We demonstrate that the signaling potential of LIGHT is affected by two polymorphisms at positions E214K and S32L. The 214 position in LIGHT directly influenced the binding avidity to a neutralizing human Ab and the LTβR, indicating this residue is located in or near the receptor-binding region. This result provides an important consideration for the use of inhibitors of LIGHT, such as decoy receptors or Abs, in analyzing patient responses in clinical trials. The 32L variant located in the intracellular domain enhanced the avidity of binding to LTβR, lowered the avidity of binding to DcR3, and decreased the membrane expression of LIGHT. Although the precise mechanism of how LIGHT32L variant affects binding avidity to LTβR and DcR3 remains to be defined, it is likely an indirect effect, perhaps related to the stability of LIGHT trimer. Increased bioavailability of LIGHT due to lower avidity for DcR3 and enhanced binding avidity of LIGHT for LTβR could combine to increase signaling activity ...

The genus Maackia is composed of 8 to 10 species all of which come from East Asia. Two of which are found in our collection. This genus is closely related to the genus Cladrastis and was once placed in there, but it differs in several important morphological characters and in its DNA. The genus is named after the Russian naturalist Richard Maack who died in 1886. We have 8 examples of Maqackia fauriei in the Arboretum. Three are from seed collected in the wild on Mt. Halla during the Nordic Arboretum Committees expedition to Korea in 1976. The rest are from seed collected from this original material. Second generation seedlings have been sent to a number of other places including: Norwegian National Arboretum, Swedens Agricultural University, Alnarp, Arboretum Trompenborg in Holland; Århus Kommune (probably the Botanic Garden, Århus). Maackia fauriei was made know to western science by Abbé Faurie in 1907, and was first introduced into European cultivation in 1917 by E. H. Wlson. It is not ...

This study assessed the presence of sialic acid α-2,3 and α-2,6 linked glycan receptors in seven avian species. The respiratory and intestinal tracts of the chicken, common quail, red-legged partridge, turkey, golden pheasant, ostrich, and mallard were tested by means of lectin histochemistry, using the lectins Maackia amurensis agglutinin II and Sambucus nigra agglutinin, which show affinity for α-2,3 and α-2,6 receptors, respectively. Additionally, the pattern of virus attachment (PVA) was evaluated with virus histochemistry, using an avian-origin H4N5 virus and a human-origin seasonal H1N1 virus. There was a great variation of receptor distribution among the tissues and avian species studied. Both α-2,3 and α-2,6 receptors were present in the respiratory and intestinal tracts of the chicken, common quail, red-legged partridge, turkey, and golden pheasant. In ostriches, the expression of the receptor was basically restricted to α-2,3 in both the respiratory and intestinal tracts and in mallards
Abstract: : Purpose: The Guinea pig conjunctival epithelium includes a cell that shares a striking morphological resemblance to the M cells (membranous cells) found over lymphoid follicles in other mucosa. The presence of M cells in the conjunctiva would be significant since intestinal M cells are known to initiate the mucosal immune response by sampling luminal antigens and translocating them to underlying lymphocytes and antigen-presenting cells. Since intestinal M cells are also the site of entry for opportunistic pathogens such as viruses or bacteria, conjunctival M cells would also represent a previously unrecognized site of pathogen entry. We hypothesized that if the conjunctiva contains M cells, it should be possible to selectively label their apical surface glycoconjugates with lectins as has been done with M cells in other mucosal locations. Methods: We screened a panel of lectins on aldehyde-fixed nodules and identified Maakia amurensis leukoagglutinin (MAL-I) as a potential ...

Herein describes a study testing the feasibility of enhanced specificity targeting tumor cells using heterobivalent ligands. Previous work has demonstrated that two functionally unrelated GPCRs, i.e., MC4R and CCK2R, or MC4R and the δ-opioid receptor (δ-OR), can be noncovalently cross-linked by their corresponding heterobivalent ligands with high avidity (12, 17, 19). Multivalency can increase binding avidity, but not necessarily increase targeting specificity (32, 33). By following the previous in vitro findings, the current work studied the parameters including linker length and flexibility, surface receptor expression level per cell, ratio of receptor expression level, receptor-ligand affinity, and ligand concentration on which this targeting specificity depends.. The linker, Pego-[PG]3-Pego, a combination of rigid and flexible linker, demonstrated dynamic mobility over time and temperature ranges and spanned distances of 30∼70 Å as predicted by computer models (Fig. 1 B-D). As evidenced ...

