In the Golgi the receptor binds newly synthesized lysosomal enzymes and transports them directly to an endosomal (prelysosomal) compartment without traversing the plasma membrane. Deletion of the carboxyl-terminal Leu-Leu-His-Val residues of the 163 amino acid cytoplasmic tail of the bovine Man-6-P/IGF-II receptor partially impaired this function, resulting in the diversion of a portion of the receptor-ligand complexes to the cell surface, where they were endocytosed. The objective of this proposal is to determine the prognostic role of expression of Insulin-Like Growth factor II in breast cancer. IGF-II is a potent mitogen for breast tumor epithelium, and is expressed in the stroma of invasive breast cancers. In this study, we analyzed expression of IGF-II mRNA and protein in two separate series of patients with invasive breast cancer, and compared the result with other clinical parameters, prognostic indicators and patient outcome. IGF-II mRNA and protein expression were easily detected in the majority of the 193 cases that were informative and had complete clinical follow up. While analysis of the cases from the two data sets individually showed that IGF-II expression was associated with clinical outcome depending on hormone receptor status. However, IGF-II expression by itself was not a prognostic indicator, and the relationship with hormone receptor status was lost when the separate data sets were analyzed together. I had a urinary infection and used the Mannose for one day thinking that was all I needed after reading info about the product. After that first day my symptoms were gone but I continued to take the Mannose the next day, but only a few teaspoons. My infection came back with a vengeance, I got a fever and it seemed that it was getting into my kidneys. I tried one antibiotic, cepro, took 2 pills and saw all the side effects and switched to another one. The next antibiodic didnt clear up the infection either although since I was taking one bottle a day of the Mannose and had no symptoms. As soon as I stopped the Mannose the symptoms returned. I went one more round with another antibiodic with no relief and stopped the Mannose so I could tell if the drugs were helping. They werent! Went to my naturopath and he recommended continuing with the mannose, the probiotics as well as his remedy which was to support the kidneys. Here, we have used a fluorescent EMSA to reveal a previously uncharacterized binding site for the endosomal Sec1/Munc18 protein, Vps45p, on the syntaxin Tlg2p. We show that a construct lacking the N-peptide of Tlg2p interacts tightly with Vps45p (Fig. 2), demonstrating the presence of a C-terminal binding site. The Vps45p-L117R mutant, which does not bind the N-peptide of Tlg2p, binds to Tlg2p(37-318) with a similar affinity as wild type. Moreover, mutation of Tlg2p residues F9 and L10 to alanine, which disrupts binding of the N-peptide to Vps45p, competes for binding to Vps45p similarly to the N-peptide deletion (Table 1). This Tlg2p-Vps45p interaction explains the lack of a trafficking defect when the Vps45p-L117R mutant is expressed in yeast as the sole copy of Vps45p (13). Kak Zeti Aktar Jaafar telah mengajak saya untuk menjadi urus setia bagi program Minggu Penulis Remaja (MPR) untuk tahun 2010. MPR 2010 juga diadakan di Port Dickson seperti tahun saya, MPR 2009 cuma lokasinya berbeza. MPR 2010 kali ini disertai oleh 21 orang peserta dari seluruh Malaysia. Paling menarik saya dapat berjumpa lagi dengan beberapa orang penulis ternama. Selain itu, salah satu pengisian MPR 2010 ialah lawatan ke Muzium Tentera di Port Dickson. Muziumnya sangatlah besar dan perlu ke tingkat atas, ke tingkat bawah serta bertukar bangunan dan paling seronok apabila melalui terowong bawah tanah. Setelah usai program, ketika di DBP, saya sempat mengajak Amir yang juga mantan MPR 2010 ke tingkat 20 DBP iaitu bahagian majalah. Waktu ketika itu sudah pun 5.30 petang, tetapi sempat juga bertemu Kak Ton, Kak Yus, Abg Armin dan Abg Ruslan. Rindu saya pada Kak Mah juga terurai apabila dapat bersemuka dengannya pada petang itu.
