An improved L-lysine amperometric biosensor based on lysine oxidase immobilised onto a platinum electrode by glutaraldehyde co-crosslinking with bovine serum albumin is described. A thoroughly optimization of the enzyme immobilization procedure permitted the fabrication of a fast-response biosensor with high sensitivity and improved stability. Moreover, the relevant electrochemical study showed the possibility to tune the overall kinetic control of biosensor from merely diffusive to enzymatic or mixed by switching the pH from weakly alkaline to weakly acid values, respectively, as well as by controlling its hydrodynamic behaviour. The response time, t(0.95), evaluated in batch addition experiments, was lower than 6 s. Novel glycine oxidase (GlyOX) from Marinomonas mediterranea depends on cysteine tryptophilquinone (CTQ) and catalyzes the oxidative deamination of glycine to produce a glyoxylate, ammonia, and hydrogen peroxide. M. mediterranea GlyOX genes (goxA and goxB) were cloned and recombinant GlyOX was heterologously expressed by E. coli. The purification of recombinant GlyOX was carried out by metal affinity and DEAE-Toyopearl 650M column chromatographies. M. mediterranea GlyOX was homotetramic with a molecular mass of 76kDa and showed optimum activity around 30°C and at pH 5.0, and stability below 50°C and between pH 5.0 to 9.0. The maximum size and age that clonal organisms can reach remains poorly known, although we do know that the largest natural clones can extend over hundreds or thousands of metres and potentially live for centuries. We made a review of findings to date, which reveal that the maximum clone age and size estimates reported in the literature are typically limited by the scale of sampling, and may grossly underestimate the maximum age and size of clonal organisms. A case study presented here shows the occurrence of clones of slow-growing marine angiosperm Posidonia oceanica at spatial scales ranging from metres to hundreds of kilometres, using microsatellites on 1544 sampling units from a total of 40 locations across the Mediterranean Sea. Temporal variability in species composition and abundance may enhance coexistence in ecological communities, especially when subjected to strong seasonal patterns. Species distribution through time may be related to many factors, such as changes in the structure and functioning of preferred habitats, variations in ecological interactions or asynchronous patterns in species life cycles. Here, data from a one-year survey of amphipod assemblages from a nearly undisturbed Posidonia oceanica meadow are used to investigate the seasonal patterns in species abundance and identity. Posidonia oceanica, an endemic seagrass of the Mediterranean Sea harbors a high diversity of N2-fixing prokaryotes. One of these is Halothece sp., a unicellular N2-fixing cyanobacteria detected through nifH analysis from the epiphytes of P. oceanica. The most related strain in culture is Halothece sp. PCC 7418 and this was used as the test organism in this study. In the Mediterranean Sea, phosphorus (P) and iron (Fe) can be the major limiting nutrients for N2 fixation. A close phylogenetic relationship appears among the species Marinomonas vaga, Oceanospirillum linum, Halomonas elongata, and Pseudomonas aeruginosa. Because of the impossibility of finding a single most closely related species, we suggest that this bacterium be assigned to a new genus, at least temporarily. The integrated nanoliter system is a measuring, separating, and mixing device that is able to measure fluids to the nanoliter, mix different fluids for a specific product, and separate a solution into simpler solutions. All features of the integrated nanoliter system are specifically designed for controlling very small volume of liquid (referred as microfluidic solutions). The integrated nanoliter systems scalability depends on what type of processing method the system is based on (refer as technology platform) with each processing method having its advantages and disadvantages. N2 - We show that graphene membranes that separate two gases at identical pressure are deflected by osmotic pressure. The osmotic pressure is a consequence of differences in gas permeation rates into a few-layer graphene enclosed cavity. The deflection of the membrane is detected by measuring the tension-induced resonance frequency with an interferometric technique. Using a calibration measurement of the relation between resonance frequency and pressure, the time dependent osmotic pressure on the graphene is extracted. The time dependent osmotic pressure for different combinations of gases shows large differences that can be accounted for by a model based on the different gas permeation rates. The class Bolidophyceae includes a single genus Bolidomonas and two species (Bolidomonas pacifica and B. mediterranea) described from marine waters.. The Bolidophyceae was established by Guillou et al. (1999), as a new class of picoplankton isolated from both the equatorial Pacific and the Mediterranean Sea. One genus, Bolidomonas, and two species, Bolidomonas pacifica and B. mediterranea, have been described. Guillou et al. also proposed a subspecies of B. pacifica: B. pacifica var. eleuthra. Bolidomonas pacifica swims differently than B. mediterranea, and their flagella are oriented differently with respect to one another (Fig. 1). The phylum Echinodermata is well known for its extensive regenerative capabilities. Although there are substantial data now available that describe the histological and cellular bases of this phenomenon, little is known about the regulatory molecules involved. Here, we use an immunochemical approach to explore the potential role played by putative members of the transforming growth factor-β (TGF-β) family of secreted proteins in the arm regeneration process of the crinoid Antedon mediterranea. We show that a TGF-β-like molecule is present in normal and regenerating arms both in a propeptide form and in a mature form. During regeneration, the expression of the mature form is increased and appears to be accompanied by the appearance of an additional isoform. Immunocytochemistry indicates that TGF-β-like molecules are normally present in the nervous tissue and are specifically localized in both neural elements and non-neural migratory cells, mainly at the level of the brachial nerve.