SAα2,6 and SAα2,3 linked sialic acid molecules on epithelial cell membrane served as receptors for influenza virus, which are specifically recognized by human and avian influenza viruses, respectively. The distribution of these two species of sialic acids in human respiratory tract from different anatomical sites and different age groups was investigated. The results showed that SAα2,3Gal species was prevalent in respiratory bronchiole and lung alveolar epithelium, but was infrequent in trachea, bronchus and bronchiole. On the contrary, the SAα2,6Gal species was more common in the trachea and bronchus and to a lesser degree in the alveolar epithelium. When compared the expression levels of SAα2,6Gal and α2,3Gal in the respiratory tract among different age groups, no significant difference was found. In the ex vivo H5N1 virus infection study, alveolus epithelium were found to be more susceptible to avian influenza than trachea and bronchus epithelial cells. These results suggest that the ...

Results: Leukocytospermic seminal fibronectin, in comparison with fibronectin of normal fertile group, showed lower relative reactivity with AAL, LTA and UEA, and higher reactivity with MAA and SNA, while the AGP of the leukocytospermic group was less reactive with AAL, and the relative reactivity. buy 5-Fluoracil with LTA and MAA was ...
In the influenza virus, the two relevant antigens are the surface proteins, hemagglutinin and neuraminidase.[4] The hemagglutinin is responsible for binding and entry into host epithelial cells while the neuraminidase is involved in the process of new virions budding out of host cells.[5] Sites recognized on the hemagglutinin and neuraminidase proteins by host immune systems are under constant selective pressure. Antigenic drift allows for evasion of these host immune systems by small mutations in the hemagglutinin and neuraminidase genes that make the protein unrecognizable to pre-existing host immunity.[6] Antigenic drift is this continuous process of genetic and antigenic change among flu strains.[7]. In human populations, immune (vaccinated) individuals exert selective pressure for single point mutations in the hemagglutinin gene that increase receptor binding avidity, while naive individuals exert selective pressure for single point mutations that decrease receptor binding avidity.[6] These ...

We sequenced and characterized the complete mitogenome (KX964606) of the Amur hedgehog Erinaceus amurensis to provide more data for comparative mitogenomics of the genus Erinaceus (Erinaceidae). The mitogenome of E. amurensis is a circular molecule 16,941 bp long, consisting of a control region and a conserved set of 37 genes containing 13 protein-coding genes, 22 tRNA genes, and two rRNA genes (12S rRNA and 16S rRNA). The mitogenome of E. amurensis is AT-biased, with a nucleotide composition of 33.9% A, 21.1% C, 32.6% T, and 12.4% G. The mitogenomes of E. amurensis and the closely related hedgehog species E. europaeus, excluding the control region (66.7%), share over 90% sequence similarity. According to the inter-generic relationship based on six mitogenomes described from five genera of Erinaceidae, the subfamilies Erinaceinae and Galericinae are strongly supported as monophyletic groups, with each genus well placed within its own subfamily. Within the subfamily Erinaceinae, E. amurensis is a sister

Using an adaptive study design, the PET study was carried out in parallel with the single ascending dose (SAD) study to relate occupancy at the A2A receptor in the brain with drug dose and plasma concentration. For each drug dose, A2A receptor occupancy was calculated for 2 plasma drug levels.. The data generated clearly demonstrated the relationship between plasma concentration and receptor occupancy in the CNS showing that the drug crosses the blood brain barrier and interacts with the target receptor. Using this data, Vernalis was able to take the drug into further development. Importantly, the quantitative data generated from imaging was used to determine the dose to be used in patient studies for efficacy, thus reducing the number of doses to be tested, saving considerable time and money.. This study clearly demonstrates the value imaging can add to aid decisions about drug development. In this way, imaging holds the key to reducing time and costs of drug development overall. ...
P-glycoprotein (P-gp) overexpression is the most frequently observed cause of multidrug resistance in neoplastic cells. In our experiments, P-gp was expressed in L1210 mice leukemia cells (S cells) by selection with vincristine (R cells) or transfection with the gene encoding human P-gp (T cells). Remodeling of cell surface sugars is associated with P-gp expression in L1210 cells as a secondary cellular response. In this study, we monitored the alteration of cell surface saccharides by Sambucus nigra agglutinin (SNA), wheat germ agglutinin (WGA) and Maackia amurensis agglutinin (MAA). Sialic acid is predominantly linked to the surface of S, R and T cells via α-2,6 branched sugars that tightly bind SNA. The presence of sialic acid linked to the cell surface via α-2,3 branched sugars was negligible, and the binding of MAA (recognizing this branch) was much less pronounced than SNA. WGA induced greater cell death than SNA, which was bound to the cell surface and agglutinated all three L1210 cell-variants
Attachment of human noroviruses to histo blood group antigens (HBGAs) is essential for infection, but how this binding event promotes the infection of host cells is unknown. Here, we employ protein NMR experiments supported by mass spectrometry and crystallography to study HBGA binding to the P-domain of a prevalent virus strain (GII.4). We report a highly selective transformation of asparagine 373, located in an antigenic loop adjoining the HBGA binding site, into an iso-aspartate residue. This spontaneous post-translational modification (PTM) proceeds with an estimated half-life of a few days at physiological temperatures, independent of the presence of HBGAs but dramatically affecting HBGA recognition. Sequence conservation and the surface-exposed position of this PTM suggest an important role in infection and immune recognition for many norovirus strains.. ...

