Using a transporter mutant having ammonium-resistant nitrate transport activity as the genetic background, we analyzed specific effects of PII on in vivo nitrate reductase activity by measuring uptake of nitrate from the medium. Chromium and uranium are highly toxic metals that contaminate many natural environments. We investigated their mechanisms of toxicity under anaerobic conditions using nitrate-reducing Pseudomonas stutzeri RCH2, which was originally isolated from a chromium-contaminated aquifer. A random barcode transposon site sequencing library of RCH2 was grown in the presence of the chromate oxyanion (Cr[VI]O42-) or uranyl oxycation (U[VI]O22+). Strains lacking genes required for a functional nitrate reductase had decreased fitness as both metals interacted with heme-containing enzymes required for the later steps in the denitrification pathway after nitrate is reduced to nitrite. Cr[VI]-resistance also required genes in the homologous recombination and nucleotide excision DNA repair pathways, showing that DNA is a target of Cr[VI] even under anaerobic conditions. High rates of denitrification have been measured in Nevada geothermal hot springs, but little is known about the thermophiles that contribute to this activity. We hypothesize that heterotrophic bacteria in the genus Thermus are the most important denitrifiers in the springs. Alternatively, other microorganisms including chemolithotrophs may also be important. To test these hypotheses, several different strategies will be used to try to enrich and isolate nitrate-reducing microorganisms. Isolates will be identified by 16S rRNA gene PCR and sequencing. Subsequently, representative isolates will be chosen for nitrate reductase gene (narG) sequencing and for studies on the kinetics of nitrate reduction at high temperature. These data will provide information on how these microorganisms may behave in situ and how their activities may affect nitrogen cycling in the hot springs.

This chapter discusses the catabolism of amino acids and other nitrogen-containing compounds. Aspartate is transported into B. subtilis by two systems, a high-affinity system energized by the proton motive force and a low-affinity system. The enzymes of the arginase degradative pathway are found in B. subtilis and B. licheniformis. In B. subtilis and B. licheniformis, the proline-degradative enzymes are induced by proline. In B. subtilis, this induction is inhibited if the growth medium also contains glucose and amino acids. Hut expression in B. subtilis is induced by histidine and repressed by rapidly metabolized carbon sources such as glucose. Growth in the presence of amino acids severely inhibits synthesis of the Hut enzymes. Dehydrogenase enzymes may play a role in the degradation of phenylalanine in Bacillus badius, of valine in Streptomyces spp, and of leucine in B. cereus. cDNA clones for two isogenes of nitrate reductase (NR) have been isolated from rapeseed (Brassica napus L.) androgenetic haploid embryos induced by microspore culture. NR mRNA accumulation can be detected by northern hybridization at 14 d after culture initiation when embryos develop to the heart/torpedo-shaped stage. Whole-mount in situ hybridization experiments demonstrate that the mRNA accumulation is developmental stage specific. In addition, even when cultured in media containing no nitrate, embryos accumulated NR mRNA to almost the same level as the control.
