No animal species is immune to the acute toxic effects of aflatoxins. Adult humans have a high tolerance for aflatoxin exposure and rarely succumb to acute aflatoxicosis,[23] but children are particularly affected, and their exposure can lead to stunted growth and delayed development, in addition to all the symptoms mentioned below.[4] High-level aflatoxin exposure produces an acute hepatic necrosis (acute aflatoxicosis), resulting later in cirrhosis or carcinoma of the liver. Acute liver failure is made manifest by bleeding, edema, alteration in digestion, changes to the absorption and/or metabolism of nutrients, and mental changes and/or coma.[23] Chronic, subclinical exposure does not lead to symptoms so dramatic as acute aflatoxicosis. Chronic exposure increases the risk of developing liver and gallbladder cancer,[24] as aflatoxin metabolites may intercalate into DNA and alkylate the bases through epoxide moiety. This is thought to cause mutations in the p53 gene, an important gene in ...

Endometriosis is a nonmalignant, but potentially metastatic, gynecological condition manifested by the extrauterine growth of inflammatory endometrial implants. Ten percent of reproductive-age women are affected and commonly suffer pelvic pain and/or infertility. The theories of endometriosis histogenesis remain controversial, but retrograde menstruation and metaplasia each infer mechanisms that explain the immune cell responses observed around the ectopic lesions. Recent findings from our laboratories and others suggest that retinoic acid metabolism and action are fundamentally flawed in endometriotic tissues and even generically in women with endometriosis. The focus of our ongoing research is to develop medical therapies as adjuvants or alternatives to the surgical excision of these lesions. On the basis of concepts put forward in this review, we predict that the pharmacological actions and anticipated low side-effect profiles of retinoid supplementation might provide a new treatment option ...
Since its first description more than 20 years ago osteopontin has emerged as an active player in many physiological and pathological processes, including biomineralization, tissue remodeling and inflammation. As an extracellular matrix protein and proinflammatory cytokine osteopontin is thought to facilitate the recruitment of monocytes/macrophages and to mediate cytokine secretion in leukocytes. Modulation of immune cell response by osteopontin has been associated with various inflammatory diseases and may play a pivotal role in the development of adipose tissue inflammation and insulin resistance. Here we summarize recent findings on the role of osteopontin in metabolic disorders, particularly focusing on diabetes and obesity.
A new study led by scientists at the University of Pennsylvania School of Veterinary Medicine has painted a clearer picture of the delicate arms race between the human immune system and a pathogen that seeks to infect and kill human cells. The research explores the strategies by which the bacterial pathogen Yersinia, responsible for causing plague and gastrointestinal infections, tries to outsmart immune cell responses and looks at the tactics used by the immune system to fight back.. The senior author of the paper, which appears online this week in Proceedings of the National Academy of Sciences, is Igor E. Brodsky, an assistant professor in the Department of Pathobiology at Penn Vet. He teamed with Penn Vets Naomi H. Philip, Annelise Snyder, Meghan Wynosky-Dolfi, Erin E. Zwack, Baofeng Hu, Louise FitzGerald and Elizabeth A. Mauldin. Alan M. Copenhaver and Sunny Shin of Penns Perelman School of Medicine were also co-authors.. The researchers set out to try to solve a seeming paradox. They ...
The mycotoxin zearalenone (ZEN) causes functional and morphological alterations in reproductive organs of pigs. In the field, diagnosis of ZEN-induced disorders is often challenging, as relevant feed lots are no longer available, or feed analysis results are not conclusive. Here, we report a field case of hyperestrogenism in newborn piglets. Surprisingly, more than 50 fungal metabolites were detected in hay pellets fed to gestating sows, including ZEN and its modified form zearalenone-14-sulfate (ZEN-14-S). Despite the broad contamination range in this unconventional feed component, a definite diagnosis of mycotoxicosis could not be achieved. In this context, current limitations regarding the confirmation of suspected cases of ZEN-induced disorders are discussed, covering both feed analysis and the biomarker approach. A piglet producer with 200 sows experienced a sudden increase in suckling piglet losses up to 30% by lower vitality and crushing. Predominant clinical signs were splay legs and signs of
Tachyzoite to bradyzoite development in Toxoplasma is marked by major changes in gene expression resulting in a parasite that expresses a new repertoire of surface antigens hidden inside a modified parasitophorous vacuole called the tissue cyst. The factors that control this important life cycle transition are not well understood. Here we describe an important transcriptional repressor mechanism controlling bradyzoite differentiation that operates in the tachyzoite stage. The ApiAP2 factor, AP2IV-4, is a nuclear factor dynamically expressed in late S phase through mitosis/cytokinesis of the tachyzoite cell cycle. Remarkably, deletion of the AP2IV-4 locus resulted in the expression of a subset of bradyzoite-specific proteins in replicating tachyzoites that included tissue cyst wall components BPK1, MCP4, CST1 and the surface antigen SRS9. In the murine animal model, the mis-timing of bradyzoite antigens in tachyzoites lacking AP2IV-4 caused a potent inflammatory monocyte immune response that ...
Abstract. Non-infectious inhaled microbial particles can cause illness by triggering an inappropriate immunological response. From the pathogenic point of view these illnesses can be seen to be related to on one hand autoimmune diseases and on the other infectious diseases.. In this review three such illnesses are discussed in some detail. Hypersensitivity pneumonitis (HP) is the best known of these illnesses and it has also been widely studied in animal models and clinically. In contrast to HP Pulmonary mycotoxicosis (PM) is not considered to involve immunological memory, it is an acute self-limiting condition is caused by an immediate toxic effect. Damp building related illness (DBRI) is a controversial and from a diagnostic point poorly defined entity that is however causing, or attributed to cause, much more morbidity than the two other diseases.. In the recent decade there has been a shift in the focus of immunology from the lymphocyte centered, adaptive immunity towards innate immunity. ...
Perennial ryegrass staggers (PRGS) is a neurological mycotoxicosis caused by the ingestion of lolitrem-B. In this study, seven horses split into two separate groups were exposed to lolitrem-B by feeding them perennial ryegrass seed and hay containing 2 ppm lolitrem-B. Paired data was collected prior to and after two weeks exposure to lolitrem-B including video-documented neurological examination, clinical examination, brainstem auditory evoked (BAEP) and magnetic motor evoked (mMEP) potentials, blood and cerebrospinal fluid, and a frusemide challenge. All horses developed tremor when exposed to lolitrem-B. The degree of tremor varied between individual horses and also depended on the level of activity, increasing during feeding and exercise. Using an ophthalmoscope a subtle, rapid (~5 Hz) tremor of the eyeball was detected in six of the seven horses. Subtle signs of ataxia were observed during handling, and motor dysfunction was exaggerated when blindfolded. Ataxia primarily involved a truncal ...

