beyond 50 μg of hapten. However, in companion experiments, in which panels of mice were pretreated with cyclophosphamide, it was determined that ...

This finger prick blood test is a convenient way to assess for 190 different potential adverse food reactions. IgG is the major antibody found in serum. IgGs are composed of two fragment antigen binding (Fab) regions that contain the antigen binding sites and the Fc region, which is responsible for most of the biologic activity of the antibodies (Figure 1). An antigen is a substance that causes the immune system to produce an antibody that specifically reacts with it. IgG-mediated reactions to food antigens may be delayed by several hours or days, whereas IgE food antibody reactions are quite immediate. Also, headaches, skin problems, joint discomfort, and body pain can be associated with elevated IgG reactions to food. Doctors often use this test to evaluate for food sensitivity reactions such as gluten sensitivity.. THE BENEFITS OF TESTING ...
