Spontaneous regression has been reported in some indolent forms of lymphoma. Mantle cell lymphoma (MCL) is an aggressive lymphoid neoplasm and has a poor prognosis. However, approximately 30% of MCL patients can exhibit indolent clinical behavior. To date, complete spontaneous regression of MCL has not been reported. We describe four cases of spontaneous regression of MCL. At the time of presentation, these patients were asymptomatic, with lymph node enlargement and mild to moderate fluorodeoxyglucose (FDG) uptake on FDG-positron emission tomography combined with computed tomography. One of the possible mechanisms of spontaneous regression of the tumor could be due to the host immune response through humoral and cellular immunity, which may have a role in the clearance of tumor cells. In this report, we support the use of a

On the basis of data from the literature, certain tumor types rarely regress, whereas others, such as testicular germ cell tumors and neuroblastoma, regress with a certain frequency. This regression can be partial or total. The present review refers only to cases of clinically complete regression.. Complete regression can occur in primary tumors as well as in metastases. The process of regression in the latter is relatively more common, particularly when one considers the regression of one or a few metastases in a patient with numerous metastases. This eventuality is relatively more frequent and clinically identified in the metastasis of renal carcinoma.. The processes involved in the spontaneous regression of tumors are mainly related to the process of apoptosis and the activity of the immune system, as well as to conditions in the tumor microenvironment. Therefore, such processes are occasionally linked more or less directly to the oncogenic suppressors of DNA.. Among tumors that present ...
Spontaneous regression of cancer is thought to be a rare event. Here, we report an extremely rare case of repeated episodes of spontaneous regression and progression of recurrent cervical adenocarcinoma. We report here a case of a 56-year-old Japanese woman who was diagnosed with cervical adenocarcinoma. Her hilar and mediastinal lymph nodes were swollen 6 years after the initial diagnosis and subsequent treatment, and were found to be pathologically malignant by mediastinal biopsy. Then, without any treatment, the hilar and mediastinal lymph nodes spontaneously regressed with decreases in tumor size and serum tumor marker levels, as confirmed by a decrease in uptake of fluorodeoxyglucose during positron emission tomography-computed tomography. Subsequently, although there were repeated episodes of increase and decrease in her serum tumor marker levels and lymph node size, her activities of daily living were and are well preserved. While spontaneous regression of a malignant tumor is a rare event, our

We investigated a new conservative management treatment for primary abdominal wall DTs. Methods. Data were collected from 147 patients between 1993 and 2012. The initial therapeutic approaches were categorized as front-line surgery [surgery group (SG), n = 41, 28 %] and initial observation or medical treatment [nonsurgery group (NSG), n = 106, 72 %]. The cumulative incidence of the last strategy modification was estimated using competing risk methods with variable censoring times. Results. Of the 147 ...

Infantile fibrosarcoma usually presents as a rapidly growing mass on the extremities or trunk. We describe spontaneous regression in a 5-month-old female infant with biopsy proven, molecularly confirmed, right leg infantile fibrosarcoma currently at 26 months of age with no signs of local recurrence. Previously reported cases of spontaneous regression are reviewed, suggesting a benign clinical course in some cases. Although evidence for spontaneous regression is anecdotal in this rare tumor type, physicians should weigh the risks and benefits of surgery and chemotherapy against watchful waiting ...
No, no, no, no! First off, the background reading was all about overdiagnosis; it has nothing to do with an increased incidence of breast cancer. Overdiagnosis, as you may recall, is the diagnosis of disease that, if left untreated, would never have progressed to trouble the patient or endanger her life. In other words, it has to do with detecting what is now being referred to as cancer of limited malignant potential. The speculation that mammography somehow resulted in a massive increase in cancer is not an interpretation listed in the study in question or even in the original studies cited. The question is, rather, what proportion of breast cancers detected mammographically constitute overdiagnosis. A helpful reminder of these concepts comes in the form of two posts Ive written.. Lets take a look at the study itself.. A study of this sort could only have been accomplished in a nation with a government-run health care system with extensive databases linking information from every hospital in ...