Aim: Traumatic brain injury (TBI) is a leading cause of mortality/morbidity and is associated with chronic neuroinflammation. Melanocortin receptor agonists including adrenocorticotropic hormone (ACTH) ameliorate inflammation and provide a novel therapeutic approach. We examined the effect of long-acting cosyntropin (CoSyn), a synthetic ACTH analog, on the early inflammatory response and functional outcome following experimental TBI.Methods: The controlled cortical impact model was used to induce TBI in mice. Mice were assigned to injury and treatment protocols resulting in four experimental groups including sham + saline, sham + CoSyn, TBI + saline, and TBI + CoSyn. Treatment was administered subcutaneously 3 h post-injury and daily injections were given for up to 7 days post-injury. The early inflammatory response was evaluated at 3 days post-injury through the evaluation of cytokine expression (IL1β and TNFα) and immune cell response. Quantification of immune cell response included cell counts of
Metabolic dysregulation is an early hallmark of neurodegenerative diseases including Alzheimers disease and age-related macular degeneration. Mapping metabolic adaptation with cellular resolution and tissue- wide context is crucial to define networks regulating neuronal survival, cell death progression, and immune cell response.. Computational Molecular Phenotyping (CMP) explores the amine metabolome (amino acids and amines). Technically, CMP metabolomics combines amine metabolite trapping, ultrathin microscopy (50-200 nm), immunodetection, pattern recognition, and clustering algorithms. Here we mapped the in situ distribution of over 30 core amine metabolites in retinal cells challenged by light-induced oxidative stress. Metabolomic profiles were phenotyped using ultrastructural, biochemical, and proteomic indices of oxidative stress.. CMP enabled precise visualization of ,30 metabolites in every retinal cell. CMP resolved and phenotyped metabolomic profiles to specific degeneration and ...
Background B cell activating factor (BAFF) plays an important role in the differentiation, survival and activation of B cells rheumatoid arthritis (RA). BAFF receptor (BAFFR) is a crucial receptor for the survival of mature B cells in developing B cells. Paeoniflorin-6-O-benzene sulfonate (CP-25) exerted its anti-inflammatory effects through regulating immune cells responses. The study was to investigate the regulatory effect of CP-25 on BAFF/BAFFR-nuclear factor of kappa B (NFkapaB) signaling in B cell of collagen induced-arthritis (CIA) mice. Methods Mice CIA was induced by injection of type II collagen. Arthritis index (AI) and swollen joint count (SJC) were assessed, spleen and joints histopathology were observed.B cells subsets, BAFF receptor were analyzed by flow cytometry. BAFF and immunoglobulin (Ig) levels were measured by protein antibody array. The expressions of TRAF2, MKK3, MKK6, p-P38, and p-NFkapaB65 in NF-kapaB signaling were analyzed by western blot ...

