We have investigated whether the recently discovered transcription element, zinc little finger BED domain-containing proteins 6 (ZBED6), is expressed in insulin-producing cells and, if thus, to what degree it affects beta cell function. function of insulin-producing beta cells. For example, genetics code for transcription elements important to the pancreatic beta cellsuch as Neurog3, Nkx6.1, NeuroD2, and v-maf musculoaponeurotic fibrosarcoma oncogene homolog A (MafA)were found to be putative joining focuses on for ZBED6 in myoblast cells (1). ZBED6-mediated control of may become relevant to beta cells because it offers been noticed that extravagant IGF2 creation in embryonic pancreas forwent the following beta cell mass anomaly that evolves in diabetic GotoCKakizaki rodents (3). Furthermore, the human being gene is usually clustered carefully collectively with (4), directing to the probability that not really just might become managed by a common ZBED6-reliant regulatory system. Therefore, it is ...

Region 3 (base pairs -8118 to -7750 relative to the transcription start site), one of six conserved 5′ cis domains of the MafA promoter, is capable of directing β-cell-line-selective expression. Transgenic reporters of region 3 alone (R3), sequences spanning regions 1 to 6 (R1-6; base pairs -10428 to +230), and R1-6 lacking R3 (R1-6 ΔR3) were generated. Only the R1-6 transgene was active in MafA+ insulin+ cells during ...
Diabetes results from a loss in the capacity of β cells to produce enough insulin to meet the bodys needs, either as a result of autoimmune destruction of β cells (i.e., in type I diabetes mellitus [35]) or of reduced function (i.e., in type II [72]). Here, we have isolated and shown that the MafA transcription factor represents a β-cell-enriched DNA-binding component of the C1/RIPE3b1 activator complex that is involved in cell type-specific and glucose-regulated expression of the insulin gene. MafA is a member of the large Maf family of basic leucine-zipper containing DNA-binding proteins. In addition to MafA, there are three other closely related members of this family-c-Maf, MafB, and NRL-each of which have very similar DNA-binding (basic leucine zipper) and N-terminal transactivation domains (5, 73).. Phosphorylation of conserved serine and threonine residues in the large Maf proteins is important in activation domain function (4). In addition, phosphorylation of a tyrosine(s) appears to ...
A glucose sensitive regulator of insulin transcription includes a glucose response element (GIRE) of a liver-pyruvate (L-PK) gent promoter, and an insulin-sensitive element of an insulin-like growth factor binding protein-1 (IGFBP-1) basal promoter. The transcriptional activity of the regulator is stimulated by glucose and inhibited by insulin.
Results - Mafa, Ins2 and Aire expression was detected in the thymus. Mafa expression was lower in NOD thymus than control, and was correlated with Ins2 expression. Targeted disruption of MafA reduced thymic Ins2 expression, and induced autoantibodies against pancreatic islets. Functional polymorphisms of MafA were newly identified in NOD mice and humans, and polymorphisms of human MAFA were associated with susceptibility to type 1 diabetes, but not to autoimmune thyroid disease. ...

Transcription factor MafK is a bZip Maf transcription factor protein that in humans is encoded by the MAFK gene. MafK is one of the small Maf proteins, which are basic region and basic leucine zipper (bZIP)-type transcription factors. The HUGO Gene Nomenclature Committee-approved gene name of MAFK is v-maf avian musculoaponeurotic fibrosarcoma oncogene homolog K. MafK was first cloned and identified in chicken in 1993 as a member of the small Maf (sMaf) genes. MafK was also identified as p18 NF-E2, a component of NF-E2 complex binding to a specific motif (NF-E2) in the regulatory regions of β-globin and other erythroid-related genes. MAFK has been identified in many vertebrates, including humans. There are three functionally redundant sMaf proteins in vertebrates, MafF, MafG, and MafK. MafK has a bZIP structure that consists of a basic region for DNA binding and a leucine zipper structure for dimer formation. Similar to other sMafs, MafK lacks any canonical transcriptional activation domains. ...
The β-cell-enriched MAFA transcription factor plays a central role in regulating glucose-stimulated insulin secretion while also demonstrating oncogenic transformation potential in vitro. No disease-causing MAFA variants have been previously described. We investigated a large pedigree with autosomal dominant inheritance of diabetes mellitus or insulinomatosis, an adult-onset condition of recurrent hyperinsulinemic hypoglycemia caused by multiple insulin-secreting neuroendocrine tumors of the pancreas. Using exome sequencing, we identified a missense MAFA mutation (p.Ser64Phe, c.191C,T) segregating with both phenotypes of insulinomatosis and diabetes. This mutation was also found in a second unrelated family with the same clinical phenotype, while no germline or somatic MAFA mutations were identified in nine patients with sporadic insulinomatosis. In the two families, insulinomatosis presented more frequently in females (eight females/two males) and diabetes more often in males (12 males/four ...

