Vaccinia virus thymidylate kinase, although similar in sequence to human TMP kinase, has broader substrate specificity and phosphorylates (E)-5-(2-bromovinyl)-dUMP and dGMP. Modified guanines such as glyoxal-dG, 8-oxo-dG, O(6)-methyl-dG, N(2)-ethyl-dG and N(7)-methyl-dG were found present in cancer cell DNA. Alkylated and oxidized dGMP analogs were examined as potential substrates for vaccinia TMP kinase and also for human TMP and GMP kinases. Molecular models obtained from structure-based docking rationalized the enzymatic data. All tested nucleotides are found surprisingly substrates of vaccinia TMP kinase and also of human GMP kinase. Tuberculosis (TB) is the primary cause of mortality among infectious diseases. Mycobacterium tuberculosis thymidylate kinase (TMPK(Mtub)) catalyzes the ATP-dependent phosphorylation of deoxythymidine 5-monophosphate (dTMP). Essential to DNA replication, this enzyme represents a promising target for developing new drugs against TB, because the configuration of its active site is unique within the TMPK family. Indeed, it has been proposed that, as opposed to other TMPKs, catalysis by TMPK(Mtub) necessitates the transient binding of a magnesium ion coordinating the phosphate acceptor. The computational results suggest that the nucleoside monophosphate must be protonated for the forward reaction while it is unprotonated in the presence of aluminium fluoride, a popular transition state analog for phosphoryl transfer reactions. 3296 Despite the fact that 5-Fluorouracil (5FU) has been used to treat colorectal cancer (CRC) for the last four decades, the mechanism(s) of acquired resistance to this drug in the clinic is not well understood. In fact, 5FU has different biological effects depending on the dose administered and time of exposure to this drug e.g. short-term exposure to high doses of 5FU (bolus) results in resistance related to the effect of the drug on RNA metabolism. Our previous studies on pulse 5FU resistant HCT-8 lines suggest that resistance to this drug is associated with lower levels of UMP kinase (UMPK) in resistant cells as compared to parental sensitive cells. Moreover, tumor samples from hepatic metastasis from patients clinically resistant to pulse 5FU, have a high incidence of decreased expression of this enzyme, both mRNA and protein, as compared to tumor samples of patients not previously treated. This study shows that NMDA receptor-driven plasticity, behavior, and signal transduction is mediated by the MAGUK protein SAP102. SAP102 was required for the induction or initiation of these signaling events consistent with its direct physical association with the receptor. Remarkable specificity in the signaling responses was observed, with only specific frequencies of synaptic activation requiring SAP102 and ERK pathways. Thus, the discrimination of different patterns of neuronal activity, all of which activate the NMDA receptor, is performed by distinct MAGUK proteins that then selectively engage downstream pathways.
