The study was conducted to assess the characterization, differentiation, and in vitro cell regeneration potential of canine mesenteric white adipose tissue-derived mesenchymal stem cells (AD-MSCs). The tissue was harvested through surgical incision and digested with collagenase to obtain a stromal vascular fraction. Mesenchymal stem cells isolated from the stromal vascular fraction were characterized through flow cytometry and reverse transcription-polymerase chain reaction. Assessment of cell viability, in vitro cell regeneration, and cell senescence were carried out through MTT assay, wound healing assay, and β-galactosidase assay, respectively. In this study, after 21 days of ceasing CCl 4 , both placebo-treated and untreated cirrhotic mice showed reduced liver damage as indicated by a decrease of AST/ALT index, recovery of histology, and reduction of fibrosis-related gene expression. Presumably, the liver of these mice were capable of self-regeneration. The cause of the self-regeneration has been explained by the ceasing of CCl 4 administration during the 21 days of treatment. The hMSC and transforming growth factor-β1 (TGFβ-1) gene infected hMSC were co-cultured with hepatoma cells. The ability of cells migration was assessed by Transwell assay. The ability of cells proliferation was detected using CCK-8 assay. Purpose: : To observe the differentiation potential of SD rat bone marrow mesenchymal stem cells (MSCs) which were subretinally transplanted into laser-injured retina in vivo without differentiation inducements in vitro. Methods: : SD rat MSCs were cultivated in vitro to passage 3 and were injected into the sub-retina space of laser-injured rat retina with Bromo-Deoxyuridine (BrdU) labeling to trace the origin of the repopulating cells. After 4 weeks, eyeballs were enucleated for continuous paraffin slides. The expressions of Nestin, NeuN, GFAP, Vimentin, Rhodopsin were detected by immunofluorescence. Results: : BrdU-positive MSCs were present in mutiple locations, including photoreceptor layer, the bipolar layers and the ganglion cell layer. Bone marrow mesenchymal stem cells (BMMSCs)-based therapy has emerged as a promising novel therapy for Traumatic Brain Injury (TBI). However, the therapeutic quantity of viable implanted BMMSCs necessary to initiate efficacy is still undetermined. Increased oxidative stress following TBI, which leads to the activation of nicotinamide adenine dinucleotide phosphate (NADPH) oxidase signaling pathway, has been implicated in accounting for the diminished graft survival and therapeutic effect. To prove this assertion, we silenced the expression of NADPH subunits (p22-phox, p47-phox, and p67-phox) and small GTPase Rac1 in BMMSCs using shRNA. Our results showed that silencing these proteins significantly reduced oxidative stress and cell death/apoptosis, and promoted implanted BMMSCs proliferation after TBI.
