The S1 gene nucleotide sequences of 10 type 3 (T3) reovirus strains were determined and compared with the T3 prototype Dearing strain in order to study sequence diversity in strains of a single reovirus serotype and to learn more about structure-function relationships of the two S1 translation products, sigma 1 and sigma 1s. Analysis of phylogenetic trees constructed from variation in the sigma 1-encoding S1 nucleotide sequences indicated that there is no pattern of S1 gene relatedness in these strains based on host species, geographic site, or date of isolation. This suggests that reovirus strains are transmitted rapidly between host species and that T3 strains with markedly different S1 sequences circulate simultaneously. The stimulation of reporter gene expression following co-transfection with the S4 gene of mammalian reoviruses was analysed. The σ3 protein of type 3 reovirus gave a five- to eightfold increase in expression of chloramphenicol acetyltransferase and β-galactosidase but this was found to be dependent upon the nature of the promoter being used to drive reporter gene expression. The σ3 protein of reovirus type 1 failed to stimulate reporter gene expression under any of the conditions used. Hybrid constructs between the S4 genes of reoviruses type 1 and 3 were used to map the stimulation characteristic to the carboxy-terminal third of the gene. Analysis of the level of σ3 protein accumulation in transfected cells showed that the reovirus type 3 protein accumulated to a much higher level than that of reovirus type 1. Using the hybrid gene constructs this higher level of protein accumulation was shown to co-segregate with the ability to stimulate reporter gene expression.

Author summary Enteric viruses are exposed to diverse environments during their replication cycle. They must be stable enough to endure outside their host, persist through changing pH and proteolytic environments encountered through passage via digestive and gastrointestinal tracts, and pliable to undergo disassembly during infection of host cells. Some enteric viruses have evolved to interact with and use members of the host microbiota to achieve optimal virion stability to infect their host. In this study, we show that the enteric mammalian reovirus (reovirus) interacts with bacteria. The interaction of the virus with bacteria or bacterial envelope components results in enhanced virion stability, which translates into enhanced reovirus infectivity following an environmental stress. The interaction of reovirus with bacterial envelope components does not alter receptor utilization, overall infection kinetics, or affect the anti-viral effects of reovirus-specific antibodies. Oncolytic viruses selectively target and replicate in cancer cells, providing us with a unique tool with which to target and kill tumour cells. These viruses come from a diverse range of viral families including reovirus type 3 Dearing (RT3D), a non-pathogenic human double-stranded RNA oncolytic virus, which has been shown to be an effective therapeutic agent, both as a mono-therapy and in combination with traditional chemotherapeutic drugs. This study investigated the interaction between RT3D and radiotherapy in melanoma cell lines with a BRAF mutant, Ras mutant or BRAF/Ras wild type genotype. The data indicates that RT3D combined with radiotherapy significantly increased cytotoxicity relative to either single agent, independent of genotype, both in vitro and in vivo.