Background: In the past decade, a variety of immunotherapy approaches focused predominantly on the adaptive immune system have shown unprecedented responses in patients with advanced-stage malignancies. However, studies in spontaneous regression/complete resistance (SR/CR) mice and humans have shown a novel innate cancer-killing activity mediated by granulocytes, which is completely transferable for prevention or therapy against established malignancies. Methods: Three patients with advanced, relapsed or refractory solid tumors for which no standard therapy was available or was refused were enrolled into this ongoing combined phase I/II open label clinical trial testing the safety, dose tolerance, and possible antineoplastic efficacy of sequential infusions of HLA-mismatched non-irradiated allogeneic white cells (68-91% granulocytes) collected by leukapheresis from young, healthy donors (age 18-35) following mobilization with granulocyte colony stimulating factor (G-CSF) and dexamethasone ...
The idea that tumors could be recognized and rejected by the immune system was initially proposed in the late 19th century upon the observation that rare spontaneous tumor regressions occurred following infections, which may result in general activation of the immune system.1. More recently, preclinical studies revealed that tumors express immunogenic antigens and that these antigens induce immune responses that are able to protect against subsequent tumor challenges.2 Patients with melanoma were demonstrated to have T cells with specific activity against melanoma cells, and these cells were shown to be able to cause cancer regression when stimulated outside the body and returned to the patient.3-5. Over the last few years it has been recognized that the immune system in many patients has the ability to respond to cancer, but is usually prevented from doing so by immune defenses present in cancer cells. Many researchers and companies are studying a variety of means to overcome these defenses and ...
Our studies have led to the establishment of a variant of the ID8 ovarian tumor model ID8-VEGF that has allowed us to develop important new insight into why subcutaneously implanted ID8 tumors regress over time. To this end, our new data is consistent with an early infiltration of activated CD8 T-cells that contribute to controlling tumor growth, as depleting CD8 T-cells slowed spontaneous regression and enhanced tumor size. Our data suggest for the first time that ID8 and SKOV-3 tumor cells can generate the low molecular weight RGDKGE collagen fragment themselves. These novel findings suggest the presence of an autocrine signaling mechanism that may regulate YAP activity in these tumor cells. Collectively, these observations provide evidence for a role for a hippo-dependent signaling mechanism by which the XL313 collagen fragment regulates ovarian tumor growth. Taken together, our data provides new mechanistic insight that may help optimize the use of Mab XL313 to treat ovarian cancer.
Speaking of breast cancer, one topic I dont think anyone really understands is the spontaneous regression of tumors (when tumors disappear by themselves). Its very rare, but it does occur. Miracle healings! One minute youre told you have a tumor, the next minute its gone! Do modern day miracles still happen? I believe they do. Scientists have postulated various mechanisms by which tumors may disappear by themselves. One researcher lists these possible explanations: immune mediation, tumor inhibition by growth factors and/or cytokines, induction of differentiation, hormonal mediation, elimination of a carcinogen, tumor necrosis and/or angiogenesis inhibition, psychologic factors, apoptosis and epigenetic mechanisms. The list doesnt include supernatural healings, but psychologic factors? Im not so sure about that ...

Occasionally spontaneous regression takes place or response may be noted to inadequate chemotherapy suggesting some sort of immune reaction and in keeping ...

https://www.greenmedinfo.com/blog/when-cancer-spontaneously-disappears Have you ever wished that someones cancer would magically disappear? For dozens, maybe hundreds of patients each year, this is exactly what happens. Its called spontaneous regression, and it has medical science baffled. While it may sound like a fairytale or, at best, a one-in-a-million anomaly, cases of spontaneous healing from cancer have been documented in the scientific literature for hundreds of years -- with surprisi

