Objective: assessment of somatoform disorder (SFMD) prevalence and impact in PD and DLB. Methods: SFMD were assessed by direct observation of symptoms in the year coincident (±6 months) with definite diagnosis of PD, DLB, AD, MSA, PSP, FTD and by interviews with patients, caregivers, General Practitioners (GPs), reviews of prior hospital admissions, in a cohort of 942 patients with neurodegenerative disorders. Matched groups of PD and DLB patients without vs. with SFMD were selected for comparisons and followed-up over 4 years. Results: The frequency of SFMD was higher in DLB (15 patients, 12%) and PD (29 patients, 7%) than in other neurodegenerative diseases (0-3%). SFMD consisted of conversion motor or sensory disorders, often accompanied by delusional thought content; in one patient catatonic symptoms were observed concomitantly with PD diagnosis. To clarify the role of the renal and hypothalamic noradrenergic systems in the antihypertensive actions of dietary potassium supplementation in salt-loaded spontaneously hypertensive rats (SHR), we measured systolic blood pressure and norepinephrine turnover, which was determined from the rate of decline of tissue norepinephrine concentration after the administration of alpha-methyl-p-tyrosine, in 5-week-old SHR or age-matched Wistar-Kyoto (WKY) rats eating normal-NaCl (0.66%) or high-NaCl (8%) diet with supplementation of 8% KCl. In WKY rats, neither high-sodium nor high-potassium diets had an effect on blood pressure with no change in renal or hypothalamic norepinephrine turnover. In SHR, however, salt loading accelerated the development of hypertension. We report the effects of a tyrosine (and phenylalanine)-free amino acid mixture on tyrosine levels, ex vivo catecholamine synthesis and in vivo catecholamine release in brain regions of the rat. Administration of a tyrosine-free amino acid load reduced tissue levels of tyrosine (-50% after 2 h) in all brain regions examined (frontal cortex, hippocampus, striatum). The tyrosine-free amino acid mixture also reduced DOPA accumulation: this effect was most marked in striatum (-44%) and nucleus accumbens (-34%), areas with a predominantly dopaminergic innervation. Smaller decreases (-20-24%) were detected in other areas (cortex, hippocampus and hypothalamus). The effect on DOPA accumulation was prevented by supplementing the mixture with tyrosine/phenylalanine. The tyrosine-free amino acid mixture did not alter 5-HTP accumulation in any region.
