A macroglobulin is used to improve the signal-to-background ratio in an affinity binding assay employing a proteinase (or precursor) as a label. The macroglobulin entraps unbound labeled reagent, thereby reducing its signal generating activity, relative to that of the affinity complex.

TY - JOUR. T1 - A Waldenstrom macroglobulin that is both a cold agglutinin and a cryoglobulin because it binds N-acetylneuraminosyl residues. AU - Tsai, C. M.. AU - Zopf, D. A.. AU - Yu, Robert K. PY - 1977/1/1. Y1 - 1977/1/1. N2 - A purified human monoclonal IgM(κ) (cold agglutinin MKV) has been characterized as both a cold agglutinin and a cryoglobulin. Since its reactivity with human erythrocytes but not with dog erythrocytes is reduced by treatment of the cells with ficin and its reactivity with both is abolished by treatment of the cells with neuraminidase, it has by definition Pr2 specificity [Roelcke, D. (1974) Clin. Immunol. Immunopath. 2, 266-280]. Presumably, the membrane receptors for cold agglutinin MKV are sialic acid-containing glycoproteins in human cells and sialic acid-containing glycolipids in dog cells. Agglutination of erythrocytes is specifically inhibited by II3-N-acetylneuraminosyllactosylceramide (GM3) and N-acetylneuraminosylparagloboside but not by their ...

Serum protein electrophoresis was performed in 63% of 1,069 consecutive patients with lymphoma. No monoclonal peaks of IgM specificity were found in sera from 345 patients with nodular lymphoma or Hodgkins disease. Of the remaining 333 patients with diffuse lymphoma, 1.5% had an IgG peak, suggesting a coincidental relationship of IgG peaks to lymphoma. IgM peaks occurred in 3.6% of patients with diffuse lymphoma, a prevalence about 60 times more frequent than that in normal subjects. Such peaks were more frequent in older patients, suggesting an increased incidence of lymphomas producing monoclonal macroglobulins with advancing age. There was a close correlation between lymphoma mass and the level of the IgM peak in individual patients. Results supported the value of identifying monoclonal components in all patients with lymphoma, particularly those with diffuse lymphocytic infiltration. ...
1. Hemoglobins and serum proteins of herring have been analyzed by agar gel electrophoresis (hemoglobins) and combined starch and agar gel electrophoresis (serum proteins). The material comprises twelve samples from the Norwegian coast and the North Sea and one sample from the east coast of Canada, a total of 1,546 specimens. 2. The results of the hemoglobin analyses were in general agreement with the ontogenetic variation described by WILKINS and ILES (1966). 3. Intraspecific variation was noted in the transferrin components. Three common transferrin types were found and assumed to be controlled by two allelomorphic genes. No relation to age or length was found. Presence of additional weak components complicated the type determination. Hemoglobins in the sera as well as prolonged storage made the type determination unreliable. 4. Observed albumin variations could be explained assuming control by two alleles, while observed variations in other serum protein groups were too weak or too diffuse ...

Ceruloplasmin (or caeruloplasmin) is a ferroxidase enzyme that in humans is encoded by the CP gene. Ceruloplasmin is the major copper-carrying protein in the blood, and in addition plays a role in iron metabolism. It was first described in 1948. Another protein, hephaestin, is noted for its homology to ceruloplasmin, and also participates in iron and probably copper metabolism. Ceruloplasmin is an enzyme (EC 1.16.3.1) synthesized in the liver containing 6 atoms of copper in its structure. Ceruloplasmin carries more than 95% of the total copper in healthy human plasma. The rest is accounted for by macroglobulins. Ceruloplasmin exhibits a copper-dependent oxidase activity, which is associated with possible oxidation of Fe2+ (ferrous iron) into Fe3+ (ferric iron), therefore assisting in its transport in the plasma in association with transferrin, which can carry iron only in the ferric state. The molecular weight of human ceruloplasmin is reported to be 151kDa. A cis-regulatory element called the ...
Fibropapillomatosis (FP) of green turtles has a global distribution and causes debilitating tumours of the skin and internal organs in several species of marine turtles. FP is associated with a presently non-cultivable alphaherpesvirus Chelonid fibropapilloma-associated herpesvirus (CFPHV). Our aims were to employ quantitative PCR targeted to pol DNA of CFPHV to determine (i) if DNA sequesters by tumour size and/or cell type, (ii) whether subculturing of cells is a viable strategy for isolating CFPHV and (iii) whether CFPHV can be induced to a lytic growth cycle in vitro using chemical modulators of replication (CMRs), temperature variation or co-cultivation. Additional objectives included determining whether non-tumour and tumour cells behave differently in vitro and confirming the phenotype of cultured cells using cell-type-specific antigens. CFPHV pol DNA was preferentially concentrated in dermal fibroblasts of skin tumours and the amount of viral DNA per cell was independent of tumour size. Copy