Our outcomes demonstrated multiple positive correlations among all MMPs and ADAMTS-4 mRNA in herniated examples indicating their feasible synergistic impact in Identification herniation. MMP-9 and -13 mRNA amounts were significantly raised in individuals with persistent pain presumably because of neovascularization and persistent inflammation. Smoking practices were found to truly have a adverse dose-dependent influence on the transcript degrees of MMP-3 and KN-62 MMP-13 and an optimistic correlation with discomfort intensity recommending an unfavorable part ...
Last year when Sinclair attempted to acquire Tribune Broadcasting for a cool $3.9 billion, you might recall the company was accused of some highly dodgy behavior in order to get the deal done. Despite the FCC doing its best to neuter most media consolidation protections to help move the deal forward, the union would have still resulted in the merged company violating media ownership limits and dominating local broadcasting in a huge number of new markets. To get around those limits, Sinclair allegedly got, uh, creative. Consumer groups accused Sinclair of trying to offload several of its companies to Sinclair-owned shell companies to pretend the deal would remain under the governments ownership cap. The company also tried something similar in trying to offload some stations to friends and other partner companies at highly discounted rates, allowing it to technically not own -- but still control -- those stations. It was all so dodgy that even the Ajit Pai FCC, which had initially been doing ...

Ben Zayn briefs the command staff. Ivanova objects to the presence of Gray; regulations prohibit the use of telepaths in routine investigations. Ben Zayn tells her there are new regulations; all staff must submit to scans. Ivanova refuses, growing angry, but Sinclair stops her and says hell check the regulations himself. Ben Zayn attaches Garibaldi to his staff over Sinclairs objections, and the two of them leave Sinclair and Ivanova alone in Sinclairs office. Ivanova reiterates her refusal to submit to a scan. Garibaldi tells Ben Zayn he thinks the whole investigation is a sham. Ben Zayn orders Garibaldi to collect all the files -- including classified ones -- on the stations staff, and report back with them later that night. Lennier intercepts Garibaldi in the hallway and says hes managed to translate the manual. He offers to help with the bike; Garibaldi eventually agrees. In his office, Garibaldi instructs the computer to put together the files Ben Zayn requested. While thats going on, ...

Spontaneous remission, also called spontaneous healing or spontaneous regression, is an unexpected improvement or cure from a disease that usually progresses. These terms are commonly used for unexpected transient or final improvements in cancer. Spontaneous remissions concern cancers of the haematopoietic system (blood cancer, e.g. leukemia), while spontaneous regressions concern palpable tumors; however, both terms are often used interchangeably. The spontaneous regression and remission from cancer was defined by Everson and Cole in their 1966 book as the partial or complete disappearance of a malignant tumour in the absence of all treatment, or in the presence of therapy which is considered inadequate to exert significant influence on neoplastic disease. It has long been assumed that spontaneous regressions, let alone cures, from cancer are rare phenomena, and that some forms of cancer are more prone to unexpected courses (melanoma, neuroblastoma, lymphoma) than others (carcinoma). ...
RCC accounts for 3% of all cancers and its incidence is steadily increasing. RCC is the most common among renal tumors in adults, accounting for 85% of neoplasms arising in the kidney (3). Approximately one-fourth of RCC patients are known to have metastatic disease at the initial diagnosis (4). The lungs (50%), bones (49%), lymph nodes (6-32%), skin (11%), liver (8%) and brain (3%) are the most frequent sites of clinical metastases (5), and these patients generally have a poor prognosis, with a reported 5-year survival of 3-11% for unoperated patients (6).. In the era of immunotherapy, cytoreductive nephrectomy followed by interferon-α treatment increased overall survival (OS) in RCC patients compared with immunotherapy alone (7,8). In this era of molecular-targeted therapy, it was also reported that cytoreductive nephrectomy may provide an OS benefit in patients with metastatic RCC (9).. Spontaneous regression of metastases in RCC is rare, occurring in ,1% of all cases (10). In 1928, Bumpus ...
Left Image: Breast Cancer Awareness Month with Pink Ribbons. Courtesy of WIkimedia Commons.. Spontaneous Remission of Breast Cancer ?. One screening study reported by Welch in the Annals of Internal Medicine actually concluded that many small breast cancers spontaneously regress. Gina Kolata wrote a New York Times piece about it. Actually, spontaneous regression of breast cancer has been reported many times in the medical literature. Sir William Osler, a legendary and revered doctor reported 14 cases himself.. See this 1901 report: The Medical Aspects of Carcinoma of the Breast, with a Note on the Spontaneous Disappearance of Secondary Growths, OSLER W., American Medicine: April 6 1901; 17-19; 63-66.. Perhaps breast cancer remission was more common during his lifetime. I personally have seen a case of spontaneous regression of breast cancer documented by follow up MRI scan.. The real challenge is for medical science to investigate spontanous regression, and once understood, use it induce a cure ...
Regarding the surgical procedure, SR of cancer can be proven only in the excised specimens; thus, we suggest that it is necessary to resect the primary cancer with secured enough margins, and the dissection of lymph nodes should correspond to preoperative staging, even if there is SR of colon cancer.. Although many of the mechanistic details of SR of cancer remain unclear, the following have been suggested as factors: immunological, endocrine, metabolic, surgical, and postoperative events; elimination of a carcinogen or antigen; inhibition of angiogenesis; tumor necrosis; changes in oncogenes, growth factors, or cytokines; genetic and epigenetic factors; induction of benign differentiation and apoptosis; and psychological factors [3, 10]. Previous reports have described a central role for immune-mediated host responses in SR of solid tumors; for example, SR has been observed in many patients with carcinomas that contained significantly higher numbers of infiltrating immune cells (e.g., activated ...

