MicroRNAs are key regulators of plant physiological activities by functioning in RNA silencing and post-transcriptional regulation of gene expression. In apple, the uncharacterized Md-miR285N is predicted to target 35 RNA transcripts, mostly coding for TMV, SCN1 and CDPK proteins involved in plant defense to pathogens. In this work, ca. 2 Kb of the Md-miR285N promoter were isolated and cloned into an expression vector with its 3-end fused to the ß-glucoronidase (GUS) reporter gene. The expression vector was used for Agrobacterium-mediated transformation of both Malus x domestica (cv. Gala) and Arabidopsis thaliana (ecotype Col-0). In transgenic apple plantlets a strong GUS activity was detected in stems, leaves and veins. In Arabidopsis seedlings the promoter was shown to be highly expressed in roots, shoot apex meristem, primary and secondary leaves and veins. To understand the functional response of Md-miR285N against pathogen infections, apple and Arabidopsis plantlets wereinoculated, ...

McIntosh apple trees at three early stages of leaf expansion were exposed to freshly discharged ascospores of Venturia inaequalis in a dew chamber at 18 19 C. Spores deposited on glass coverslips positioned next to the leaves at each node were counted. Spores on some coverslips were also tested for viability. Lesions were noted after the trees had been kept in a growth chamber (12 hr of light at 25 C followed by 12 hr of darkness at 20 C) for 11 27 days. The efficiency of lesion production was calculated as the ratio of the number of lesions per unit of leaf surface area at the time of inoculation to the number of spores per unit of coverslip area. Lesion-forming efficiency averaged 5, 6, and 14% for the three groups of trees.. ...

Fruit growth in apple (Malus × domestica Borkh.) is mediated by cell production and expansion. Genes involved in regulating these processes and thereby fruit growth, are not well characterized. We hypothesized that the apple homolog(s) of AINTEGUMENTA (ANT), an APETALA2-repeat containing transcription factor, regulates cell production during fruit growth in apple. Two ANT genes, MdANT1 and MdANT2, were isolated from apple and their expression was studied during multiple stages of fruit development. MdANT1 and MdANT2 expression was high during early fruit growth coincident with the period of cell production, rapidly declined during exit from cell production, and remained low during the rest of fruit development. The effects of increase in carbohydrate availability during fruit growth were characterized. Increase in carbohydrate availability enhanced fruit growth largely through an increase in cell production. Expression of MdANT1 and MdANT2 increased sharply by up to around 5-fold in response to an

This study investigated the diel emission of volatile organic compounds (VOCs) from intact apple (Malus x domestica Borkh., cv. Golden Delicious) and grape (Vitis vinifera L., cv. Pinot Noir) foliage. Volatiles were monitored continuously for 48 h by proton transfer reaction - time of flight - mass spectrometry (PTRToF-MS). In addition, volatiles were collected by closed-loop-stripping-analysis (CLSA) and characterized by gas chromatography-mass spectrometry (GC-MS) after 1 h and again 24 and 48 h later. Fourteen and ten volatiles were characterized by GC-MS in apple and grape, respectively. The majority of these were terpenes, followed by green leaf volatiles, and aromatic compounds. The PTR-ToF-MS identified 10 additional compounds and established their diel emission rhythms. The most abundant volatiles displaying a diel rhythm included methanol and dimethyl sulfide in both plants, acetone in grape, and mono-, homo- and sesquiterpenes in apple. The majority of volatiles were released from both ...

