Vaccinia virus thymidylate kinase, although similar in sequence to human TMP kinase, has broader substrate specificity and phosphorylates (E)-5-(2-bromovinyl)-dUMP and dGMP. Modified guanines such as glyoxal-dG, 8-oxo-dG, O(6)-methyl-dG, N(2)-ethyl-dG and N(7)-methyl-dG were found present in cancer cell DNA. Alkylated and oxidized dGMP analogs were examined as potential substrates for vaccinia TMP kinase and also for human TMP and GMP kinases. Molecular models obtained from structure-based docking rationalized the enzymatic data. All tested nucleotides are found surprisingly substrates of vaccinia TMP kinase and also of human GMP kinase. Interestingly, O(6)-methyl-dGMP is the only analog specific for the vaccinia enzyme. Thus, O(6)-Me-dGMP could be useful for designing new compounds of medical interest either in antipoxvirus therapy or in experimental combined gene/chemotherapy of cancer. These results also provide new insights regarding dGMP analog reaction with human GMP kinase and their slow ...

The reaction mechanism of phosphoryl transfer catalyzed by UMP/CMP-kinase from Dictyostelium discoideum was investigated by semiempirical AM1 molecular orbital computations of an active site model system derived from crystal structures that contain a transition state analog or a bisubstrate inhibitor. The computational results suggest that the nucleoside monophosphate must be protonated for the forward reaction while it is unprotonated in the presence of aluminium fluoride, a popular transition state analog for phosphoryl transfer reactions. Furthermore, a compactification of the active site model system during the reaction and for the corresponding complex containing AlF3 was observed. For the active site residues that are part of the LID domain, conformational flexibility during the reaction proved to be crucial. On the basis of the calculations a concerted phosphoryl transfer mechanism is suggested that involves the synchronous shift of a proton from the monophosphate to the transferred ...

Kinases control virtually all aspects of biology. Forty-eight human proteins have a kinase-like domain that lacks at least one of the conserved catalytic residues; these proteins are therefore predicted to be inactive and have been termed pseudokinases. Here, we describe exciting work suggesting tha …

Department of Medical Biochemistry and Microbiology, Uppsala University, PO Box 582, SE-751 23 Uppsala, Sweden. [email protected] Megalin is an integral membrane receptor belonging to the low-density lipoprotein receptor family. In addition to its role as an endocytotic receptor, megalin has also been proposed to have signalling functions. Using interaction cloning in yeast, we identified the membrane-associated guanylate kinase family member postsynaptic density-95 (PSD-95) as an interaction partner for megalin. PSD-95 and a truncated version of megalin were co-immunoprecipitated from HEK-293 cell lysates overexpressing the two proteins, which confirmed the interaction. The two proteins were found to be co-localized in these cells by confocal microscopy. Immunocytochemical studies showed that cells in the parathyroid, proximal tubuli of the kidney and placenta express both megalin ...
Signaling of TJ-related proteins was mainly described in nonendothelial cells, because of the lack of suitable in vitro systems of TJ-bearing ECs. However, the available results open new concepts for signaling events downstream of TJs in ECs (Figure 1).. Zonula occluden-1 (ZO-1), together with the homologous proteins ZO-2/-3, are members of the membrane-associated guanylate kinase homologues (MAGUKs), which exhibit a PDZ-binding domain in the C-terminus and a src homology region 3 (SH3). PDZ domains are known to mediate the anchorage of transmembrane proteins to the cortical actin cytoskeleton. ZO-1 exists in 2 splicing variants, characterized by the presence of the 80-amino acid α domain. The α+ isoform is present in epithelial cells, whereas the α− isoform is restricted to ECs and Sertoli cells.52 Interestingly, ZO-1 localizes to the nucleus in sparse or migrating cells when TJs are not or only poorly developed.53 The loss-of-function mutation of the Drosophila MAGUK family member ...

