A qualitative male-specific DNA marker (OT-24) was amplified by spPCR (single-primer polymerase chain reaction) from chinook salmon (Oncorhynchus tshawytscha) DNA along with several non-sex-linked products. The termini of the male-specific product were sequenced, and a pair of PeR primers were constructed for marker-specific PCR amplification. Dual primer PCR (dpPCR), with the marker-specific primers, amplified a product from both nudes and females. The amount of dpPCR product amplified from males was at least 100-fold greater than that from females. The quantitative difference between males and females was consistent among geographically distinct populations from western U.S. rivers. In addition, DNA sequence analysis indicated that OT-24 was highly conserved among geographically distinct salmon populations. The qualitative spPCR product segregated through several genetic crosses indicating equal sex ratios among progeny. Identification of the male and female juveniles by dpPCR was consistent with the

Sterols are required by almost all eukaryotes, but their structural diversity varies across different lineages. It is unclear if there are any systematic variations in sterol structures between metazoa and their unicellular relatives. MIT graduate student Jonathan Grabensatter analyzed sterols from four unicellular relatives of metazoa. For comparison, sterols from the basal metazoans hexactinellid sponges and ctenophores were analyzed as well. Sterols were identified by gas chromatography and mass spectrometry. Choanoflagellate sterol profiles are largely similar, containing two or three C27 compounds with 2, 3 and 4 double bonds and one C28 with three double bonds. Ichthyospora are dominated by a C27Δ5, 24 but also produce cholesterol, three C28 sterols and two 4-methyl C27 sterols. Capsaspora produce cholesterol, a tetraunsaturated C27 and two C28 sterols. Quantification of aboveground biomass (AGB) in Alaskas boreal forest is essential to the accurate evaluation of terrestrial carbon stocks and dynamics in northern high-latitude ecosystems. Our goal was to map AGB at 30 m resolution for the boreal forest in the Yukon River Basin of Alaska using Landsat data and ground measurements. We acquired Landsat images to generate a 3-year (2008-2010) composite of top-of-atmosphere reflectance for six bands as well as the brightness temperature (BT). We constructed a multiple regression model using field-observed AGB and Landsat-derived reflectance, BT, and vegetation indices. A basin-wide boreal forest AGB map at 30 m resolution was generated by applying the regression model to the Landsat composite. The fivefold cross-validation with field measurements had a mean absolute error (MAE) of 25.7 Mg ha−1 (relative MAE 47.5%) and a mean bias error (MBE) of 4.3 Mg ha−1(relative MBE 7.9%). Many of these compounds are the products of non-ribosomal peptide synthetase (NRPS) or polyketide synthase (PKS) pathways. However, current understanding of the diversification of these pathways is largely based on the chemical structure of the bioactive compounds, while the evolutionary forces driving their remarkable chemical diversity are poorly understood. We evaluated the survival of juvenile salmon through turbines in Columbia River dams and found no differences between two operations but strong evidence of delayed mortality from turbine passage. After tagging with a passive integrated transponder (PIT) tag and a radio tag, yearling Chinook salmon Oncorhynchus tshawytscha were released at McNary Dam on the Columbia River through a turbine operating both within 1% of peak efficiency (a discharge rate of 317 m(3)/s) and outside the 1% range at the maximum blade angle (464 m(3)/s). Estimated relative survival to a detection array 15 km downstream was 0.871 at 317 m(3)/s and 0.856 at 464 m(3)/s and 0.858 and 0.814, respectively, to an array 46 km downstream. The highest point estimates of survival occurred under the lower discharge, suggesting that operating turbines within 1% of peak efficiency is a useful guideline for fish protection at McNary Dam. In a concurrent evaluation using balloon tags, estimated mean direct survival ranged from 0.930 to 0.946.

Salmon are paramount to the economy, ecology, and history of the Pacific Northwest. Viruses constitute one of the major threats to salmon health and well-being, with more than twenty known virus species that infect salmon. Here, we describe the isolation and characterization of the fall Chinook aquareovirus, a divergent member of the species Aquareovirus B within the family Reoviridae. The virus was first found in 2014 as part of a routine adult broodstock screening program in which kidney and spleen tissue samples from healthy-appearing, adult fall Chinook salmon (Oncorhynchus tshawytscha) returning to a hatchery in Washington State produced cytopathic effects when inoculated onto a Chinook salmon embryo cell line (CHSE-214). The virus was not able to be confirmed by an RT-PCR assay using existing aquareovirus pan-species primers, and instead was identified by metagenomic next-generation sequencing.