This study enabled an empirical assessment of a model carcinogen borne in soil and its potency on human receptors. SOS-lux biosensors had numerous practical advantages over the traditional assays including procedural simplicity, ease of measurement and in vivo analysis without cell disruption. A key advantage of the application of biosensors is their ability to be deployed to genuine samples. This means that while chromatographic analysis requires extraction, purification, clean-up and then analysis, the biosensor can be used in crude and turbid solutions. This could be exclusively for a carcinogenic sensor or for a wider suite of sensors for specific analytes and generic toxicity. Samples could be exposed to a suite of sensors simultaneously allowing a real time response that could prove valuable for the protection of target receptors at a timescale commensurate with intervention. The data presented confirm that the sensors could be fabricated to permit compact analysis and the use of disposable

I recently heard a radio show where two callers claimed that their blood types had changed. One caller claimed that he was born O-, but recent tests said he was A+. Another caller claimed to change from AB- to AB+. Both callers claim that they had multiple tests before and after the change.. What I noticed is that both cases claim a conversion from Rh- to Rh+, or from type O to some other type. These could potentially be explained by a person carrying a gene for Rh factor or some other blood type, but failing to express the protein. Then the gene starts expressing, changing their blood type.. Of course both stories could be false, intentionally or otherwise. This is purely anecdotal evidence, and one caller claimed that her blood type change was due to extraterrestrial interference. I couldnt find any better documented cases of blood type changes after a quick search on the internet, so I thought Id ask here.. Are there any documented cases of a change in blood type?. ...

Feline calicivirus (FCV) is a major causative agent of respiratory disease in cats. It is also one of the few cultivatable members of the family Caliciviridae. It has recently been reported that FCV binding is in part due to interaction with junction adhesion molecule-A. This report describes the characterization of additional receptor components for FCV. Chemical treatment of cells with sodium periodate showed that FCV recognized carbohydrate moieties on the surface of permissive cells. Enzymic treatment with Vibrio cholerae neuraminidase demonstrated that sialic acid was a major determinant of virus binding. Further characterization using linkage-specific lectins from Maackia amurensis and Sambucus nigra revealed that FCV recognized sialic acid with an α2,6 linkage. Using various proteases and metabolic inhibitors, it was shown that α2,6-linked sialic acid recognized by FCV is present on an N-linked glycoprotein.

By engaging SIRPα, CD47 limits the ability of macrophages to engulf tumor cells, which acts as a major phagocytic barrier. In this study, we developed an exosome-based immune checkpoint blockade that antagonizes the interaction between CD47 and SIRPα. These exosomes harboring SIRPα variants (SIRPα-exosomes) were sufficient to induce remarkably augmented tumor phagocytosis, lead to prime effective anti-tumor T cell response. Given that clustering of native CD47 provides a high binding avidity to ligate dimerized SIRPα on macrophage, nature-derived exosomes could be appreciable platform to ...
In the few years since the beginning of glycomics in 21th century, many achievements have been made in the study of glycomics, especially in the discovery of sugar genes and the elucidation of the new structure of sugar chain. Although glycomics research has made remarkable achievements in these fields, there are many unknown intermediate links between sugar genes and sugar chains, sugar chains and glycan binding proteins, and there is a general lack of detailed research on the relationship between structure and function.. These fields are not only related to the progress of glycomics itself, but also directly related to the progress of proteome planning, so it has become a major bottleneck in the development of life science.. The advancement of analytical techniques has always been a direct driving force in the development of glycomics. It is the rapid development and wide application of modern instrument analysis technology that lead to the overall prosperity of glycomics research, and inject ...

This is a Phase 1, dose escalation study of EC0489 administered by intravenous bolus (IV) during weeks 1 and 3 of a 4-week cycle in PART A and weekly on a 4-week cycle in PART B.. Both Parts are open to patients with refractory or metastatic cancer who have exhausted standard therapeutic options. EC0489 is a drug that is specifically designed to enter cells via a folate vitamin receptor. Experimental evidence shows that the target receptor is over-expressed in many human cancers. There are no previous human studies of EC0489; however, lab research (research in test tubes or animals) using EC0489 has shown activity against tumors in animals. This activity in animal models suggests that EC0489 may be useful as chemotherapy against human cancers. The primary objective of this study is to determine the safety and maximum tolerated dose of EC0489 given by intravenous bolus. The efficacy of treatment will also be measured. ...