Tamou, coded for by polychaetoid, a Drosophila cell-cell junction-associated protein, is homologous to mammalian ZO-1, a member of the membrane-associated guanylate kinase homolog family. It is suggested that TAM is in an emc activating pathway. Mutation in tam gives a phenotype resembling mutation in emc. Mammalian ZO-1 can bind directly to the cytoskeletal element alpha-spectrin, and also binds occludin, an integral membrane protein at the tight junction. It is proposed that ZO-1 plays a role in the structural linkages between the tight junction and cytoskeletal networks. ZO-1 also colocalizes with cadherins in nonepithelial cells lacking tight junctions. Mutation in tam reduces the transcription of emc and causes enlargement of proneural clusters resulting in emergence of supernumerary precursor cells, and consequently in extra mechanosensory organs (Takahisa, 1996). Irregular facets (If) is a dominant mutation of Drosophila that results in small eyes with fused ommatidia. Previous results ...
TJP2 Full-Length MS Protein Standard (NP_004808), Labeled with [U- 13C6, 15N4]-L-Arginine and [U- 13C6, 15N2]-L-Lysine, was produced in human 293 cells (HEK293) with fully chemically defined cell culture medium to obtain incorporation efficiency at Creative-Proteomics. This gene encodes a zonula occluden that is a member of the membrane-associated guanylate kinase homolog family. The encoded protein functions as a component of the tight junction barrier in epithelial and endothelial cells and is necessary for proper assembly of tight junctions. Mutations in this gene have been identified in patients with hypercholanemia, and genomic duplication of a 270 kb region including this gene causes autosomal dominant deafness-51. Alternatively spliced transcripts encoding multiple isoforms have been observed for this gene.
Pak D.T., Yang S., Rudolph-Correia S., Kim E., Sheng M.. The PSD-95/SAP90 family of scaffold proteins organizes the postsynaptic density (PSD) and regulates NMDA receptor signaling at excitatory synapses. We report that SPAR, a Rap-specific GTPase-activating protein (RapGAP), interacts with the guanylate kinase-like domain of PSD-95 and forms a complex with PSD-95 and NMDA receptors in brain. In heterologous cells, SPAR reorganizes the actin cytoskeleton and recruits PSD-95 to F-actin. In hippocampal neurons, SPAR localizes to dendritic spines and causes enlargement of spine heads, many of which adopt an irregular appearance with putative multiple synapses. Dominant negative SPAR constructs cause narrowing and elongation of spines. The effects of SPAR on spine morphology depend on the RapGAP and actin-interacting domains, implicating Rap signaling in the regulation of postsynaptic structure.. Disks large homolog 5 is a protein that in humans is encoded by the DLG5 gene. This gene encodes a member of the family of discs large (DLG) homologs, a subset of the membrane-associated guanylate kinase (MAGUK) superfamily. The MAGUK proteins are composed of a catalytically inactive guanylate kinase domain, in addition to PDZ and SH3 domains, and are thought to function as scaffolding molecules at sites of cell-cell contact. The protein encoded by this gene localizes to the plasma membrane and cytoplasm, and interacts with components of adherens junctions and the cytoskeleton. It is proposed to function in the transmission of extracellular signals to the cytoskeleton and in the maintenance of epithelial cell structure. Alternative splice variants have been described but their biological nature has not been determined. DLG5 has been shown to interact with SORBS3. This gene is a member of a conserved family of genes that share a common DNA-binding domain, the T-box. T-box genes encode transcription factors involved in the regulation of numerous developmental processes. In mouse, the ortholog of this gene is expressed in the cerebral cortex, hippocampus, amygdala and olfactory bulb and is thought to play an important role in neuronal migration and axonal projection. In mouse, the C-terminal region of this protein was found to be necessary and sufficient for association with the guanylate kinase domain of calcium/calmodulin-dependent serine protein kinase. 3296 Despite the fact that 5-Fluorouracil (5FU) has been used to treat colorectal cancer (CRC) for the last four decades, the mechanism(s) of acquired resistance to this drug in the clinic is not well understood. In fact, 5FU has different biological effects depending on the dose administered and time of exposure to this drug e.g. short-term exposure to high doses of 5FU (bolus) results in resistance related to the effect of the drug on RNA metabolism. Our previous studies on pulse 5FU resistant HCT-8 lines suggest that resistance to this drug is associated with lower levels of UMP kinase (UMPK) in resistant cells as compared to parental sensitive cells. Moreover, tumor samples from hepatic metastasis from patients clinically resistant to "pulse" 5FU, have a high incidence of decreased expression of this enzyme, both mRNA and protein, as compared to tumor samples of patients not previously treated. In order to further confirm the role of UMPK in resistance, we over expressed UMPK cDNA in ...
For Koreans, Miyeok Guk is associated with birth. We eat it on our birthday. And mothers eat Miyeok Guk for weeks after giving birth. Its believed to revive and replenish the body after the labor of giving birth. But this also applies to miscarriages and abortions. I totally remember my mom eating Miyeok Guk after giving birth to my brothers. Memories of the youngest brother stands out the most because I was a little older then. My grandma made a huge pot of Miyuk Guk for my mom to eat. And it seems like she ate nothing but that for days, then weeks. And of course, it became what we (the family) ate at the time too. I am pretty sure the rest of us didnt eat Miyeok Guk every single day like my mom did, but we certainly did have our share. My mom makes Miyeok Guk for each of our birthdays. As much as I dont like her cooking, I eat it anyways. I make my own every now and then. Its rarely for a birthday, but just whenever I get the craving. I was browsing around Maangchis site last week and her ...