c-Maf, which is one of the large Maf transcription factors, can bind to Maf recognition element (MARE) and activates transcription of target genes. Although c-Maf is expressed in macrophages and directly regulates the expression of interleukin-10, detailed information regarding its function in the null mutant phenotype of tissue macrophages remain unknown. In this study, we demonstrated that c-Maf is specifically expressed in the F4/80 positive fetal liver and adult macrophages. The expression of F4/80, which is a tissue macrophage-specific seven trans-membrane receptor, was dramatically suppressed in the c-Maf-deficient macrophage, whereas the expression of Mac-1 was not affected, suggesting that c-Maf is not necessary for the lineage commitment of macrophages. Luciferase reporter and EMSA showed that c-Maf directly regulates the expression of F4/80 by interacting with the half-MARE site of the F4/80 promoter. These results suggest that c-Maf is required for the F4/80 expression in macrophages in vivo.

Analysis of MafB(-/-) mice has suggested that the MAFB transcription factor was essential to islet α- and β-cell formation during development, although the postnatal physiological impact could not be studied here because these mutants died due to problems in neural development. Pancreas-wide mutant mice were generated to compare the postnatal significance of MafB (MafB(Δpanc)) and MafA/B (MafAB(Δpanc)) with deficiencies associated with the related β-cell-enriched MafA mutant (MafA(Δpanc)). Insulin(+) cell production and β-cell activity were merely delayed in MafB(Δpanc) islets until MafA was comprehensively expressed in this cell population. We propose that MafA compensates for the absence of MafB in MafB(Δpanc) mice, which is supported by the death of MafAB(Δpanc) mice soon after birth from hyperglycemia. However, glucose-induced glucagon secretion was compromised in adult MafB(Δpanc) islet α-cells. Based upon these results, we conclude that MafB is only essential to islet α-cell ...
Glucose is an essential regulator of insulin gene expression from β-cells, and it is likely that glucose is required during any physiological scenario when increased insulin production is needed (2,11,29-31). In the present study, we identify signaling mechanisms by which GLP-1 synergistically enhances insulin gene transcription in the presence of glucose. Glucose and GLP-1 have similar effects on β-cells, arising from distinct and overlapping signaling pathways. For example, glucose elevates intracellular calcium, but also provides factors to enhance insulin gene transcription by pathways distinct of calcium (10,32,33). GLP-1 increases cAMP and activates PKA, factors both involved in calcium-dependent and -independent pathways in pancreatic β-cells (34-36). Thus, both glucose factors and GLP-1 factors exert multiple effects on the β-cell, and the points of convergence that synergistically upregulate insulin gene transcription are not well understood.. The obvious common effect produced by ...
My daughter is halfway to her 16th birthday. When she was younger, I wanted to do everything for her. I treated her like she was made of glass. I would give the other kids a hard time on the playground if they werent nice to her. Ill never forget when she was in second or third grade she told me, Im not going to call you mommy anymore, Im going to call you mom. It made me a little sad, but I got over it. I was so tied into her that when she went to preschool at age 4, I would stare at the clock, waiting for her to come home from school. My world at the time revolved around taking care of her. What I wouldnt give now for a day with 25 hours in it! As shes gotten older, Ive had to learn to distance myself somewhat. Its a slippery slope, you want to help, but how much is too much? When she had to do a project making a lighthouse in elementary school, we went to the hobby store and bought some supplies. When I went to the school to see what all the other kids made, I was astounded by the ...
Title: Maf transcription factors in the development of somatosensory neurons By Patrick Carroll, INSERM U-583, F34091 Montpellier, France.
MIT researchers have created a computer model that can predict how glucose-responsive insulin (GRI) will affect patients blood sugar based on chemical traits such as how quickly it becomes activated in the presence of glucose. This could help scientists design insulin that lingers in a patients bloodstream and becomes active only when needed, such as right after a meal.

