Microsporidia are obligate intracellular parasites that infect eukaryotic cells and have emerged as major opportunistic human pathogens. Due to the difficulties in definitive laboratory diagnosis and insufficient knowledge, ocular microsporidiosis is infrequently reported in India. To improve diagnostic facilities, we have developed a novel duplex PCR (dPCR) for the simultaneous identification of both genera and species of isolates with microsporidian aetiology that cause keratitis. The material scraped from the corneas of 12 clinically diagnosed microsporidial keratitis patients was subjected to routine microbiological examinations and molecular diagnosis using a novel dPCR that targeted the small-subunit rRNA gene (SSU-rRNA) of microsporidia and Vittaforma corneae using genus- and species-specific primers. Of the 12 corneal scrapes, 6 showed positive results in smears, while dPCR provided positive amplification with both pan-microsporidial and V. corneae species-specific primers for 9 corneal scrapes.

To date, surveys of bumble bee pathogens in the United States have only been conducted on workers and males, yet the health of a population is ultimately dependent on the success of colony-founding queens. We conducted a molecular-diagnostic survey of the prevalence of N. bombi and trypanosomatids, such as Crithidia bombi, in six species of spring queens (n = 142) collected in 2011 and 2013 at three sites in central Kansas. Nosema bombi was found in 27% of Bombus pensylvanicus and 13% of B. auricomus but was not found in the other species sampled. Trypanosomatids were only found in B. pensylvanicus (9%) during the May 2013 sampling period. The high prevalence of N. bombi in B. pensylvanicus is consistent with other surveys for this pathogen in other castes, but the high prevalence of N. bombi in B. auricomus is a novel finding. ...

The microsporidian Encephalitozoon cuniculi possesses one of the most reduced and compacted eukaryotic genomes. Reduction in this intracellular parasite has affected major cellular machinery, including the loss of over fifty core spliceosomal components compared to S. cerevisiae. To identify expression changes throughout the parasites life cycle and also to assess splicing in the context of this reduced system, we examined the transcriptome of E. cuniculi using Illumina RNA-seq. We observed that nearly all genes are expressed at three post-infection time-points examined. A large fraction of genes are differentially expressed between the first and second (37.7%) and first and third (43.8%) time-points, while only four genes are differentially expressed between the latter two. Levels of intron splicing are very low, with 81% of junctions spliced at levels below 50%. This is dramatically lower than splicing levels found in two other fungal species examined. Watershed clearcut logging is the most widespread land-use modification to which Canadian boreal lakes are exposed. The present study investigated whether a stress-response of increased Thelohania can be used as an early biomonitor of the health and viability of crayfish (Orconectes virilis) inhabiting such watersheds. Prevalences remained low (|1.5%) in 4 coldwater fishery lakes over an 8-year period irrespective of experimental clearcutting.
