The rodent parasite Nippostrongylus brasiliensis (N. brasiliensis) models the salient features of helminth infection including skin penetration, migration from tissues to lung, maturation and egg production in the gut. As a potent activator of systemic and mucosal Th2 immune responses, Nippostrongylus brasiliensis has been extensively used to study host protective immunity and in vivo regulation of Th2 immune response. Six to eight week old C57Bl/6J, Balb/c mice or any other strains are suitable, as all are susceptible to infection. Inocula of 150-650 L3 larvae can be administered by subcutaneous injection, but for greatest consistency a dose of 550 L3 larvae is routinely used for experimental purposes. We have optimized three different protocols for the isolation of larvae from the lungs of mice infected with the L3 stage of Nippostrongylus brasiliensis. The intestinal epithelium is rich in γδ T cells and the gut is a site of residence for a wide variety of pathogens, including nematodes. Although CD4+ T-cell receptor (TCR) -αβ+ T helper type 2 T cells are essential for the expulsion of intestinal nematodes, little information is available on the function of γδ T cells in this type of infection. Here, we demonstrate two major functions of γδ T cells as a potently protective T-cell population against Nippostrongylus brasiliensis infection using γδ T-cell-deficient (TCR-δ(-/-) ) mice. First, γδ T cells are required to initiate rapid expulsion of adult worms from the intestine and to limit egg production. Nippostrongylus brasiliensis infections generate pulmonary pathologies that can be associated with strong TH2 polarization of the hosts immune response. We present data demonstrating N. brasiliensis-driven airway mucus production to be dependent on smooth muscle cell interleukin 4 receptor-α (IL-4Rα) responsiveness. At days 7 and 10 post infection (PI), significant airway mucus production was found in IL-4Rα−/lox control mice, whereas global knockout (IL-4Rα−/−) and smooth muscle-specific IL-4Rα-deficient mice (SM-MHCCre IL-4Rα−/lox) showed reduced airway mucus responses. The integrin α4β1 is expressed on a wide variety of peripheral leukocytes, and functions in recruitment of these cells to sites of inflammaiton. We infected august (AUG) rats with the helminth parasite Nippostrongylus brasiliensis and later used leukocytes from the peritoneum of the sensitized animals and antibodies against α4β1 integrin to localize that adhesion molecule on the cell surfaces after antigen challenge. We found that after antigen challenge, eosinophils, monocytes, and small lymphocytes, but not mast cells, expressed α4β1 integrin if the animals had been previously infected, but cells from uninfected animals did not show expression of this molecule. Our study describes a novel protocol for immunohistochemical analysis of rat eosinophils.
One of my colleagues did her honours (kind of like a one year practice PhD, I dont know if its done anywhere outside of Australia) looking at Nippostrongylus brasiliensis infection in mice. Apparently you can find them in the lung but nobody could work out how the worm got from the site of infection to the lung. Anywho, they used whole animal bioluminescent imaging to track the worms throughout infection. They were trying to do it in both real-time for the acute infection and by compiled photos to map the migration over longer time frames. CX3CR1GFP/PGRPdsRed reporter mice have previously been used to monitor trafficking of monocytes and neutrophils, respectively. With this new tool, we examined myeloid cell and neutrophil behavior during murine infection with the helminth, Nippostrongylus brasiliensis (Nb). Nb acutely infects the lungs leading to significant infiltration of PGRPdsRed neutrophils and CX3CR1GFP monocytes compared to naïve mice. Using ex vivo time-lapse confocal microscopy, we discovered that Nb infection increased chemotaxis of CX3CR1GFP monocytes and PGRPdsRed neutrophils. Because CX3CR1GFP expression is a loss of function knock-in mutation, we tested the function of CX3CR1 in the immune response to Nb. After inoculation with infective larvae, CX3CR1GFP mice were less susceptible to infection compared to wild-type mice and had lower worm counts and parasite eggs in the feces. Ym1 and Fizz1 are secreted proteins that have been identified in a variety of Th2-mediated inflammatory settings. We originally found Ym1 and Fizz1 as highly expressed macrophage genes in a Brugia malayi infection model. Here, we show that their expression is a generalized feature of nematode infection and that they are induced at the site of infection with both the tissue nematode Litomosoides sigmodontis and the gastrointestinal nematode Nippostrongylus brasiliensis. At the sites of infection with N. brasiliensis, we also observed induction of other chitinase and Fizz family members (ChaFFs): acidic mammalian chitinase (AMCase) and Fizz2. The high expression of both Ym1 and AMCase in the lungs of infected mice suggests that abundant chitinase production is an important feature of Th2 immune responses in the lung. Author Summary Retnla is a member of a family of cysteine-rich secreted proteins, referred to as resistin-like molecules or found in inflammatory zone that increase in expression during allergic reactions and following infection with a variety of metazoan parasites. Retnla was originally hypothesized to function as an effector molecule during helminth-induced Th2-type immune responses. Studies conducted here with Retnla-deficient mice, however, suggest that Retnla primarily functions as a regulatory molecule during helminth infection. Using three helminth model systems affecting three different organ systems, we show that Retlna is induced by IL-4 and IL-13 as a mechanism to suppress Th2-type immunity. Retnla deficiency increased inflammation in the lung following i.v. challenge with Schistosoma mansoni eggs. Five experiments were conducted comparing differential intestinal immune responses to two isolates of Eimeria acervulina (EA), EA1 and EA2. In three experiments, broiler chicks were divided into control (non-challenged), EA1, or EA2 challenged (14 days of age) groups. By using the combination of reverse-transcription PCR and rapid amplification of cDNA ends methods, a cDNA encoding mast cell tryptase was successfully cloned from the small intestine of Mongolian gerbil, Meriones unguiculatus, infected with Nippostrongylus brasiliensis. The cDNA was 1219 bp long including 810 bp of an open reading frame. Com a finalidade de demarcar mais precisamente o espectro de ação do albendazol, foi estudada a atividade terapêutica desse anti-helmíntico em ratos experimentalmente infectados com Strongyloides venezuelensis, tendo sido usada, como termo de comparação, a ação do cambendazol e do mebendazol, dois outros benzimidazólicos. Os três compostos mostraram-se eficientes quando utilizadas doses únicas de 6,75, 12,5, 25 e 50 mg/kg, pois motivaram desaparecimento total das formas adultas no intestino. Com a posologia de 5 mg/kg sucederam porcentagens médias de reduções dos números de vermes de 87%, 98% e 80%, respectivamente, como decorrência do emprego do albendazol, do cambendazol e do mebendazol, traduzindo superioridade da segunda droga citada..