Acute aflatoxin exposure can cause death and disease (aflatoxicosis) in humans. Aflatoxicosis fatality rates have been documented to be as high as 40% in Kenya. The inclusion in the diet of calcium silicate 100 (ACCS100), a calcium montmorillonite clay, may reduce aflatoxin bioavailability, thus potentially decreasing the risk of aflatoxicosis. We investigated the efficacy, acceptability and palatability of ACCS100 in a population in Kenya with recurring aflatoxicosis outbreaks. Healthy adult participants were enrolled in this double-blinded, crossover clinical trial in 2014. Following informed consent, participants (n = 50) were randomised to receive either ACCS100 (3 g day-1) or placebo (3 g day-1) for 7 days. Treatments were switched following a 5-day washout period. Urine samples were collected daily and assessed for urinary aflatoxin M1 (AFM1). Blood samples were collected at the beginning and end of the trial and assessed for aflatoxin B1-lysine adducts from serum albumin (AFB1-lys). AFM1 ...
We examined the relationship between various health parameters and aflatoxin B1 (AFB1) albumin adduct levels in plasma. Design. A cross-sectional field study was conducted in four villages in the Ashanti region of Ghana. A survey on socio-demographic and health characteristics was administered to 162 volunteers and blood (20 ml) was donated by 140 participants. AFB1 albumin adduct levels, liver function, hepatitis B and C viruses (HBV, HCV) and malaria infections were determined. AFB1 levels ranged from 0.12 to 2.995 pmolmg21 albumin (mean¡standard deviation50.89¡ 0.46) and was categorized based on the median as low (,0.80 pmolmg21) or high (,0.80 pmol mg21) and used in the analyses. By multivariate analysis, significantly higher levels of AFB1 were obtained for participants who reported symptoms of acute aflatoxicosis: history of yellow mouth (odds ratio55.5, confidence interval51.04-29.07, p50.04); history of sore swollen stomach (odds ratio54.54, confidence interval51.28-15.62, p50.01). Fungi can affect animals, including humans, in several ways. A mycosis is a fungal disease that results from infection and direct damage. Fungi attack animals directly by colonizing and destroying tissues. Mycotoxicosis is the poisoning of humans (and other animals) by foods contaminated by fungal toxins (mycotoxins). Mycetismus describes the ingestion of preformed toxins in poisonous mushrooms. In addition, individuals who display hypersensitivity to molds and spores develop strong and dangerous allergic reactions. Fungal infections are generally very difficult to treat because, unlike bacteria, fungi are eukaryotes. Antibiotics only target prokaryotic cells, whereas compounds that kill fungi also harm the eukaryotic animal host.. Many fungal infections are superficial; that is, they occur on the animals skin. Termed cutaneous (skin) mycoses, they can have devastating effects. For example, the decline of the worlds frog population in recent years may be caused by the chytrid fungus ...

Apremilast (Otezla) is an oral medication approved for the treatment of moderate to severe plaque psoriasis. Phosphodiesterase-4 (PDE4) is an enzyme that regulates the levels of cyclic adenosine monophosphate (cAMP), a key modulator of immune cell responses. Apremilast is a PDE4 inhibitor, which results in increased intracellular cAMP levels to help modulate the balance between pro-inflammatory and anti-inflammatory mediators produced by immune cells. Apremilast regulates the production of inflammatory mediators such as interleukin-17 and tumor necrosis factor-alpha, naturally occurring proteins that regulate the immune system and are implicated in immune mediated inflammatory disorders. Apremilast first received FDA approval on March 21, 2004 for treatment of psoriatic arthritis. It is the first oral medication in the U.S. approved for the treatment of psoriatic arthritis with a recommended starting dose of 30 mg twice daily. This medication is particularly beneficial in patients with multiple ...

Due to their structure and properties it is hypothesized that they are also capable of binding LPS in vitro. Three trials were conducted to determine if they are indeed able to bind LPS. The first two trials used a fixed amount of LPS, the third trial used an in vitro fermentation model to create an acidosis and subsequent rise in LPS. Three substances were used: activated charcoal, montmorrilonite and Amadeite. From the first two trials it appears that activated charcoal and montmorrilonite are ...
Ulexite, like other borates, is a structurally complex mineral. The basic structure of ulexite contains chains of sodium, water and hydroxide octahedrons linked in endless chains. The chains are linked together by calcium, water, hydroxide and oxygen polyhedra and massive boron units. The basic boron unit has a formula of B5O6(OH)6 and a charge of negative three (-3). It is composed of three borate tetrahedrons and two borate triangular groups. Ulexite is found with the mineral borax and is directly deposited in arid regions from the evaporation of water in intermittent lakes called playas. The playas form only during rainy seasons due to runoff from nearby mountains. The runoff is rich in the element boron and is highly concentrated by evaporation in the arid climate. Eventually the concentration is so great that crystals of ulexite, borax and other boron minerals form and accumulate to great thickness. Specimens of ulexite may form a cotton ball tuft of acicular crystals. These can be ...
