In pre-clinical safety studies, drug-induced vascular injury (DIVI) is defined as an adverse response to a drug characterized by degenerative and hyperplastic changes of endothelial cells and vascular smooth muscle cells. Inflammation may also be seen, along with extravasation of red blood cells into the smooth muscle layer (i.e., hemorrhage). Drugs that cause DIVI are often discontinued from development after considerable cost has occurred. An in vitro vascular model has been developed using endothelial and smooth muscle cells in co-culture across a porous membrane mimicking the internal elastic lamina. Arterial flow rates of perfusion media within the endothelial chamber of the model induce physiologic endothelial cell alignment. Pilot testing with a drug known to cause DIVI induced extravasation of red blood cells into the smooth muscle layer in all devices with no extravasation seen in control devices. The image-based arterial geometries used in patient-specific arterial fluid-structure interaction (FSI) computations, such as aorta FSI computations, do not come from the zero-stress state (ZSS) of the artery. We propose a method for estimating the ZSS required in the computations. Our estimate is based on T-spline discretization of the arterial wall and is in the form of integration-point-based ZSS (IPBZSS). The method has two main components. (1) An iterative method, which starts with a calculated initial guess, is used for computing the IPBZSS such that when a given pressure load is applied, the image-based target shape is matched. (2) A method, which is based on the shell model of the artery, is used for calculating the initial guess. In the first part of the thesis, the computational meshes obtained from the medical imaging techniques are used for the computation of hemodynamic parameters associated with the rupture potency of the cerebral aneurysms. The main result is the computation within twenty geometries of aneurysms. It is shown that the aneurysm size has more important role in wall shear stress distribution than the fact whether the aneurysm is ruptured or unruptured. We analyze the performances of two types of Luenberger observers -- namely, the so-called Direct Velocity Feedback and Schur Displacement Feedback procedures, originally devised for elasto-dynamics -- to estimate the state of a fluid-structure interaction model for hemodynamics, when the measurements are assumed to be restricted to displacements or velocities in the solid. We first assess the observers using hemodynamics-inspired test problems with the complete model, including the Navier-Stokes equations in Arbitrary Lagrangian-Eulerian formulation, in particular. Then, in order to obtain more detailed insight we consider several well-chosen simplified models, each of which allowing a thorough analysis -- emphasizing spectral considerations -- while illustrating a major phenomenon of interest for the observer performance, namely, the added mass effect for the structure, the coupling with a lumped-parameter boundary condition model for the fluid flow, and the fluid dynamics effect per se. Although spiral waves are ubiquitous features of nature and have been observed in many biological systems, their existence and potential function in mammalian cerebral cortex remain uncertain. Using voltage-sensitive dye imaging, we found that spiral waves occur frequently in the neocortex in vivo, both during pharmacologically induced oscillations and during sleep-like states. While their life span is limited, spiral waves can modify ongoing cortical activity by influencing oscillation frequencies and spatial coherence and by reducing amplitude in the area surrounding the spiral phase singularity. During sleep-like states, the rate of occurrence of spiral waves varies greatly depending on brain states. These results support the hypothesis that spiral waves, as an emergent activity pattern, can organize and modulate cortical population activity on the mesoscopic scale and may contribute to both normal cortical processing and to pathological patterns of activity such as those found in epilepsy.

In this paper we consider the numerical solution of the three-dimensional fluid-structure interaction problem in haemodynamics, in the case of real geometries, physiological data and finite elasticity vessel deformations. We study some new inexact schemes, obtained from semi-implicit approximations, which treat exactly the physical interface conditions while performing just one or few iterations for the management of the interface position and of the fluid and structure non-linearities. We show that such schemes allow to improve the efficiency while preserving the accuracy of the related exact (implicit) scheme. To do this we consider both a simple analytical test case and two real cases of clinical interest in haemodynamics. In the present study, we developed a multiscale model capable of incorporating global cardiovascular properties into the simulation of blood flows in local vascular segments. The multiscale model was constructed by coupling a three-dimensional (3D) model of local arterial segments with a zero-one-dimensional (0-1-D) model of the cardiovascular system. An original hybrid gear model is introduced, which combines lumped parameter and finite elements along with a specific interface aimed at coupling mismatched discrete models. A mortar-based interface is presented, which eliminates the numerical errors induced by direct collocations between the tooth contact and gear body models. It is shown that the proposed interface can capture the instant contact conditions in the profile and lead directions for both spur and helical gears. A number of quasi-static and dynamic simulation results are presented, which illustrate the potential and practical interest of the methodology. Computational cardiac models provide important insights into the underlying mechanisms of heart function. Parameter estimation in these models is an ongoing challenge with many existing models being overparameterised. Sensitivity analysis presents a key tool for exploring the parameter identifiability. While existing methods provide insights into the significance of the parameters, they are unable to identify redundant parameters in an efficient manner. We present a new singular value decomposition based algorithm for determining parameter identifiability in cardiac models. Using this local sensitivity approach, we investigate the Ten Tusscher 2004 rapid inward rectifier potassium and the Mahajan 2008 rabbit L-type calcium currents in ventricular myocyte models. We identify non-significant and redundant parameters and improve the models by reducing them to minimum ones that are validated to have only identifiable parameters. Computational modelling of cardiovascular dynamics has received notable attention over the last two decades. The modelling approaches range from lumped parameter, zero-dimensional (0D) models of the circulation [1,2], to one-dimensional (1D) models of blood pressure and flow propagation [3-7], to three-dimensional (3D) fluid-structure interaction techniques [8-11]. Each approach has its own merits and limitations. Lumped parameter methods provide a computationally inexpensive, mathematically accessible and intuitive framework to study whole-system dynamics. However, they are not suitable for studying pulse propagation phenomena or complex flows. Nonlinear 1D methods can accurately describe pulse wave propagation phenomena in extensive vascular networks while keeping the computational cost down.
