The anti-tumor effect of aconitine in melanoma cell line B16 has been studied in this paper. We found that B16 cells showed significantly reduced growth rates and increased apoptotic effects in the presence of aconitine. Furthermore, aconitine inhibited the PI3K/AKT and MAPK/ERK1/2 signaling pathways, thus regulating the levels of protein and mRNA of PCNA and apoptotic related signaling molecules. Above all, we found that aconitine showed an anti-melanoma effect in suppressing tumor growth in vivo. In conclusion, we show that aconitine may be a useful anticancer drug in the future.

The antioxidant capacity of the extracts was evaluated by DPPH radical scavenging activity (DPPH-RSA), ABTS radical scavenging activity (ABTS-RSA) and ferric reducing capacity (FRAP) assay. A central composite design (CCD) with four variables and five levels was employed for optimization of process parameters, and response surface plots were constructed in accordance with a second order polynomial model. Jerusalem artichoke (Helianthus tuberosus L.) produces substantial shoots not used as food. To test its potential as a sustainable bioenergy crop, we studied the effects of synthetic fertilizer and intercropped legumes as nitrogen (N) sources on the growth, aboveground biomass dry matter yield and energy qualities of this crop. Plant height, leaf area index (LAI), SPAD-value, biomass yield, ash content and mineral element composition were determined. Mean aboveground biomass yields were not significantly affected by N source (legume intercrops and synthetic fertilizer) and ranged from 13 to 17 t ha(-1). Remarkably, plants given no fertilizer yielded equally to plants given 90 N kg ha(-1). We have combined high-performance liquid chromatography (HPLC) separations using a monolithic column with acidic potassium permanganate and tris(2,2'-bipyridyl)ruthenium(II) chemiluminescence detection in a rapid and highly sensitive method to monitor the process of extracting opiate alkaloids from Papaver somniferum. Due to the high flow rates allowed with the monolithic column and the inherent selectivity of the chemiluminescence reactions, the four predominant alkaloids - morphine, codeine, oripavine and thebaine - were determined in less than 2 min. The results obtained with numerous process samples compared favourable with those of the standard HPLC methodology. Higenamine is a tetrahydroisoquinoline present in several plants that has β-adrenergic receptor agonist activity. Study of the biosynthesis of higenamine has shown the participation of norcoclaurine synthase, which controls the stereochemistry to construct the (S)-isomer. However, when isolated from nature, higenamine is found as the racemate, or even the (R)-isomer. We recently reported the isolation of higenamine 4′-O-β-d-glucoside. Herein, its (R)- and (S)-isomers were synthesized and compared to precisely determine the stereochemistry of the isolate. Owing to their similar spectral properties, determination of the stereochemistry based on NMR data was considered inappropriate. Therefore, a high-performance liquid chromatography method was established to separate the isomers, and natural higenamine 4′-O-β-d-glucoside was determined to be a mixture of isomers.

The chromosomal and molecular PCR-ISSR+RAPD pattern of high-mountain Aconitum sect. Aconitum in the Sudetes and Carpathians were analyzed to test whether the taxon has common markers in these two mountain systems. In the Sudetes the taxon forms an autopolyploid chromosome complex (2n = 32), and allopolyploid in the neighboring Western Carpathians. The chromosome Giemsa C banding pattern of the allotetraploid Carpathian A. firmum was found to be common to both Sudetic autopolyploid A. plicatum and diploid A. variegatum. The Sudetes are geologically older than the Carpathians, and it is argued that an ancient Sudetic taxon may have contributed to the genome of the Carpathian taxon. The Quaternary glaciations and corresponding range extensions of alpine floras may have facilitated their secondary contact(s).
