Phalaenopsis orchid is an important potted flower with high economic value around the world. We report the 3.1 Gb draft genome assembly of an important winter flowering Phalaenopsis KHM190 cultivar. We generated 89.5 Gb RNA-seq and 113 million sRNA-seq reads to use these data to identify 41,153 protein-coding genes and 188 miRNA families. We also generated a draft genome for Phalaenopsis pulcherrima B8802, a summer flowering species, via resequencing. Comparison of genome data between the two Phalaenopsis cultivars allowed the identification of 691,532 single-nucleotide polymorphisms. Proliferous protocorm-like bodies were obtained from shoot tips and axillary buds of V. hookeriana and shoot tips and in florescence of T. obtusum. Shoot tips and axillary buds of P. niveum grew into single or multiple plantlets. In florescence culture of T. obtusum developed into multiple branches of in florescences, calli or produced roots. Young leaf tip or leaf base of P. niveum and root of T. obtusum were unable to differentiate into callus. However, culture of young flower bud of P. niveum produced leaf after seven months. Paphiopedilum exul (Ridl.) Rolfe is one of terrestrial or lithophytic orchids also known as the ladys slipper orchid. The distribution is native around limestone mountain at western and eastern coasts of Thailand. The suitable conditions for P. exul seed germination were constructed. Seeds from 5-month-old fruits cultured on half-strength Murashige and Skoog agar medium (½ MS) at different pH levels (5, 5.6, 6.2, and 6.8) and supplemented with 10, 20, and 30 g L-1 sucrose. Seeds were cultured in dark conditions for 2 months and then cultured at 25±3°C under light intensity of 37 µmol m-1 s-1 for 16 h d-1 for 1 month. The results showed that different pH levels affected seed development to germination. The pH 5, 5.6, 6.2, and 6.8 gave seed germination of 28.07, 26.33, 21.75 and 19.12%, respectively. Phylogenetic relationships within the orchid subtribe Oncidiinae sensu Chase were inferred using maximum likelihood analyses of single and multilocus DNA sequence data sets. Analyses included both nuclear ribosomal internal transcribed spacer DNA and plastid regions (matK exon, trnH-psbA intergenic spacer and two portions of ycf1 exon) for 736 individuals representing approximately 590 species plus seven outgroup taxa. Based on the well resolved and highly supported results, we recognize 61 genera in Oncidiinae. Mimicry of oil-secreting Malpighiaceae and other floral syndromes evolved in parallel across the subtribe, and many clades exhibit extensive variation in pollination-related traits. Because previous classifications heavily emphasized these floral features, many genera recognized were not monophyletic.
