Using a nude-mouse xenograft model, we show for the first time the antitumor activity of the HDACi panobinostat in three GISTs carrying distinctive oncogenic KIT mutations. Xenografts were obtained with the imatinib-sensitive GIST882 cell line or with biopsies from progressive patients on imatinib. According to the high mitotic index, all xenografts were highly aggressive. Mice received a 12-day treatment with panobinostat, imatinib, or combination of these drugs.. All GIST xenografts responded rapidly to panobinostat, early after treatment initiation. Panobinostat reduced significantly proliferation and increased apoptosis in all xenografts. The tumor cells were arranged in organoid clusters or sheets and composed of round to polygonal cells with central round nucleus, abundant eosinophilic or clear cytoplasm and distinct cell membrane (IMAGES 1, 2). Myxoid matrix was also present between the tumor cells (IMAGE 3). The tumor had increased cellularity with nuclear pleomorphism and significant mitotic activity (IMAGES 4, 5). The in vitro transformation responses of lymphocytes to stimulation with phytohemagglutinin (PHA) and smallpox vaccine (vaccinia) were studied in cells from 20 patients with ocular and malignant diseases receiving chemotherapy. The transformation of lymphocytes to lymphoblast-like cells was reduced from 71% in the pretreatment PHA-stimulated cultures to 1.5% during therapy. The response to vaccinia was reduced from 12% before therapy to 0% during therapy. The mitotic indices fell from 1.5% (PHA) and 1.2% (vaccinia) to 0% for each during therapy.. Intensive combination therapy with parenteral 6-mercaptopurine and methotrexate, with or without prednisolone completely abolished transformation after 3 days of treatment. Since hemodynamic factors have been implicated in the localization and pathogenesis of atherosclerosis, the effect of hemodynamic stress on endothelium was investigated. The endothelium of the inferior vena cava of 24 rabbits with aortocaval fistulas was stained in situ and examined by the Hautchen technique at postoperative intervals ranging from 1 to 50 weeks. Cell counts revealed that, in the anastomosed vein of the fistula, an initially high frequency of abnormal cells decreased 3-12 weeks postoperatively but rose again within 20-50 weeks to levels significantly higher than those in either normal venous endothelium or sham-operated veins. An originally high mitotic index at 1-2 weeks decreased with time but still remained significantly larger than that observed in sham-operated rabbits. In most rabbits with arteriovenous shunts, a small area of the inferior vena cava near the fistula was devoid of endothelium, covered with a thin deposit of mural thrombi, and considered to be a jet lesion. ...

Denervated forelimbs and contralateral innervated forelimbs of Ambystoma larvae were injured internally distal to the elbow by compression with watchmakers forceps. Innervated controls completely repaired the crush injury within one week; denervated limbs failed to repair the injury and exhibited varying degrees of limb regression. This study was done to determine the severity and extent of abnormalities of duodenal mucosal architecture in non-specific (non-ulcerative) and ulcer-associated duodenitis. The effect of successful treatment with cimetidine on these changes has also been assessed. A method of microdissection and measurement of villus height, crypt depth, and mitotic figure count per crypt was applied to endoscopic biopsies from the duodenum. Five groups of patients were studied: untreated ulcer-associated duodenitis, untreated non-specific duodenitis, healed ulcer-associated and non-specific duodenitis after cimetidine treatment, and controls. Significant reduction in villus height, increase in crypt length, and increase in mitotic figure count per crypt were all found in both ulcer-associated and severe non-specific duodenitis as compared with controls. DISCUSSION. The present study retrospectively evaluated the prognostic factors and survival of a patient cohort diagnosed with gastrointestinal stromal tumors that underwent surgery in a Turkish clinic over a 16-year period. Patient survival was associated with GIST Miettinen risk group, tumor location and recurrence. Furthermore, Ki-67 and mitotic indices were identified as poor prognosis factors while C-kit was identified as a good prognosis factor. These results are by and large in agreement with previous reports, with the exception of tumor size and gender, which were not associated with survival in the present study. For this aim, music samples from Wagner, Mozart, Musorgsky, (Boris Godunov) Chopin, Tchaikovski, Schubert were chosen. We found correlation between root elongation and Mitotic Index (MI). Both kinds of music have positive effects on root growth and mitotic divisions in onion root tip cells but rhythmic dynamically changing lyrics affected much better. We have shown here that cultured primary human DMVEC cultures can be infected at different efficiencies by KSHV virions derived from several PEL cell lines. The infection causes a phenotype involving aligned and swirling bunches of elongated spindle-shaped cells that closely resembles that displayed by the pathogenic spindle cells of nodular KS lesions. Importantly, KSHV infection of the DMVEC cultures and the associated spindle cell conversion phenotype correlate almost completely with constitutive LANA1 protein expression in the same cells. Recent studies by Dupin et al. (10) have also confirmed that almost all spindle cells in late nodular KS lesions express LANA1. The similarity to the latent state found in both PEL cell lines and KS lesions is emphasized by the punctate nuclear character of the LANA1 signal (4), implying that the viral genomes also establish a multicopy episomal state in infected spindloid DMVEC. Major component of the study was the radiobiological characterization of murine LLC1 cells in vitro. These cells are culture transferred single cells emanated from a widely used in vivo mouse tumor model. The solid, undifferentiated tumor clusters in vivo corresponded to a non-small cell lung cancer, the culture transferred individual cells were classified as malignant due to nuclear pleomorphism, hyperchromasia, atypical mitoses and moved nuclear- cytoplasmic ratio. If obtained under optimal conditions in culture the LLC1-cells showed a fast, stable growth pattern - with a doubling time of 16 hours- even over long culture periods, but reacted very sensitively to changes in the cell culture environment, such as composition of the growth medium, seeding density or type and coating of the growth surface.