The abscopal effect is mediated by a systemic anti-tumor immune response and reflects the regression of non-irradiated metastatic lesions at a distance from the primary site of irradiation. This review will focus on understanding the biological rationale behind the abscopal effect of radiotherapy (RT), which has a recently renewed interest as a result of the successes achieved with immunotherapy and RT in combination. Both RT and immunotherapy are standard components of modern treatment regimens. Combination of these two modalities results in an increased response in the irradiated lesions themselves and the metastatic regions distant from the site of irradiation. We will summarize the abscopal effect of radiotherapy, in particular, the synergistic effect of RT and immunotherapy.

Neuroblastoma is the most frequently diagnosed extracranial solid tumour in childhood. While a subset of tumours show spontaneous regression or complete remission following conventional treatment, a substantial number remain resistant to intensive multimodal therapies. Survival rates approaching 40% place high-risk neuroblastoma as one of the greatest challenges in paediatric oncology. This contemporary review provides an update on the diagnosis, risk stratification and management for this enigmatic tumour.. Neuroblastoma is the most common extracranial solid tumour in childhood and the most frequently diagnosed neoplasm during infancy.1 This malignant tumour consists of undifferentiated and/or differentiating cells originating from neural crest-derived sympathoadrenal precursors. Neuroblastoma is often described as enigmatic and unpredictable because of the broad spectrum of clinical behaviour ranging from life-threatening progression despite intensive treatment to complete spontaneous ...
OBJECTIVES: I. Determine the incidence of spontaneous regression of localized neuroblastoma in infants. II. Determine how many infants with neuroblastoma do not need chemotherapy. III. Evaluate the time course of regression by radiologic criteria and catecholamine metabolites. IV. Determine the reliability of risk estimation by molecular characteristics (N-myc amplification, CD44, del 1p) compared to clinical criteria. V. Evaluate whether reduced therapeutic toxicity results in a decrease in treatment-related deaths. VI. Correlate cytostatic drug levels with chemotherapy side effects.. OUTLINE: All patients undergo resection of the primary tumor and N-myc determination within 6 months of age, unless critically ill, then are treated according to risk. Patients with amplified N-myc or with indeterminate N-myc amplification but with other risk features are treated per protocol GER-NB90. Stage 4S patients who are critically ill or thrombocytopenic receive doxorubicin, vincristine, and ...
This book addresses neuroblastoma, a type of embryonic tumor derived from neural crest cells and one of the most frequent extra-cranial solid tumors in children. However, the term also describes a heterogeneous group of tumors, the prognosis of which can differ greatly according to age, stage and biology. Some forms undergo spontaneous regression, and some are cured by surgery alone or after chemo-reduction, while others exhibit extremely aggressive behavior. Their successful treatment is one of the best examples of tailored medicine, which involves close collaboration between pediatric surgeons, pediatric oncologists, radiologists, nuclear medicine specialists, biologists, oncogeneticists and radiotherapists. The book pursues a unique approach, as it combines most essential insights from all of these fields, together with key information regarding epidemiology, physiopathology and palliative care. The respective chapters were written by the leading international experts on neuroblastoma, and ...
