MCCORD, PER HILDING. Genetic, Genomic, and Transgenic Approaches to Understand Internal Heat Necrosis in Potato. (Under the direction of G. Craig Yencho and Bryon R. Sosinski.) Internal heat necrosis (IHN) is a physiological disorder of potato tubers. A multifaceted approach was followed to better understand IHN. This approach entailed three major projects, involving genetic, genomic, and transgenic techniques. The genetics approach involved developing linkage maps of tetraploid potato using AFLP and SSR markers, and identifying QTL for IHN. Our mapping population consisted of 160 individuals from a cross between ‘Atlantic’, an IHN-susceptible cultivar, and B1829-5, an IHN-resistant breeding line. Phenotypic data indicated that the distribution of IHN is highly skewed toward resistance. Early foliage maturity was positively correlated with reduced IHN. QTL for resistance to IHN were located in multiple years on chromosomes IV, V, and group VII of ...

The Chen lab studies mechanisms underlying cell-cell fusion, a fundamental cellular process in the conception, development and physiology of multicellular organisms. We approach this question using a multifaceted approach including genetics, molecular biology, biochemistry, biophysics, live imaging, super-resolution microscopy and electron microscopy. Starting with a forward genetic screen in Drosophila, we have identified multiple evolutionarily conserved core components of the myoblast fusion signaling cascade, and, more importantly, discovered a novel cellular mechanism underlying myoblast fusion. We show that myoblast fusion is an asymmetric process in which one cell invades its fusion partner using actin-propelled membrane protrusions to promote fusion pore formation. Building on insights we learned from myoblast fusion in vivo, we have reconstituted high-efficiency cell-cell fusion in an otherwise non-fusogenic, non-muscle cell line and uncovered a previously unrecognized function for the ...
Theres no magic bullet for perimenopausal skin problems. But that doesnt mean you have to accept midlife as the beginning of the end of a beautiful, radiant complexion. Like any complex condition, the acne and signs of aging that tend to occur during these years require treatment that addresses not only the visible symptoms that result, but also their underlying causes.. A holistic skin care regime combines gentle, yet powerful topical treatments and healthy lifestyle habits to help you achieve clear, younger-looking skin. This multifaceted approach not only helps restore skins strength, resilience, and youthful luster by improving blood circulation, boosting elastin and collagen production, and neutralizing free radicals, but also calms irritated, acne-prone skin with plant-derived organic extracts that promote hormonal balance and fight bacteria and inflammation. I call this treatment plan the HERS program to remind my clients to focus on the fundamentals of caring for their skin during ...
For example, while investigating pharmacologically accessible signaling pathways downstream of oncogenic RAS, we recently demonstrated that genetic ablation of the high affinity Cu transporter CTR1 responsible for Cu uptake resulted in decreased RAF-MEK-ERK signaling through loss of the interaction between Cu and the kinases MEK1/2. This is the first example demonstrating Cu directly regulates the activity of a mammalian kinase, and hence has opened up a new way to explore how metals interact with signaling pathways. Capitalizing on the dependence of oncogenic mutations in the RAS effector protein BRAF for MEK1/2 activity, a multifaceted approach was used to examine this new signaling mechanism in the context of BRAF mutation-positive cancer. Specifically, we reported that decreasing the levels of CTR1, or introducing mutations in MEK1 that disrupt Cu binding, decreased BRAFV600E-driven signaling and tumorigenesis. Furthermore, Cu chelators used in the treatment of Wilson disease decreased the ...

Alternative therapies for fibroids may be used to great effect and therefore are extremely popular as conventional treatment has severe limitations. The nature of fibroids implies that conventional medication can provide little apart from symptomatic relief which is why the reasons doctors recommend a wait and find outInch approach.. Fibroids form because of the subtle interaction of numerous secondary and primary factors in the human body. These may be the effect of a poor diet, through genes, lifestyle factors and loss of focus. Very frequently there arent any apparent causes which is virtually difficult to look for the precise causes within an individual lady.. One way of addressing this issue is by using a multifaceted approach which systematically eliminates every single known reason for fibroids, although strengthening and supplementing the defense mechanisms. These alternative therapies for fibroids could use the next protocols:-. * Extensive nutritional modifications. * Nutritional ...

