Human leukocytes in suspension or in monolayer cultures have been processed for electron microscopy by fixation in a freshly made cold mixture of glutaraldehyde and osmium tetroxide and by postfixation in uranyl acetate. Simultaneous exposure to glutaraldehyde and osmium tetroxide eliminates many of the shortcomings seen when either of these agents is used alone as the initial fixative. Specimens are processed to the stage of dehydration as single cell suspensions or as very small clumps to assure rapid penetration of fixatives and efficient washing. The technique is rapid and reproducible. Electron micrographs presented in this report illustrate the ultrastructural features of human white cells prepared by this method.. ...

This was a controversy in the early days of electron microscopy, and is discussed in textbooks (Hayat, Glauert etc). Its some time since I read about this stuff, but OsO4 is certainly reduced to OsO2 by alcohol groups and also by some amino acid side-chains. However, its much more soluble in hydrophobic domains (middle of membrane) than in hydrophilic ones (inside or outside surface). The unit membrane (Robertson) appearance was explained at least partly by migration of the insoluble OsO2 molecules (or of an osmate indermediate) from the ...

This morning brings the news, via ABC, that the recently discovered bomb plot in London involved a quantity of osmium tetroxide. Thats a surprise. I know the reagent well, but its not what anyone would call a common chemical, despite the news story above that calls it easily obtained. Its quite odd that someone

A novel molecular scale device is described which is bactericidal, fungicidal and algicidal. The antipathogenic properties of the device are attributed to electron activity indigenous to diamagnetic semiconducting crystals of tetrasilver tetroxide (Ag4 O4) which contains two monovalent and two trivalent silver ions in each molecular crystal. When the crystals are activated with an oxidizing agent, they release electrons equivalent to 6.4 10-19 watts per molecule which in effect electrocute pathogens. A multitude of these devices are effective at such low concentrations as 0.3 PPM where they can kill 100% of 100 K/cc Streptococcus faecalis, and E. coli colonies in three minutes meeting the ten-minute EPA criteria of 100% kills within ten minutes for swimming pool and hot-tub applications. The devices can be used in utilitarian bodies of water, such as municipal and industrial water reservoirs.
Professor Miquel Colls team from the Spanish Research Council in Barcelona was able to obtain the molecular level picture of how the drug interacts with the DNA using a technique called X-ray crystallography at the European Synchrotron facility at Grenoble in France. Professor Miquel Coll says, In 1999 we solved the structure of the four-way DNA junction -also called Holliday junction- which is how two DNA helices can recombine (swop genetic information) and which is important in producing genetic diversity in humans and other organisms. But that junction was rather compact, without cavities or holes that could be used for drug binding. Now we have discovered that three-way DNA junctions are much more suitable for drug design: they leave a central cavity where a drug can fit perfectly and this opens a door for the design of new and quite unprecedented anti-DNA agents ...

This chapter focuses on the role of bacterial cell surfaces in catalyzing biomineralization processes and in promoting microfossil formation. Ultrathin sections were prepared by conventional embedding without the addition of osmium tetroxide or uranyl acetate as heavy-metal fixatives and contrasting agents. The electron density in these ultrathin sectioned samples as well as in the whole mounts presented in this chapter was provided by the naturally immobilized metals. The domains Bacteria and the Archaea represent the two major bacterial groups that have been identified based on 16S rRNA phylogenetic studies. The domain Bacteria is divided into the gram-positive or gram-negative groups based on cell envelope structure and chemistry (although gram-variable organisms also exist). The formation of many secondary minerals in natural as well as laboratory systems is catalyzed by microorganisms. These precipitation reactions have been divided into two general categories: passive and active mineralization.

Sampling of biofilm. Biofilm samples were analysed twice during the study period. To collect samples, the pipes containing biofilm developing devices were closed with valves before they were disconnected and the coupons were removed using sterile forceps. The filters in the mini tap filter devices were aseptically removed from the cartridge. Coupons were placed immediately into sterile 100-mL Schott bottles that contained water from the particular sampling site. Samples were transported on ice to the laboratory for analysis. Upon arrival in the laboratory, the coupons were removed from the bottles. Those coupons intended for scanning electron microscopy were stored in 100% alcohol and the remaining coupons were analysed for bacterial growth.. Scanning electron microscopy. The biofilm structure was investigated using scanning electron microscopy (SEM). Biofilm samples were fixed by 2.5% glutaraldehyde and 2% osmium tetroxide, dehydrated sequentially in increasing concentrations of ethanol (70%, ...