The major findings of this study are that BMK1 is a novel regulator of HIF1α and angiogenesis by increasing HIF1α ubiquitination and degradation in EC. BMK1 was recently shown to play a critical role in EC function because the conditional BMK1 knockout mouse rapidly dies because of disruption of vascular integrity.16 We recently showed an essential role for BMK1 in preventing EC apoptosis induced by serum deprivation and tumor necrosis factor-α.17 The present study further supports the key role of BMK1 in EC function. In particular, our data show a specific role for BMK1 in regulating HIF1α protein expression and subsequently VEGF expression during hypoxia. An interesting finding is that the effect of BMK1 to promote HIF1α degradation was independent of hydroxylation on Pro 402 and Pro 564, because the HIF1α mutant (P402A/P564G) lacking the consensus prolines was still ubiquitinated and degraded after BMK1 activation.. The mechanisms that regulate HIF1α function have been extensively ...

This epidemiological study investigated the association between β-cell function (a predictive factor of diabetes development) and the organic arsenic arsenobetaine (an agent less toxic to humans). Urine arsenobetaine was revealed to be associated with HOMA-β in the normal population (without diabetes), especially in males, even after adjusting for factors affecting diabetes and β-cell function, while no association was shown with HOMA-IR which is associated with insulin resistance.. Despite several conflicting results, arsenic has been reported to be correlated with diabetes in many epidemiological studies [6, 9, 23-31], and arsenic can affect insulin resistance and β-cell function [7]. In some experimental studies, arsenite was found to induce oxidative stress in pancreatic β-cells and reduce insulin transcription and secretion [32, 33], caused by the altered expression of essential genes such as the Pdx1 gene [34, 35] and insulin gene [36] for pancreas development, insulin production, and ...
BACKGROUND: Reconstruction of large defects in the temporal region can be performed with skin grafts or pedicled or free flaps. Results are often not optimal because of the patch of a skin graft, lack of availability of local flaps, and distant skin from free flaps. A technique for reconstruction of these defects with local tissue is presented in this article that uses superficial musculoaponeurotic system (SMAS) plication to allow wide advancement of a cervicofacial flap. METHODS: Once the defect is outlined, a face-lift-like skin incision is used to raise the flap. The SMAS is plicated with two purse-string sutures that relieve tension on the flap and allow maximal advancement. Thirteen face-lift SMAS plication flaps were used in 12 patients (mean age, 70.2 years) after cancer resection, which was bilateral in one case. Defects up to 8 cm in largest diameter can be closed. In one case of an 8 × 6-cm defect, a 1 × 1.5-cm skin graft was necessary. RESULTS: All flaps healed uneventfully, and no ...

The results from this study show that control of a key developmental regulator through an optimized tetracycline-regulation system can be used to manipulate the formation of a compound gland during fetal organogenesis and the maintenance of glandular function in mature animals. We incorporated several modifications to help ensure that regulated expression of PDX1 from a transgene could compensate for the absence of both endogenous Pdx1 alleles. The correct timing of activation of the Pdx1 transgene was optimized by inserting the coding region of the tTA transcriptional activator into the Pdx1 locus to place tTA under control of the endogenous Pdx1 promoter. Sufficient tTA was produced to activate the Pdx1 transgene to an effective level. By screening multiple lines bearing the tetracycline-responsive Pdx1 transgene, we were able to identify several that could restore fetal pancreatic development in the absence of a functional endogenous Pdx1 allele.. The rescue of pancreatic development by the ...
