The first thing we wanted to demonstrate was co-culture growth in which an E. coli culture was able to grow on the carbon shared by Synechocystis. We inoculated 48-well plates with different starting ratios of Synechocystis and E. coli. We used both the lactate-producing Synechocystis strains whose genetic instability we showed earlier, as well as the acetate-producing strain based on the acs knock-out. Cells were counted using a coulter-counter before innoculation and re-counted every 24 hours for seven consecutive days. Results are shown in figure 1 for the acetate producer & E. coli consortium (the lactate-producer & E. coli consortium shows similar dynamics). Overall, the consortia seem to converge to a common final ratio, showing robust population dynamics governed by the carbon-sharing dependency of E. coli on Synechocystis. We all know that the transition from being a common nursery-grown plant (including pre-planted boxes) to the extreme environment of a rooftop poses severe challenges. Green roof plant nurseries typically have "great" advice and recommend the installation of temporary or permanent irrigation systems. In some areas this might be helpful when operated correctly but in all cases this advice is defeating the purpose of an extensive Green Roof (water retention). In almost all cases any type of irrigation caused a spiral of failures and it is wrong to irrigate extensive Green Roofs in areas with limited water for the cause of just having a green roof (Green Washing). A mathematical model, involving diffusion of extracellular compounds and cell interactions, has been adapted, in order to assess the quorum sensing process occurring in biofilms. Quorum sensing is a diffusion dependent phenomenon; therefore, accumulation levels of extra-cellular substances can directly affect cellular development. In order to modeling quorum sensing from artificially structured microbial consortia (ASMC), accumulation of N-acyl-Lhomoserine lactone (AHL) was studied within a biofilm formed with three bacterial species, Agrobacterium tumefaciens KYC6, Escherichia coli JM109 and green fluorescent protein (GFP), producer and reporter, Escherichia coli JM105. The use of lignocellulosic biomass as a substrate in agricultural biogas plants is very popular and yields good results. However, the efficiency of anaerobic digestion, and thus biogas production, is not always satisfactory due to the slow or incomplete degradation (hydrolysis) of plant matter. In this study, azo dye adapted mixed microbial consortium was used to effectively remove colour from azo dye mixtures and to simultaneously generate bio-electricity using microbial fuel cells (MFCs). Operating temperature (20°C-50°C) and salinity (0.5% w/v-2.5% w/v) were varied during experiments. Reactor operation at 50°C improved dye decolourisation and COD removal kinetic constants by approximately two fold compared to the kinetic constants at 30°C. Decolourisation and COD removal kinetic constants remained high (0.28h-1 and 0.064h-1 respectively) at moderate salinity (1% w/v) but deteriorated approximately four fold when the salinity was raised to 2.5% (w/v). Molecular phylogenetic analysis of microbial cultures used in the study indicated that both un-acclimated and dye acclimated cultures from MFCs were predominantly comprised of Firmicutes bacteria. The aim of this study was to determine the effect of linear alkylbenzene sulfonate (LAS), anthracene and a LAS anthracene mixture on the growth of a microbial consortium isolated from polluted sediment. The microbial consortium was grown in a sterile glass bottle with mineral medium containing 1 g/L of glucose. Microbial growth inhibition produced by LAS, anthracene and combinations of LAS and anthracene was determined by viable count in nutritive agar; inhibitory concentration 50 (IC50) was calculated. The concentrations evaluated were 0.16, 0.8, 1.6, 16 and 160 mg/L of LAS or anthracene. The LAS anthracene mixtures were prepared by fixing either LAS or anthracene at 0.16 mg/L while increasing the other compound at the above concentrations. Microbial growth was sensitive to LAS at an IC50 of 8.22 and to anthracene at an IC50 of 5.2 mg/L. In this study we focus on one such catabolic glycoside hydrolase from B. ovatus. The enzyme, termed BoMan26A, is a -mannanase that takes part in the hydrolytic degradation of galactomannans. The crystal structure of BoMan26A has previously been determined to reveal a TIM-barrel like fold, but the relation between the protein structure and the mode of substrate processing has not yet been studied. Here we report residue-specific assignments for 95% of the 344 backbone amides of BoMan26A.
