Treatments were 1) control (C); 2) injection of exogenous bovine somatotropin (bST); 3) TMR top dressed with 300 mg of monensin/day (M); and 4) monensin and somatotropin in combination (bST+M) during the last 28 days before expected parturition. A 500-mg subcutaneous injection of sustained release somatotropin was administered adjacent to the tail head at d -28 and -14 relative to expected calving. Diet and management were the same for all cows after parturition. Production and intake were measured daily until 63 d in milk. In experiment 1, 75 cows (30 primiparous) were fed controlled-energy, high-forage (CEHF) diets or were overfed energy (OVERFED) from dry-off to parturition, or CEHF for the first 40 d after dry-off, followed by OVERFED for the last approximately 20 d before parturition (2-stage). During the periparturient period, cows fed CEHF had lower lipid accumulation in the liver than OVERFED cows and there was no advantage to using the 2-stage feeding strategy. The rate of aggregation was dependent on the growth state of the cells, with growing cells aggregating more rapidly than growth-arrested cells. There was considerable variation between individuals in the rate at which cells aggregated but no differences were detected in the aggregation of normal and DMD cells measured by this method. To test whether angiotensinogen might be targeted to dense core secretory granules in cells containing a regulated secretory pathway, we expressed rat angiotensinogen in AtT-20 cells, a mouse pituitary cell line that has the demonstrated ability to correctly sort proteins to the constitutive or regulated pathway. We compared the pattern of secretion of angiotensinogen with that of endogenous adrenocorticotropin hormone, which is secreted by AtT-20 cells through the regulated pathway. When cells were incubated for 5 hours with dibutyryladenosine cyclic monophosphate or KCl, adrenocorticotropin hormone secretion was significantly higher than control, whereas monensin had no effect. In contrast, angiotensinogen secretion was markedly reduced by monensin, but no stimulation was observed with dibutyryladenosine cyclic monophosphate or KCl.
