Two Cellmatics (Difco) Herpes simplex virus (HSV) detection systems, one with mink lung cells (ML) and the other with primary rabbit kidney cells (PRK) and the Cultureset (Ortho) system with Vero cells were compared for their ability to detect previously positive specimens. One hundred fifty patients specimens positive for HSV in either Vero or PRK cells prior to freezing at -70 degrees C were thawed once, diluted to 1:8, and inoculated in duplicate into each cell system. Positive cultures were detected using each kits immunoperoxidase method. All three cell lines were stained at 24 hr followed by a final staining of negative cultures at 48 hr for Cultureset and at 72 hr for Cellmatics as per kit instructions. At 24 hr percent detection was: Cellmatics (ML) = 85%, Cellmatics (PRK) = 81%, Cultureset (Vero) = 74%. At final staining percent detection was 94%, 92%, and 84%, respectively. The present study was under taken to assess the effectiveness of cleaning procedure being used in one of the WHO approved Pharmaceutical plant in Gujarat having common facilities for manufacture of dosage forms of Beta Lactam group of antibiotics where there is a risk of potential hazards of cross contamination. Pantoea agglomerans is an ecologically diverse taxon that includes commercially important plant-beneficial strains and opportunistic clinical isolates. Standard biochemical identification methods in diagnostic laboratories were repeatedly shown to run into false-positive identifications of P. agglomerans, a fact which is also reflected by the high number of 16S rRNA gene sequences in public databases that are incorrectly assigned to this species. Several mosquito-borne diseases affecting humans are emerging or re-emerging in the United States. The early detection of pathogens in mosquito populations is essential to prevent and control the spread of these diseases. In this study, we tested the potential applicability of the Lawrence Livermore Microbial Detection Array (LLMDA) to enhance bio-surveillance by detecting microbes present in Aedes aegypti, Aedes albopictus and Culex mosquitoes that are major vector species globally, including in Texas. The sensitivity and reproducibility of the LLMDA was tested in mosquito samples spiked with different concentrations of dengue virus (DENV) revealing a detection limit of |100 but |1000 pfu|/mL. Additionally, field-collected mosquitoes from Chicago, Illinois and College Station, Texas of known infection status (West Nile virus (WNV) and Culex flavivirus (CxFLAV) positive) were tested on the LLMDA to confirm its efficiency. In interviews it was found that reusable bags are seldom if ever washed and often used for multiple purposes. Large numbers of bacteria were found in almost all bags and coliform bacteria in half. Escherichia coli were identified in 12% of the bags and a wide range of enteric bacteria, including several opportunistic pathogens. When meat juices were added to bags and stored in the trunks of cars for two hours the number of bacteria increased 10-fold indicating the potential for bacterial growth in the bags.