Lamellar phase refers generally to packing of polar-headed long chain nonpolar-tail molecules in an environment of bulk polar liquid, as sheets of bilayers separated by bulk liquid. In biophysics, polar lipids (mostly, phospholipids, and rarely, glycolipids) pack as a liquid crystalline bilayer, with hydrophobic fatty acyl long chains directed inwardly and polar headgroups of lipids aligned on the outside in contact with water, as a 2-dimensional flat sheet surface. Under transmission electron microscope (TEM), after staining with polar headgroup reactive chemical osmium tetroxide, lamellar lipid phase appears as two thin parallel dark staining lines/sheets, constituted by aligned polar headgroups of lipids. Sandwiched between these two parallel lines, there exists one thicker line/sheet of non-staining closely packed layer of long lipid fatty acyl chains. This TEM-appearance became famous as Robertsons unit membrane - the basis of all biological membranes, and structure of lipid bilayer in ...
The ability of proteins to catalyze hydrogen evolution has been known for more than 80 years, but the poorly developed d.c. polarographic pre-sodium wave was of little analytical use. Recently, we have shown that by using constant current chronopotentiometric stripping analysis, proteins produce a well-developed peak H at hanging mercury drop and solid amalgam electrodes. Peak H sensitively reflects changes in protein structures due to protein denaturation, single amino acid exchange, etc. at the picomole level. Unmodified DNA and RNA do not yield such a peak, but they produce electrocatalytic voltammetric signals after modification with osmium tetroxide complexes with nitrogen ligands [Os(VIII)L], binding covalently to pyrimidine bases in nucleic acids. Recently, it has been shown that six-valent [Os(VI)L] complexes bind to 1,2-diols in polysaccharides and oligosaccharides, producing voltammetric responses similar to those of DNA-Os(VIII)L adducts. Electrocatalytic peaks produced by Os-modified ...
1. The free border of the intestinal absorptive cells of cyprinids is nearly similar to that in tetrapods. This border has been described for four other species of fish and the possible evolution of the border from a ciliated epithelium has been described.. 2. The mitochondria of the absorptive cells are arranged in a supranuclear group of mainly rod-like bodies and a subnuclear group of mainly granular ones.. 3. The mitochondria are coarser and the Golgi element more readily impregnated with osmium tetroxide in the carnivorous Gobio than in the omnivorous Rutilus or the herbivorous Cyprinus. In all three fish both organoids are associated with fat absorption.. 4. The goblet cells secrete both mucus and zymogen. Their stomata are permanently open.. 5. Granular cells are described in Rutilus and Cyprinus, and in seven other species of fish, but they are absent from Gobio. Their varying reactions to numerous cytological techniques are described.. 6. Because of the volatility and extreme toxicity of its oxide, osmium is rarely used in its pure state, but is instead often alloyed with other metals for high-wear applications. Osmium alloys such as osmiridium are very hard and, along with other platinum-group metals, are used in the tips of fountain pens, instrument pivots, and electrical contacts, as they can resist wear from frequent operation. They were also used for the tips of phonograph styli during the late 78 rpm and early LP and 45 record era, circa 1945 to 1955. Osmium-alloy tips were significantly more durable than steel and chromium needle points, but wore out far more rapidly than competing, and costlier, sapphire and diamond tips, so they were discontinued.[53]. Osmium tetroxide has been used in fingerprint detection[54] and in staining fatty tissue for optical and electron microscopy. As a strong oxidant, it cross-links lipids mainly by reacting with unsaturated carbon-carbon bonds and thereby both fixes biological membranes ...

In this miniseries, we will be showcasing a number of different rarely mentioned elements, both in industry and day-to-day from the periodic table. Shining light on their true nature and amazing properties.. In this weeks instalment, a rarely mentioned noble metal Osmium (Os). Number 76 on the periodic table, only 3 doors next to everyones favourite precious metal, Gold (Au). Funnily enough, Os is considered as a platinum group metal. Additionally it is much rarer than gold, about 1000 times. Being the 2nd least abundant stable metal on planet earth (1st being Iridium, Osmiums neighbour). Discovered relatively recently compared to other metals, in 1803 by Smithson Tennant. The name Osmium comes from the Greek word osme meaning odour. Nowadays, the production of this metal comes as a by-product of Nickel (Ni) and Copper (Cu) mining. From mines in South Africa, Russia and Canada.. Interestingly, being about 1000 times rarer than Gold. Osmium is still not as expensive as Gold, being around ...
