We investigated sequence-specific DNA alkylation using conjugates between the N-methylpyrrole (Py)-N-methylimidazole (Im) polyamide and the DNA alkylating agent, chlorambucil, or 1-(chloromethyl)-5-hydroxy-1, 2-dihydro-3H-benz[e]indole (seco-CBI). Polyamide-chlorambucil conjugates 1-4 differed in the position at which the DNA alkylating chlorambucil moiety was bound to the Py-Im polyamide. High-resolution denaturing polyacrylamide gel electrophoresis (PAGE) revealed that chlorambucil conjugates 1-4 alkylated DNA at the sequences recognized by the Py-Im polyamide core moiety. Reactivity and sequence specificity were greatly affected by the conjugation position, which reflects the geometry of the alkylating agent in the DNA minor groove. To investigate the biochemical and cellular basis for the rise in polymorphonuclear leukocyte (PMN) count during epinephrine administration, PMN from subjects receiving epinephrine were studied for their capacity to adhere to nylon wool fibers and endothelial cell monolayers. A set of aminoalkylamine substituted anthraquinones (AAQs) was successfully synthesised by aminating 1- or 1,5-chloroanthraquinones in a single reaction step. These substituted anthraquinones, 1C3 or 1,5C3, were tethered to minor groove binding polyamides with between 1 - 3 N-methyl pyrrole units (Py) to produce a library of Py-AAQ compounds. It was envisaged that these compounds could improve cytotoxicity through bimodal damage to DNA relative to the individual polyamide or AAQ moieties. A set of py-AAQ platinum complexes was also successfully synthesised by a platinum-mediated alkylamine to nitrile addition. This improved membrane is prepared using a cationic polymeric wetting agent in an aqueous solution containing a polyfunctional amine reactant which is interfacially polymerized with an acyl halide to form a thin film polyamide discriminating layer on a microporous support. Alternatively, the wetting agent can be applied directly to the substrate and the interfacial polymerization can occur on said treated substrate. The use of heat treatment to improve solute rejection and fouling resistance of a polyamide reverse osmosis (RO) membrane was investigated in this study. Heat treatment was carried out by immersing the membrane samples in Milli-Q water at 70 °C for a specific duration. Heat treatment (24 h) reduced the pure water permeability from 4.1 to 2.8 L/m2hbar but improved conductivity rejection from 95.5 to 97.0%. As a result, a correlation was observed between changes in the two parameters. Marginal changes in the membrane surface characteristics (i.e. zeta potential, hydrophobicity, chemistry and roughness) were observed as a result of heat treatment. Heat treatment significantly improved the fouling resistance property of the RO membrane. When the secondary effluent was filtrated at an elevated permeated flux, the virgin RO membrane exhibited 30% flux decline while the heat-treated membrane showed only 12% flux decline.