Membrane-associated guanylate kinase, WW and PDZ domain-containing protein 1 is an enzyme that in humans is encoded by the MAGI1 gene. The protein encoded by this gene is a member of the membrane-associated guanylate kinase homologue (MAGUK) family. MAGUK proteins participate in the assembly of multiprotein complexes on the inner surface of the plasma membrane at regions of cell-cell contact. The product of this gene may play a role as scaffolding protein at cell-cell junctions. Alternatively spliced transcript variants encoding different isoforms have been identified. CARMA1 belongs to the membrane-associated guanylate kinase (MAGUK) protein family, whose members function as molecular scaffolds for the localized assembly of such multiprotein complexes [reviewed in (6)]. Upon T cell activation by TCR and costimulatory molecule engagement, CARMA1 associates with a complex containing Bcl10 and MALT1 (Mucosa-Associated Lymphoid tissue lymphoma Translocation-associated gene 1; also known as MLT or Paracaspase) and recruits these proteins to lipid rafts of the immunological synapse, where they activate the IKK complex, leading to degradation of IκB and subsequent activation of NF-κB (7;8). The CARMA1/Bcl10/MALT1 complex functions similarly in B cells to activate NF-κB in response to BCR engagement (9). NF-κB controls the proliferation, differentiation and survival of B and T cells by activating the transcription of target genes, including various cytokines.. CARMA1 mutants have normal numbers and differentiation of B cells in the bone marrow, but IgDhighIgMlow ...
This study shows that NMDA receptor-driven plasticity, behavior, and signal transduction is mediated by the MAGUK protein SAP102. SAP102 was required for the induction or initiation of these signaling events consistent with its direct physical association with the receptor. Remarkable specificity in the signaling responses was observed, with only specific frequencies of synaptic activation requiring SAP102 and ERK pathways. Thus, the discrimination of different patterns of neuronal activity, all of which activate the NMDA receptor, is performed by distinct MAGUK proteins that then selectively engage downstream pathways. These results are consistent with published experiments showing MAGUK selectivity in relation to NMDA receptor subunits, downstream signaling, and plasticity effector mechanisms. Biochemical and electrophysiological evidence suggests that PSD-95 preferentially associates with NR2A-containing NMDA receptors, whereas SAP102 associates with NR2B-containing receptors (Sans et al., ...

For cellular organisms, the transport of specific molecules across compartmentalizing membrane barriers is essential in order to exchange content with their environment and with other individuals. For example, content exchange between individuals enables horizontal gene transfer, an important factor in the evolution of cellular life.[30] While modern cells can rely on complicated protein machineries to catalyze these crucial processes, protocells must have accomplished this using more simple mechanisms. Protocells composed of fatty acids[31] would have been able to easily exchange small molecules and ions with their environment.[1] Membranes consisting of fatty acids have a relatively high permeability to molecules such as nucleoside monophosphate (NMP), nucleoside diphosphate (NDP), and nucleoside triphosphate (NTP), and may withstand millimolar concentrations of Mg2+.[32] Osmotic pressure can also play a significant role regarding this passive membrane transport.[1] Environmental effects have ...