To understand the role of the immune system with respect to disease in reptiles, there is the need to develop tools to assess the hosts immune response. An important tool is the development of molecular markers to identify immune cells, and these are limited for reptiles. We developed a technique for the cryopreservation of peripheral blood mononuclear cells and showed that a commercially available anti-CD3 epsilon chain antibody detects a subpopulation of CD3 positive peripheral blood lymphocytes in the marine turtle Chelonia mydas. In the thymus and in skin inoculated with phytohemagglutinin, the same antibody showed the classical staining pattern observed in mammals and birds. For Western blot, the anti-CD3 antibodies identified a 17.6 kDa band in membrane proteins of peripheral blood mononuclear cell compatible in weight to previously described CD3 molecules. This is the first demostration of CD3+ cells in reptiles using specific antibodies....

This snake venom is then injected in small amounts into mammals such as horses, sheep, or rabbits. These animals have an immune response whereby antibodies against the venom are generated naturally. The Antivenom is then harvested from the blood of the animal, purified and stored to treat future envenomation for snake bite victims. Antivenins are normally administered to the victim as soon as possible following a snake bite attack - usually with 4-5 hours. Since the advent of Antivenom, many snake bites which were almost always fatal have become only fatal rarely if it can administered to the victim in a timely manner. Even though the antivenins are purified by multiple processes, it may contain other serum proteins and some individuals may have an extreme hypersensitive reaction to the injection and is only administered exercising caution." ...
Egg defence against bacterial contamination relies on immunoglobulins (IgY) concentrated in the yolk and antimicrobial peptides/proteins predominantly localized in the egg white (EW). Hens contaminated with pathogenic microorganisms export specific IgYs to the egg (adaptative immunity). No evidence of such regulation has been reported for the antimicrobial peptides/proteins (innate immunity) which are preventively secreted by the hen oviduct and are active against a large range of microbes. We investigated whether the egg innate defences can be stimulated by the environmental microbial contamination by comparing the antimicrobial activity of EW of hens raised in three extreme breeding conditions: Germ-free (GF), Specific Pathogen Free (SPF) and Conventional (C) hens. The difference in the immunological status of GF, SPF and C hens was confirmed by the high stimulation of IL-1β, IL-8 and TLR4 genes in the intestine of C and SPF groups. EW from C and SPF groups demonstrated higher inhibitory effect

Recently, there have been substantial advances in techniques to detect, measure and characterize monoclonal gammopathies. For example, capillary zone electrophoresis can crisply separate serum components, detecting subtle monoclonal proteins. In addition, immunosubtraction (immunotyping) can aid in measuring M-proteins that co-migrate with other serum proteins. However, there interferences can result from a variety of substances including antibiotics and radiocontrast dyes. By use of real time audience response systems a variety of challenging and instructive cases will demonstrate the problems of false positive and false negative tests. The information will be synthesized into a practical approach to effective and efficient detection of M-proteins.. ...
Application of the DLW method in the present study gave MR estimates that were greater than respirometry by 8% and 4% for the two-sample and multiple-sample approaches, respectively, when using Eqn 7 with fed turtles. The difference between DLW and respirometry MR estimates in both cases was not significant (Table 1). The two-sample technique is typically the only approach available to field researchers as recapturing the study animals multiple times can be difficult and disrupts the animals natural behavior, but our data suggest that the two-sample technique results in twice the error of the multiple-sample approach in marine turtles. Overestimation by ≤8% for energy expenditure by the DLW method, when compared with respirometry, is common when group means are used (Speakman, 1997; Butler et al., 2004); however, the percentage difference between DLW and respirometry MR estimates for individuals ranged from -51 to 82%. As stated by Speakman (Speakman, 1997), the DLW technique is extremely ...

