Signaling by receptor tyrosine kinases (RTKs) typically involves binding of extracellular ligands and subsequent activation of intracellular kinase domain, which in turn mediates autophosphorylation and downstream signal transduction. However, several RTKs, such as ErbB3 (HER3) and Ryk/Derailed, lack key conserved (and catalytically important) residues in their intracellular kinase domain, and are thought to be inactive `pseudo-kinases. Much less is known regarding how these receptor tyrosine `pseudo-kinases mediate transmembrane signaling. The present study was undertaken to analyze the role of the MUC4 mucin in pancreatic tumor development and progression. Mucins are generally known to protect surface epithelia from injuries and infections under normal physiological conditions but often display deregulated expression in malignant cells (4 , 6 , 10) . We have previously reported that the human mucin gene MUC4 is aberrantly expressed in pancreatic tumors and cell lines but has no detectable expression in the normal pancreas (10) . The present in vitro and in vivo data reveal that the silencing of MUC4 expression results in altered tumor cell behavior, decreased growth, and a marked reduction in metastatic incidences in an orthotopic mouse model of pancreatic cancer. The high death rate of pancreatic cancer is attributed to the lack of reliable methods for early detection and underlying molecular mechanisms of its aggressive pathogenesis. Mucins are large glycoproteins that cover epithelial surfaces of the body. All mucins contain at least one PTS domain, a region rich in proline, threonine and serine. Mucins are also characterized by von Willebrand D (VWD) domains or SEA domains. We have developed computational methods to identify mucin genes and proteins based on these properties of the proteins. Using such methods we are able to characterize different organisms where genome sequence is available with respect to their mucin repertoire. We have here made a comprehensive analysis of potential mucins encoded by the chicken (Gallus gallus) genome. Three transmembrane mucins (Muc4, Muc13, and Muc16) and four gel-forming mucins (Muc6, Muc2, Muc5ac, and Muc5b) were identified. The gel-forming mucins are encoded within a locus similar to the corresponding human mucins. This suggests that there is a differential contribution of each subtype of interneuron in the regulation of the network. In particular, cholecystokinin (CCK) expressing interneurons differ in chemical features and their postsynaptic targets, yet the role of each of these subtypes in the regulation of neuronal network activity is poorly understood. In this study, we describe a subpopulation of CCK interneurons co-expressing the vesicular glutamate transporter 3 (VGlut3) that also shows tyrosine kinase ErbB4 receptor expression. Introduction: Pancreatic Cancer (PC) is the fourth leading cause of cancer-related deaths in the United States. Over 80% of human pancreatic ductal adenocarcinomas (PDA) overexpress MUC1, a transmembrane mucin glycoprotein. MUC1high tumors are aggressive and patients with such tumors have poor prognosis. Interestingly, MUC1high PDA tumors overexpress cyclooxygenase-2 (COX-2), an inducible pro-inflammatory enzyme known to promote tumor progression and metastasis. Bone is the most common site of metastasis for breast cancer, however the reasons for this remain unclear. We hypothesise that under certain conditions mammary cells possess osteomimetic capabilities that may allow them to adapt to, and flourish within, the bone microenvironment. Mammary cells are known to calcify within breast tissue and we have recently reported a novel in vitro model of mammary mineralization using murine mammary adenocarcinoma 4T1 cells. In this study, the osteomimetic properties of the mammary adenocarcinoma cell line and the conditions required to induce mineralization were characterized extensively. It was found that exogenous organic phosphate and inorganic phosphate induce mineralization in a dose dependent manner in 4T1 cells. Ascorbic acid and dexamethasone alone have no effect. 4T1 cells also show enhanced mineralization in response to bone morphogenetic protein 2 in the presence of phosphate supplemented media. Among several genes identified by SST-REX cloning, Endomucin is noteworthy, as it encodes a protein that belongs to the sialomucin family. CD34 is the prototypic member of this family and has been widely used as a marker of HSCs and/or progenitor cells and of endothelial cells. The highly conserved amino acid sequence, protein structure, and genomic organization of CD34, endoglycan, and podocalyxin suggest that they are closely related to each other (29). These molecules reportedly are expressed on HSCs and/or progenitor cells and on endothelial cells and are supposed to have redundant functions (29, 30). Although Endomucin exhibits a protein structure highly related to that of these three molecules, its genomic structure is distant, indicating a different phylogeny (19)..
