A simple and highly sensitive procedure is described enabling the simultaneous determination of biogenic polyamines (PAs) and their related monoacetyl derivatives in stomach tissue. The method is based on HPLC using octane sulfonate as an ion-pairing agent employed in acetate buffers at pH 4.5. The aim of this study was to investigate an in-capillary derivatization capillary electrophoresis (CE) technique that was performed to determine the concentration of gamma-aminobutyric acid (GABA) and alanine (Ala) in tea after being derivatized with o-phthaldialdehyde/2-mercaptoethanol (OPA/2-ME) to form fluorescence-labeled products. The conditions of labeled derivatization and CE separation were optimized and then applied to real sample analysis. The labeled derivatization with 20 mM OPA and 26.67 mM 2-ME (mol ratio = 0.75) at pH 10 offered the most sensitive detection, and the separation with 30 mM sodium tetraborate buffer (pH 10.0) under 21 kV achieved good selectivity within 14 min. The detections were linear in the range of 0.05-5 mu M with correlation coefficients (R-2) of 0.9995 and 0.9964 and with detection limits of 0.004 and 0.02 mu M for GABA and Ala, respectively. The simultaneous determination of 17 free amino acids and 8 biogenic amines in Alicante Monastrell wines was investigated for the first time. The quantification was carried out by using a RP-HPLC method. based on a pre-column derivatization with o-phthaldialdehyde (OPA) and fluorescence detection. From the results obtained it may be concluded that the most abundant free amino acids were Glu, Arg, Ala Asp, and Lys. None of the wine samples analysed had histamine (HIM) or Tyramine (TYM) levels above the limits considered as a possible toxic risk for healthy individuals. No measurable amounts of cadaverine (CAD) or methylamine (MEA) were found, showing no spoilage symptoms of sensory properties of the wines. Tryptamine (TRM) content was significantly higher in aged wines compared to young wines. However ethanolamine (ETA) content was lower. The bactericidal effect of disinfectants against biofilms is essential to reduce potential endoscopy-related infections caused by contamination. Here, we investigated the bactericidal effect of a high-level disinfectant, peracetic acid (PAA), against Staphylococcus aureus and Pseudomonas aeruginosa biofilm models in vitro. S. aureus and P. aeruginosa biofilms were cultured at 35 °C for 7 days with catheter tubes. The following high-level disinfectants (HLDs) were tested: 0.3% PAA, 0.55% ortho-phthalaldehyde (OPA), and 2.0% alkaline-buffered glutaraldehyde (GA). Biofilms were exposed to these agents for 1-60 min and observed after 5 min and 30 min by transmission and scanning electron microscopy. A Students t test was performed to compare the exposure time required for bactericidal effectiveness of the disinfectants.
