This thesis describes the fabrication, characterization, and application of radiolabeled multifunctional microspheres for Cerenkov luminescence imaging and targeting of angiogenesis in cancer and atherosclerosis. Cerenkov luminescence imaging has gained attention as a new type of molecular imaging modality for its potential to bridge optical imaging with nuclear imaging. The engineered microsphere design focuses on intensifying the Cerenkov luminescence emission and shifting the emission wavelengths toward the near-infrared for better penetration of light in biological tissue. This includes the use of a high-refractive index oil core that increases the intensity of Cerenkov luminescence by lowering the Cerenkov radiation energy threshold, as well as near-infrared quantum dots that can be optically excited by Cerenkov luminescence and generate higher-wavelength optical emission. Background: Cerenkov luminescence imaging (CLI) provides a great potential for clinical translation of optical molecular imaging techniques through using clinical approved radiotracers. However, it is difficult to obtain the Cerenkov luminescence signal of deeper biological tissues due to the small magnitude of the signal. To efficiently acquire the weak Cerenkov luminescence, we developed an endoscopic Cerenkov luminescence imaging (ECLI) system to reduce the in vivo imaging depth with minimum invasion, and validated the system on small animal tumor models. Molecularly-targeted microbubbles (MBs) are increasingly being recognized as promising contrast agents for oncological molecular imaging with ultrasound. With the detection and validation of new molecular imaging targets, novel binding ligands are needed that bind to molecular imaging targets with high affinity and specificity. In this study we assessed a novel class of potentially clinically translatable MBs using an engineered 10th type III domain of human-fibronectin (MB-FN3VEGFR2) scaffold-ligand to image VEGFR2 on the neovasculature of cancer. The in vitro binding of MB-FN3VEGFR2 to a soluble VEGFR2 was assessed by flow-cytometry (FACS) and binding to VEGFR2-expressing cells was assessed by flow-chamber cell attachment studies under flow shear stress conditions. Although the scientific community continues to combat male bias in preclinical research, representation of women in late-phase clinical trials has improved markedly in recent years.21 The National Institutes of Health initiative to increase the use of female organisms and cells in preclinical research ensures that the scientific community as a whole will grow to fully address the health of both men and women. A major obstacle to the incorporation of female animals into studies, however, has been the belief that the rodent estrous cycle would render the study of these animals prohibitively expensive or difficult. In the past years, one of my research directions has been focused on the development of probes and technologies for Imaging of Metabolically Abnormal Degeneration (MAD) diseases, which include diabetes and obesity.