With the rising incidence of neonatal abstinence syndrome (NAS) over the last decade, healthcare professionals are recognizing that close collaboration and standardized protocols aid in effectively diagnosing and treating newborns with drug withdrawal symptoms. Maternal use of prescription or illicit drugs during pregnancy can result in the newborn experiencing NAS, with symptoms of excessive or high-pitched crying, irritability, poor feeding, sleep problems, slow weight gain, and seizures. Pennsylvania Patient Safety Authority analysts identified 797 events involving newborns diagnosed with NAS that occurred from January 2005 through December 2014, with an increasing number of events reported each year. Healthcare professionals use a standardized scoring process, combinations of pharmacologic and nonpharmacologic interventions, and close care team collaboration to aid in the diagnosis and treatment of newborns with NAS. With the help of healthcare professionals, mothers and families can provide ...
An improved understanding of indoor air chemistry would significantly benefit human health. It is estimated that at least 15% of new-onset asthma cases can be attributed to workplace exposures. The 2011 American Thoracic Society report on work exacerbated asthma identified 1) better exposure assessment methods to identify and characterize the complex exposures in workplaces and 2) investigating pathophysiological mechanisms such as neurogenic inflammation and epithelial response to oxidant stress as two of the major research gaps for addressing work exacerbated asthma reduction. Unfortunately even though indoor air is identified as a significant link between occupational lung disease and its prevention/reduction, it has been poorly understood or characterized. Research in gas-phase and surface-phase chemistries, improved field and analytical sampling methods and mechanism-based toxicology may all be used to improve indoor air quality ...

There are more than 40,000 OB/Gyns in the US working in over 12,000 clinics. The recent expansion in treatment technology for intrauterine procedures and the demand by women for less invasive alternatives to hysterectomy and female sterilization has created a need for tools allowing gynecologists to diagnose and implement treatment in both the surgery center and office setting. Based on market research and recent global market analysis, we expect an annual rate of 6MM-8.5MM hysteroscopy procedures, diagnostic and therapeutic, that would occur in the US market, with an expected global CAGR of 4.5% (2016-2022).. When these procedures are performed in the office, reduced anesthesia may be used, which is a benefit for both physicians and patients. Clinical review suggests that with a multifaceted approach to office anesthesia/analgesia, most patients will tolerate and actually prefer both diagnostic and operative hysteroscopic procedures in the office.* Once approved for use, these can be achieved ...
Although noncommunicable diseases have caused a considerable burden on public health worldwide, infectious and parasitic diseases are still responsible for an unfathomable impact on human and animal health. Moreover, with the advent of molecular biology, new infectious and parasitic diseases are continuously being discovered, and emerging zoonoses are being identified. To address this complex scenario in toto, a holistic and multifaceted approach is required. This brings us to the concept of One Health Medicine, which is the order of the day in the field of public health. This concept invites public health professionals, human health and veterinary clinicians to adopt a global approach when dealing with health risks resulting from human-animal interactions. The book Human-Animal Medicine, Clinical Approaches to Zoonoses, Toxicants and Other Shared Health Risks comes to consolidate this approach, in practice.. The book is organized into 14 chapters authored by one or more of 23 contributors. ...

Atopic dermatitis (AD) is a common, chronic skin disorder that can significantly impact the quality of life of affected individuals as well as their families. Although the pathogenesis of the disorder is not completely understood, it appears to result from the complex interplay between defects in skin barrier function, environmental and infectious agents, and immune dysregulation. There are no diagnostic tests for AD; therefore, the diagnosis is based on specific clinical criteria that take into account the patients history and clinical manifestations. Successful management of the disorder requires a multifaceted approach that involves education, optimal skin care practices, anti-inflammatory treatment with topical corticosteroids and/or topical calcineurin inhibitors, the management of pruritus, and the treatment of skin infections. Systemic immunosuppressive agents may also be used, but are generally reserved for severe flare-ups or more difficult-to-control disease. Topical corticosteroids are the

Media education has the potential to reduce these harmful effects through the process of educating children, adolescents, and adults about media. Media education is defined as the study and analysis of mass media. A media-literate public is able to decipher the purpose and message of media rather than accepting it at face value. With an educated understanding of media images and messages, users can recognize medias potential effects and make good choices about their and their childrens media exposure.. Media education represents a multifaceted approach to understanding and eliminating the negative impact of media images and messages on young people. At the same time, it allows the positive and prosocial uses of media to be explored and appreciated. Media education includes the ability to access, analyze, evaluate, and produce media products. A media-educated person understands the following: all media messages are constructed; media messages shape our understanding of the world; individuals ...