Background: Neuroblastoma is a genetically heterogenic tumor diagnosed in childhood which exhibits broad clinical diversity ranging from rapid disease progression to spontaneous regression. Although amplification of the MYCN oncogene is one of the most prominent prognostic markers of this disease it is only present in around 25% of patients. Hence, additional molecular markers are needed for patients lacking MYCN amplification to enable better stratification of patient risk groups.. To date, genome wide association studies have identified few gene candidates in neuroblastoma patients that can be further developed for prognostic use in the clinic. DNA methylation is a major epigenetic mechanism for gene silencing in a wide range of human cancers and methylation sites may reveal new prognostic targets in neuroblastoma. Recent studies have suggested that the methylation status of specific gene promoters may be useful for clinically discriminating subgroups of children diagnosed with neuroblastoma. ...
The treatment of hemangiomas in infancy may be associated with significant morbidity. In addition to morbidity, an objective response cannot be obtained because of the absence of targeted therapeutic options. Herein, we present an infant with a segmental hemangioma and marked glucocorticoid toxicity due to prior steroid therapy that was successfully treated with propranolol. Propranolol was tolerated well and no side effects were observed during the treatment. The only problem to occur was disease recurrence following the withdrawal of propranolol at age 13 months, which was not within the age of spontaneous regression (generally considered as ,18 months ...
The non-Hodgkins lymphomas include a broad range of neoplasms derived from the T cells and B cells and their precursors in the lymphoid system. Although they are not among the most common cancers, the lymphomas have engendered a great deal of interest among researchers because of their interesting biology and responsiveness to therapy. The non-Hodgkins lymphomas include at least ten major subtypes of diseases with different morphologic characteristics and clinical behavior. Based upon survival characteristics, it is convenient to divide the lymphomas into three broad categories, low grade, intermediate grade, and high grade. The low grade lymphomas usually arise in middle age or older individuals (median age, 55 years). They are derived from B cells and often have a follicular architectural pattern. They usually present with advanced stages of disease, often by virtue of bone marrow involvement. Nevertheless, patients are usually asymptomatic and may even have spontaneous regressions of ...

In this study, we show in breast and colon carcinoma models that fractionated local radiotherapy to one palpable tumor can synergize with CTLA-4 blockade to induce antitumor T-cell immunity and inhibit a second palpable tumor outside the radiation field. This abscopal effect was not seen with radiotherapy alone. Although localized tumor irradiation by itself has been shown to enhance the generation of tumor-specific T cells in both preclinical models and patients, the therapeutic effects of this response remain undetermined (3, 19). Clearly, irradiation to the primary tumor was required in our models to induce growth inhibition of the secondary tumors outside the field because CTLA-4 blockade by itself was ineffective (Figs. 2 and 6). This is consistent with the hypothesis that radiation-induced immunogenic tumor cell death as well as its induction of danger signals contribute to generate an in situ vaccine (20-22). Whereas the response generated is not sufficient to be therapeutically ...