We mapped DNA polymorphisms generated by 41 sets of Simple Sequence Repeat (SSR) primers, developed independently in four laboratories. All primer sets gave polymorphisms that could be located on our `White Angel x `Rome Beauty map for apple [Malus sylvestris (L.) Mill. Var. domestica (Borkh.) Mansf.]. The SSR primers were used to identify homologous linkage groups in `Wijcik McIntosh, NY 75441-58, `Golden Delicious, and `Liberty cultivars for which relatively complete linkage maps have been constructed from isozyme and Random Amplified Polymorphic DNA (RAPD) markers. In several instances, two or more SSRs were syntenic, and except for an apparent translocation involving linkage group (LG) 6, these linkages were conserved throughout the six maps. Twenty-four SSR primers were consistently polymorphic, and these are recommended as standard anchor markers for apple maps. Experiments on a pear (Pyrus communis L.) population indicated that many of the apple SSRs would be useful for mapping in ...
SUMMARY: Series of apple and pear shoots were inoculated with the scab pathogens Venturia inaequalis and V. pirina and injected with polyphenolic metabolites extracted from apple and pear leaves, and with urea. The translocation and persistence of arbutin in Cox leaves was demonstrated chromatographically. Injection of metabolites of the highly susceptible varieties Cox and Williams back into the same varieties, markedly inhibited the pathogens, which suggests that these potential resistance factors do not occur only in varieties which are scab resistant. Inhibition of infection was most marked on leaves already developing a mature resistance and the injection treatments were most effective when applied near the time of inoculation. Injection of urea increased susceptibility to V. inaequalis, and further results indicated the importance of optimal balance mechanisms between polyphenolic and nitrogenous metabolites. Chlorogenic acid and isochlorogenic acid strongly inhibited both pathogens and there was
Rarely do you see a home orchard missing the king of fruits. Apples today can be grown from the desert to the sea. Apples offer a remarkable diversity of flavors, colors and textures and have a lifespan that far exceeds many other fruit trees. The heirloom apple varieties that we offer fruit from mid July to late November, offering almost six months of superior flavors rarely found on the supermarket shelf. The home grown heirloom apple tree has historically been the central figure in the providing the nutrients and treats for past generations. Lastly, the suggested uses for each of these antique apple trees are just that-- suggestions. Experiment and have fun! Our certified organic apple trees growing in our field range from 5-8 feet in height, and are pruned to approximately 4-5 feet in height when shipped. The great majority of our apple varieties range from 5/8 to 3/4 in caliper and most have side branches (feathered), which often leads to earlier fruiting. Pick an apple for sample flavor and seed

People who have stomach ulcers or acid reflux may find that apple cider vinegar makes their conditions worse.. On the other hand, for some people with heartburn, resulting from reduced stomach acid, apple cider vinegar may improve heartburn or acid ...
Nitric oxide production, nitric oxide synthase (NOS) and mitochondrial nitrite-reducing activities in roots, leaves and stems of different developmental stages were investigated, using potted 3-year-old apple (Malus domestica Borkh.) trees. The arginine-dependent NOS activity is sensitive to NOS inhibitor L-NAME and aminoguanidine (AG), with L-NAME being more effective than AG. Endogenous NO production, NOS and mitochondrial nitrite-reducing activities are predominately presented in young leaves and especially in young white roots and young stems. Root and stem mitochondria can reduce nitrite to nitric oxide at the expense of NADH, however, this mitochondrial nitrite-reducing activity is absent in leaves ...

ISHS 2009. Acta Hort. 814:59-66. Interpretive Summary: Technical Abstract: Malus sieversii, a wild apple species native to Central Asia has been recognized as the major progenitor of the domestic apple. Collection trips to Central Asia have verified that M. sieversii is very diverse and has all the qualities present in M. ×domestica. Nearly 130,000 seeds from 950 wild M. sieversii trees were collected in eight distinct sites, mostly in Kazakhstan, on four collection trips from 1989 to 1996. The investigation of 1480 seedlings from these diverse sites is summarized. All seedlings were screened as young seedlings for resistance to Venturia inaequalis (apple scab) and planted as own-rooted seedlings in a high density orchard in Geneva, New York. Resistance was recorded in 41% of the seedlings (7%-66% among ...

