Summary. Plant organogenesis requires control over division planes and anisotropic cell wall growth, which each require spatial patterning of cells. Polyhedral plant cells can display complex patterning in which individual faces are established as biochemically distinct domains by endomembrane trafficking. We now show that, during organogenesis, the Arabidopsis endomembrane system specifies an important additional cellular spatial domain: the geometric edges. Previously unidentified membrane vesicles lying immediately beneath the plasma membrane at cell edges were revealed through localization of RAB-A5c, a plant GTPase of the Rab family of membrane-trafficking regulators. Specific inhibition of RAB-A5c activity grossly perturbed cell geometry in developing lateral organs by interfering independently with growth anisotropy and cytokinesis without disrupting default membrane trafficking. The initial loss of normal cell geometry can be explained by a failure to maintain wall stiffness specifically ...
Plant hormones are important signaling molecules that control many developmental processes, including cell division, differentiation, organogenesis. In contrast to their animal counterparts, plant hormones can regulate a multitude of apparently unrelated physiological processes, their roles often overlap and they mutually modulate their effects. This indicates the important role of synergistic and antagonistic interactions between different plant hormones. However, the molecular basis of hormonal interactions is still largely unknown. The physiologically best characterized interaction is between auxin and cytokinin. These two plant hormones exhibit synergistic interaction to promote cell division in cell culture, but antagonistic interaction to regulate lateral root development or lateral bud outgrowth. The main aim of our studies is to reveal the molecular components and mechanisms balancing the output of auxin and cytokinin pathways in order to regulate plant organogenesis. We use lateral root ...

Plant biology, developmental biology and physiology, genetics, cellular and molecular biology University of Helsinki. Yrjö Helariutta is Professor of Plant Developmental Biology at the University of Helsinki and Research Director of the universitys Institute of Biotechnology. His research fields are plant biology, developmental biology and physiology, genetics, and cellular and molecular biology. Helariutta and his research team focus their research on the development of vascular systems of plants. They will investigate how the plant stem cell population, called vascular cambium, is genetically controlled to pattern and promote the formation of the vascular tissues in a tree trunk. The research team uses the Arabidopsis root procambium as a model for cambial development. Helariuttas team has greatly contributed to that Arabidopsis root development is widely seen as a key model for plant organogenesis.. During his term as Academy Professor, Helariutta will investigate cytokinin as a regulatory ...
Control of cell division and expansion is crucial for developmental patterning and is likely to be mediated by factors operating at different organisational levels (Irish and Jenik, 2001). Although some mutations causing altered division rates or aberrant cytokinesis affect overall plant morphogenesis (Liu et al., 1997), others do not (Blilou et al., 2002; Cleary and Smith, 1998). However, the increasing number of mutations reported to generate cytokinetic defects that lead to alterations in cell fate (Assaad et al., 1996; Chen and McCormick, 1996; Kessler et al., 2002) further demonstrate the importance of the precise timing and proper execution of cell division in plant organogenesis.. The maize seed is composed of an embryo and a persistent endosperm, and is proving an effective model system for the study of basic mechanisms of plant development. The endosperm is a simple structure that originates as a triploid central cell, the result of the fusion of the diploid central cell nucleus with ...
The uptake and metabolism of exogenous tritium-labelled benzyl adenine was studied during the shoot induction period of petunia leaf explants in tissue culture. Transfer experiments with Petunia `MD1 leaf explants (1 cm2) on MS media with 2.2 uM BA show that 27% and 100% of the leaf explants are committed to shoot induction on days 6 and 10, respectively. To study BA uptake and metabolism, leaf explants were placed on media containing tritium-labled BA for 1, 3, 6 and 10 days. BA was taken up from the media on days 1-6. BA metabolizes were analyzed using HPLC, a UV absorbancc detector and enzymatic techniques. Metabolizes produced include: BA, BAdo, BA 7G, BA 9G, BAdoMP, BAdoDP, BAdoTP and 3 unidentified compounds. BA and BAdo were detected on days 1 and 3 but not during day 6-10, the time of shoot induction. The pool of ribotide metabolites decreased from days 1 to 10, from 26.5% of all metabolites to 1.6%. Glucosylated compounds, BA 7G and BA 9G, increased continuously from 24.9% to 69.8% ...

The plants leaves are divided, narrow, terete and about 5 cm long. It was first described by Robert Brown in 1810. In 1891, German botanist Otto Kuntze published Revisio generum plantarum, his response to what he perceived as a lack of method in existing nomenclatural practice. Because Isopogon was based on Isopogon anemonifolius, and that species had already been placed by Richard Salisbury in the segregate genus Atylus in 1807, Kuntze revived the latter genus on the grounds of priority, and made the new combination Atylus formosus for this species. However, Kuntzes revisionary program was not accepted by the majority of botanists. Ultimately, the genus Isopogon was nomenclaturally conserved over Atylus by the International Botanical Congress of 1905. ...