Achieving greater blood pressure control requires a multifaceted approach that raises awareness of hypertension, uses effective therapies, and improves adherence. Particular classes of antihypertensive therapy have beneficial actions beyond blood pressure and studies have evaluated differences in
To help address industry problems, Panthee takes a multifaceted approach - part conventional breeding, part molecular marker-assisted selection (MAS). MAS isnt genetic engineering; its simply a breeding short-cut thats especially helpful when it comes to developing disease-resistant varieties using DNA-based markers.. In conventional breeding methods, scientists would inoculate plants with a disease to see which ones are resistant, but that runs the risk of spreading the disease, Panthee explains. However, with MAS, scientists can look for what are called markers - sequences of nucleotides that make up a segment of DNA - that are near the genes of interest in the genome.. To develop a tomato resistant to tomato mosaic virus, for example, Panthee has identified the molecular marker that is tightly linked with the TMV resistance gene, so as he breeds successive generations of plants, he selects only plants that contain that marker.. I keep on selecting plants that contain the TMV resistance ...
Abstract: The viral enzymes reverse transcriptase and protease are the current targets for antiretroviral therapy (ART). The use of a combination of inhibitors targeting these enzymes can reduce viral load for a prolonged period and delay disease progression. However, complications of ART, including the emergence of viruses resistant to current drugs, are driving the development of new antiretroviral agents targeting not only the reverse transcriptase and protease enzymes but novel targets as well. CCR5 and Integrase inhibitors have recently been approved for use in HIV treatment. The durg targets, which are under investigation, include CCR5 and CXCR4, maturation inhibitors, gene therapy, viral envelope protein (env). Novel virus that targets HIV-1-infected cells and Human deoxyhypusine synthase (DHS) are discussed as novel targets for HIV therapy.A multifaceted approach to ART, using combinations of inhibitors that target different steps of the viral life cycle, has the best potential for ...

We are using the nematode Caenorhabditis elegans to investigate aspects of nerve cell development and function. The wealth of developmental, anatomical, genetic, and molecular information available for C. elegans provides a powerful and multifaceted approach to these studies. Our work has focused on the study of a set of six neurons that are the sensory receptors for gentle touch (the touch cells), to address two questions: 1) How is neuronal cell fate determined? and 2) What is the molecular basis of mechanosensation, a sensory modality that underlies a variety of senses(e.g., touch, hearing, and balance)? End result was that I had nice teeth, but my bite was still bad and kept giving me problems, especially in the ears and jaw joints.. If you do a Google Search for Paula Deens dentist, you can see her testimony. He didnt say much about her latest controversy, so I didnt ask.. Because Brad teaches other dentists clinical dentistry and I teach communication, teamwork, etc., we did a trade out, and I spent two weeks in his office with his team, and he reworked my teeth and bite. What a great learning experience for both of us. Plus, I got new, beautiful teeth to the right bite.. Importance of bite ...

Rehabilitation of edentulous spaces in esthetic areas is a challenge to the clinician due to the loss of soft tissues. In these clinical situations, it would be desirable to evaluate and predict the gingival architecture to recover in the oral rehabilitation. To fulfill this need, the diagnostic wax should anticipate the final rehabilitation with the integration of hard and soft tissue. Thus, it is essential to produce a diagnostic waxup that integrates these two components that are simultaneously seeking to recreate the harmony of white and pink esthetic. This diagnostic waxup will be the basis for the creation of the provisional prosthesis and a soft tissue mock-up. After placing the provisional prosthesis in the mouth, the soft tissue mock-up can be applied to assess its esthetic impact at facial and intraoral level. Dentist and patient should objectively assess the appearance of the final result. After approval of this rehabilitation concept, the virtual surgical planning can be performed ...