The dynamic changes of secondary metabolites and tyrosinase activity of Malus pumila flowers were investigated during blooming for full use of M. pumila flowers. Phlorizin, astragalin and afzelin were selected as the indicators to determine the optimum extraction conditions of M. pumila flowers by HPLC analysis and three-factor and three-level orthogonal design experiments. When the smashing mesh number was 40 mesh, the ultrasonic time was 30 min, the solid-liquid ratio was 1:120, and the centrifugal speed was 10,000 r/min, the total content of the three compounds was the highest. The total content of phlorizin, astragalin and afzelin increased firstly and then decreased, and the highest was on the third day (176.74 mg/g). At the same time, the tyrosinase activity of M. pumila flowers showed that it had an activation effect on tyrosinase during early blooming period, however it exhibited inhibitory effect during late blooming period.

Considering the many different types of apples we see at farmers markets and supermarkets, it may be hard to believe that apple trees are not as diverse as they should be. But it isnt the fruit-bearing part of the apple tree thats the problem, its the apple trees rootstock. Most of todays commercially produced apples are from trees that were bred in two parts-the fruit-bearing scion that makes up the higher branches and tree tops, and the rootstock that forms the roots and lower trunk.

Maybe its the influence of the English language, or maybe its something idiosyncratic, but, to me, except when speaking imprecisely, a potato is not a potato plant, a blueberry is not a blueberry bush, an apple is not an apple tree, and so on. Perceived with precision (and without the conceptual sloppiness to which casual speech is prone), a potato is a tuber, not necessarily a plant; a blueberry is a berry, not necessarily a bush; an apple is a fruit, not necessarily a tree. Consider the prime definition of apple in the Merriam-Webster Collegiate Dictionary: "the fleshy usu. rounded and red or yellow edible pome fruit of a tree (genus Malus) of the rose family." In this case, even the old Romans & Greeks had separate words for the fruit (malum, mēlon) and the tree (malus, mēléa). A reasonable way of sorting these concepts is to have an article Malum for the fruit and an article Malus domestica for the tree (not to mention Malus (genus) for the genus). ¶ The easiest principle to invoke is ...

Theres good news today for parents who give their kids apple juice: The U.S. Food and Drug Administration has announced new draft guidelines that call for no more than 10 parts per billion of inorganic arsenic in apple juice - roughly the same level allowed in tap water.. Although some apple juices have tested high for arsenic, all 95 samples tested by the FDA already fell within the 10 ppb - reassurance that when it comes to arsenic levels, apple juice is safe.. "The most important message is we are confident in the safety of our juice supply," said FDA Commissioner Dr. Margaret Hamburg.. Pressure to develop the guidelines stemmed from a 2011 report on the Dr. Oz Show and arsenic testing by Consumer Reports, both of which raised questions about the safety of apple juice.. Arsenic and Apple Juice: New Testing Finds Low Levels In response, the FDA undertook additional testing and commissioned a risk assessment study to determine an arsenic limit based on the amount of juice a child might drink ...

Shown here is a section through the apple from skin, through a layer of compact epidermal cells, to the first layer of the interior cortex parenchyma (storage) cells. Dense epidermal tissue is developed in external layers; its cells have thick walls and the outer surface is covered with waxy cuticle. Compared to many other fruits and vegetables, apples contain relatively low amounts of vitamin C, as well as a several other antioxidant compounds. They may also help with heart disease, weight loss and controlling cholesterol, as they do not have any cholesterol, have fibre, which reduces cholesterol by preventing reabsorption, and are bulky for their caloric content like most fruits and vegetables. your apple trees can be sprayed with Bordeaux (lime-sulfur). This fungicide must be applied early and thoroughly to protect new growth. Apply according to label directs; as the buds break until the leaves are fully expanded. Do this at prepink, pink, calyx, and first cover. Since the fungus overwinters in fallen leaves and fruit, be sure to thoroughly clean up and dispose of plant debris in the fall months. The brown trails through the fruit indicate apple maggot presence. There has been some success with with phermone (sex attractant) traps to catch the adult fly. You can also make sticky-traps to hang in your tree; use red balls or old plastic Christmas tree ornaments coated with petroleum jelly to trap the females. (They lay their eggs oi developing apple fruits and the eggs hatch into little worms burrow their way through the fruit. The brown trails are the tunnels where theyve been.) Again, be sure to dispose of, or bury any fruit that has fallen on the ground. Because your apple tree ...