Induction and emergence of hairy roots. A. rhizogenes strain K599 could induce hairy root formation in 47% excised leaf explants. The time period required for hairy root induction and frequency of transformation varied with respect to the age of explants. Leaf explants obtained from 3 and 4 weeks old in vitro cultures were only the responsive explants (Figure 1) and exhibited 46.6 and 35.0% TF after 25 and 28 days of bacterial infection respectively. Young leaf explants showed higher TF (47%) which decreased with an increase in explants age. A number of previous studies showed that type and age of explant have a great influence on hairy root induction since age of explant is a major factor that alters the physiological properties of the cell (Dupre et al. 2000). A strong correlation between TF and the age of wheat donor plants was observed when two different varieties of wheat were used for transformation with Agrobacterium. The mean TF increased from 0.7% to 5% using immature embryo from old ...

AU - Lin, Hui Li. AU - Wu, Chia Chang. AU - Shaw, Shyh Yu. PY - 2019/5. Y1 - 2019/5. N2 - The aim of this study is to discuss the non-catechin flavonoids (NCF)from Camellia sinensis (L.)O. Kuntze seed improving TNF-α impaired insulin stimulated glucose uptake and insulin signaling. Flavonoids had anti-metabolic syndrome and anti-inflammatory properties. It had widely been known for biological activity of catechins in tea, but very few research reports discussed the biological activity of non-catechin flavonoids in tea seed. We used HepG2 cell to treat with 5 μM insulin or with 5 μM insulin + 30 ng/ml TNF-α. Detecting the glucose concentration of medium, insulin decreased the glucose levels of medium meant that insulin promoted glucose uptake into cells, but TNF-α inhibited the glucose uptake effect of ...
Nitraria schoberi (Ghar-e-Daghin in Persian) from Zygophyllaceae (Nitrariaceae) family is considered as a wonderful exquisite plant in terms of its economical, medicinal, animal feed, significant values and also its unique characteristics of being tolerant to the soil salinity, drought and erosion. On the other hand, this plant is challenging with severe problems such as seed dormancy, plant growth inhibitors and also hard seed coat which lead to retard germination and subsequently to reduce the chance of life and reproduction. The present study was conducted for the first time to cope with its seed germination obstacles and to induce plantlet regeneration in N. schoberi through tissue culture method. Effect of cotyledonary leaf and hypocotyl explants and various concentrations of plant growth regulators were studied on shoot and root induction and further development. Based on the obtained results, culturing seeds after removing its seed coat effectively improved germination rate (80%) within a week.

Medicinal plants and their bioactive fractions have significant roles in various disorders. These fractions have been shown to improve the learning behaviors and memories of experimental animals [16]. For determination of disorders of the central nervous system in experimental animals, a passive avoidance task (fear-aggravated test) is used for evaluation of learning and memory. In the present study, on the 5th day, rats were placed in a dark chamber to evaluate their vision and neuronal activity. The results of the present study revealed that the values of STL differed significantly, whereas no change was found in IL values. Similar findings were reported by Ellman et al.[15] in their study on Vaccinium ashei supplementation in mice. Our results suggested that administration of Launaea procumbens significantly improved memory and learning in experimental animals when using passive avoidance tests. Similar results have been reported in other experimental animals [16]. The data of the present ...

Hansen 536 (Prunus dulcis × Prunus persica) is an important commercial rootstock for peach and almond. However, susceptibility to wet soil and bacterial canker has limited its use primarily to areas with less annual rainfall. Genetic engineering techniques offer an attractive approach to improve effectively the current problems with this cultivar. To develop an efficient shoot regeneration system from leaf explants, 10 culture media containing Murashige and Skoog (MS) or woody plant medium (WPM) supplemented with different plant growth regulators were evaluated, and adventitious shoot regeneration occurred at frequencies ranging from 0% to 36.1%. Optimal regeneration with a frequency of 32.3% to 36.1% occurred with WPM medium containing 8.88 µm 6-benzylamino-purine (BAP) and 0.98 to 3.94 µm indole-3-butyric acid (IBA). The regenerated shoots had a high rooting ability, and 80% of the in vitro shoots tested rooted and survived after being transplanted to substrate directly. Transient ...
Stentors. Light microscope footage of two Stentor sp. ciliate protozoa. Stentor species are filter-feeding freshwater protozoa which can reach lengths up to 2 millimetres, and are among the largest known unicellular organisms. A ring of prominent cilia surrounds the anterior part of the cell to sweep in food and aid in swimming. Some species live symbiotically with certain species of green algae. After being ingested, the algae live on and continue to photosynthesize, while their host absorbs nutrients they produce, and the algae receive protection and feed on the Stentors metabolic wastes. - Stock Video Clip K005/3589
