2,3,5,6-Tetrachloro-4-nitroanisole, an agricultural fungicide and acaricide, was selected for bioassay by the National Cancer Institute because of its structural similarity to pentachloronitrobenzene, a pesticide classified as tumorigenic by the Secretarys Commission on Pesticides and their Relationship to Environmental Health.. A bioassay for possible carcinogenicity of 2,4,5,6-tetrachloro-4-nitroanisole was conducted using Fischer 344 rats and B6C3F1 mice. 2,3,5,6-Tetrachloro-4-nitroanisole was administered in the feed, at either of two concentrations, to groups of male and female animals of each species. The high and low dietary concentrations used in the chronic bioassay were 0.012 and 0.006 percent, respectively, for both species. After a 104-week period of chemical administration, observation of rats continued for up to 3 weeks and observation of mice continued for up to 1 week. For rats 50 animals of each sex were placed on test as controls, while for mice 55 animals of each sex were ...

N2 - Noninvasive measurement of cardiac output (Q̇T) is problematic during heavy exercise. We report a new approach that avoids unpleasant rebreathing and resultant changes in alveolar PO2 or PCO2 by measuring short-term acetylene (C2H2) uptake by an open-circuit technique, with application of mass balance for the calculation of Q̇T. The method assumes that alveolar and arterial C2H2 pressures are the same, and we account for C2H2 recirculation by extrapolating end-tidal C2H2 back to breath 1 of the maneuver. We correct for incomplete gas mixing by using He in the inspired mixture. The maneuver involves switching the subject to air containing trace amounts of C2H2 ...

That said, with a typical 7-day fast were not talking about excessive amounts here, and if you lose a pound or two of muscle itll most likely come back within a month once youre off the fast. And lets not forget the obvious; You could be carrying pounds of dead weight in your midsection, which isnt exactly conducive to the coveted bodybuilding X-shape. This means a slew of bad things. I wanted to see what the experts in gaining/losing weight (bodybuilders) had to say. Bodybuilding.comâ and BodySpace® are trademarks of Bodybuilding.com. I responded with a 5 minute lecture about the need for a continuous supply of nutrients, the effect of protein deficiency on muscle mass and wrongly concluded that fasting was a generally bad idea except for religious purposes. Talk about working around the clock! Aim to get at least 1-2 gallons of pure drinking water in addition to the juices. Even if you stick to the recommended 24 to 72 hours, a water fast can still be detrimental to people with ...

Until I started receiving local British vegetables through my organic veg box scheme, I had only ever purchased cauliflower as a large white vegetable swaddled in crinkly translucent plastic at the supermarket. However, this weeks veg bag came with a huge green lump about the size and weight of a dodgeball. At first, I thought it was some sort of unknown English cabbage. However, upon peeling away a few of the thick, dusky green leaves, I discovered a tiny cauliflower the size of my fist nestled amongst the paler, thinner stalks. Ive never even seen so many cauliflower leaves before, much less a version of the vegetable that consisted of about 80% leaf and 20% flower.. This left me with an interesting challenge. While Im okay with the fact that organically grown vegetables are often smaller than their conventionally grown versions, the mini-cauliflower didnt leave me with much to cook for dinner. So, in the spirit of no waste and innovation in the kitchen, I decided to cook the cauliflower ...
ETHNOPHARMACOLOGICAL RELEVANCE: Shikonin, a naphthoquinone pigment abundant in the root of the Chinese herb Lithospermum erythrorhizon, has been widely used to treat inflammatory diseases for thousands of years. Whether shikonin changes drug metabolism remains unclear. AIM OF THE STUDY: We investigated whether shikonin modulates the expression of hepatic drug-metabolizing enzymes and transporters as well as the possible mechanisms of this action. MATERIALS AND METHODS: Primary hepatocytes isolated from Sprague-Dawley rats were treated with 0-2 μM shikonin and the protein and mRNA levels of drug-metabolizing enzymes and transporters as well as the activation of aryl hydrocarbon receptor (AhR) and NF-E2-related factor 2 (Nrf2) were determined ...
Rph1 and Gis1 are two related yeast zinc finger proteins that function as downstream effectors in the Ras/PKA, TOR and Sch9 nutrient signaling pathways. Both proteins also contain JmjC histone demethylase domains, but only Rph1 is known to be an active enzyme, demethylating lysine 36 of histone H3. We have studied to what extent the demethylase activity of Rph1 contributes to its role in nutrient signaling by performing gene expression microarray experiments on a yeast strain containing a catalytically inactive allele of RPH1. We find that the enzymatic activity of Rph1 is not essential for its role in growth phase dependent gene regulation. However, the ability of Rph1 to both activate and repress transcription is partially impaired in the active site mutant, indicating that the demethylase activity may enhance its function in vivo. Consistent with this, we find that the Rph1 mutation and a deletion of the histone H3 methylase Set2 affect the same target genes in opposite directions. Genes that ...
Induction of cytochrome P-450-dependent monooxygenases with phenobarbital (PB) or other hepatic drug-metabolizing enzyme inducers in the rat is associated with enhanced cocaine hepatotoxicity both in vivo and in cultured rat hepatocytes. To demonstrate whether the major PB-inducible P-450 subfamily (P-450IIB) could be involved in the metabolic activation of cocaine, rates of cocaine N-demethylation (the first step of cocaine bioactivation) and the rate of irreversible (covalent) binding of tritiated cocaine to hepatic microsomal proteins (a measure for the overall bioactivation) were determined in microsomes from saline or PB-pretreated rats. PB pretreatment augmented Vmax (6-fold), but not KM, of cocaine N-demethylation. Similarly, the rate of irreversible protein binding was 3-fold increased in microsomes from PB-pretreated rats as compared with those from saline controls. Addition of benzphetamine, a substrate of P-450IIB, markedly inhibited cocaine irreversible binding. In addition, various ...
Phenytoin is extensively bound to plasma proteins and is prone to competitive displacement. Phenytoin is metabolized by hepatic cytochrome P450 enzymes CYP2C9 and CYP2C19, and is particularly susceptible to inhibitory drug interactions because it is subject to saturable metabolism. Inhibition of metabolism may produce significant increases in circulating phenytoin concentrations and enhance the risk of drug toxicity. Monitoring of phenytoin serum levels is recommended when a drug interaction is suspected.. Phenytoin is a potent inducer of hepatic drug-metabolizing enzymes. ...