Rat alpha 1-macroglobulin (alpha 1M), rat alpha 2-macroglobulin (alpha 2M) migrated as single bands on non-denaturing gels when purified by the methods described. All three proteins demonstrated increased mobility after reaction with trypsin. A single saturable pathway rapidly cleared complexes of trypsin and the alpha-macroglobulins of mouse, rat and human from the circulation of mice. None of the native alpha-macroglobulins competed for clearance with the trypsin complexes. [14C]Methylamine incorporation was 4.1, 3.9, 2.6 and 3.2 mol/mol of proteinase inhibitor for human alpha 2M, rat alpha 1M, rat alpha 2M and mouse alpha 2M, respectively. Only rat alpha 2M, the acute-phase alpha-macroglobulin studied, showed no evidence of conformational change when subjected to electrophoresis after reaction with methylamine. The clearance of rat alpha 2M-methylamine was comparable with that of the native molecule. The other alpha-macroglobulin-methylamine complexes cleared faster than the inhibitors that ...
When I received the package, I was astounded to see how huge it was. If you calculate the quantity versus the price, this one is worth it and could last you for months depending how often you use it. For me, I have always used it every night prior to my moisturizer. The formula is more on the watery side. Not slimy at all compared to my other serums. It is good, if you like it to dry faster which it does. Or sometimes, you cant help but wonder about the water content. Upon application, it does nothing, no tingling or whatsoever, so I think this is more on the hydration side and less on abrasive chemicals. It is refreshing for me so any issue with the water content or ingredients, I have not complaints whatsoever. It is what it is and Im just thankful I have enough to last me for awhile. ...

The method used in the current study allowed measurement of swimming effort and V̇O2 within 2 min of hatchlings being placed in the water. I found that the greatest swimming effort and oxygen consumption occurred within the first 10 min of hatchlings entering the water, and that swimming effort and oxygen consumption decreased rapidly during the first 2 h of swimming (Fig. 2). V̇O2 directly followed the decline in swimming effort during the first 12 h of swimming, an observation consistent with the assumption that hatchling sea turtle swimming is powered predominantly by aerobic metabolism (Butler et al., 1984; Wyneken, 1997). As a consequence, as hypothesised, there was a strong correlation between swimming effort and V̇O2 (Table 1; Fig. 3). Only a few studies have measured V̇O2 in hatchling sea turtles while swimming [leatherback Dermochelys coriacea (Lutcavage and Lutz, 1986; Wyneken, 1991; Wyneken, 1997; Jones et al., 2002; Jones et al., 2007); olive ridley Lepidochelys olivacea (Jones ...

The binding of 125I-labelled transforming growth factor-beta 1 (TGF-beta 1) to human alpha 2-macroglobulin (alpha 2M) was studied by native PAGE and autoradiography. TGF-beta 1 bound preferentially to alpha 2M-methylamine and minimally, if at all, to native alpha 2M. Preparations of alpha 2M-proteinase complex were generated by incubating a standard concentration of alpha 2M (0.4 microM) with different concentrations of trypsin, chymotrypsin or neutrophil elastase (0.04-2.0 microM). The 125I-TGF-beta 1-binding activity depended on the initial ratio of active proteinase to alpha 2M, or r value, used to form the alpha 2M-proteinase complex. With all three proteinases, r values of 2 or greater yielded preparations with unchanged or decreased TGF-beta 1-binding activity relative to native alpha 2M. By contrast, r values near 1 yielded preparations with significantly increased TGF-beta 1-binding activity. The results of [3H]thymidine-incorporation studies performed in mouse keratinocytes were ...

The inhibition obtained by the number of molecules in 1 µg rCCP1-CCP2-SP per ml was. thus said to be equivalent to the number of molecules in 1·76 (79 247/45 073) µg MASP-1 per ml. We added the rCCP1-CCP2-SP to 10% fetal calf serum before performing the dilutions in order to obtain a similar matrix and to obtain comparable slopes of the dilution curve of the standard plasma and the recombinant material (the antibodies employed do not cross-react with bovine MASP-1). To test for the specificity of the assay, purified rMAp44 or rMASP-3 (produced and purified as described in Degn et al.
