The November 2012 podcast features an interview with Michel Scholtz from GSI Darmstadt, Germany, senior author of the paper entitled Modeling Cell Survival after Photon Irradiation Based on Double-Strand Break Clustering in Megabase Pair Chromatin Loops appearing in the November 2012 issue of Radiation Research. The interview is hosted by Massimo Pinto and touches on the research efforts of Michael Scholtz, and his colleagues, who have been focused for a long time toward a deeper understanding of the biological effects of high linear energy transfer radiations that are used in cancer therapy with accelerated ion beams. Mutation rates at two expanded simple tandem repeat loci were studied in the germ line of first- and second-generation offspring of inbred male CBA/H, C57BL/6, and BALB/c mice exposed to either high linear energy transfer fission neutrons or low linear energy transfer x-rays. Paternal CBA/H exposure to either x-rays or fission neutrons resulted in increased mutation rates in the germ line of two subsequent generations. Comparable transgenerational effects were observed also in neutron-irradiated C57BL/6 and x-irradiated BALB/c mice. The levels of spontaneous mutation rates and radiation-induced transgenerational instability varied between strains (BALB/c,CBA/H,C57BL/6). Pre- and postmeiotic paternal exposure resulted in similar increases in mutation rate in the germ line of both generations of CBA/H mice, which together with our previous results suggests that radiation-induced expanded simple tandem repeat instability is manifested in diploid cells after fertilization. Previous studies by our group and others have shown that Artemis deficient cells exhibit elevated radiation sensitivity, yet have only a slight defect in DSB repair, implicating a role for Artemis in the repair of a specific class of DSBs[6, 13, 16]. Radiation-induced breaks consist of a combination of closely spaced single-strand breaks, DSBs, sugar damage, base damage and breaks with modified ends. Only specific subsets of these breaks are thought to be processed by Artemis, with other proteins apparently unable to substitute for Artemis in this function[10, 11, 13]. In G1 cells, the majority of radiation-induced DSBs are rejoined quickly by NHEJ and this rapid repair appears not to require Artemis or ATM, however a small subset of slow repairing damages have been shown to require ATM and Artemis for repair[13, 27, 28]. PURPOSE In an effort to assess the characteristics of mutation induced by different linear energy transfer (LET) radiation in higher plants, the mutational effects of carbon-ion beams and gamma-rays were investigated in Arabidopsis. MATERIALS AND METHODS The rpsL (Escherichia coli ribosomal protein small subunit S12) transgenic Arabidopsis (Arabidopsis/rpsL) mutation detection system was adopted. Dry seeds of Arabidopsis/rpsL were irradiated with gamma-rays and 208-MeV carbon ions (208-MeV (12)C(5+)), and the mutation frequency and mutation spectrum were examined. RESULTS The frequency of mutant clones increased following irradiation with 208-MeV (12)C(5+) and gamma-rays. Mutation spectrum analysis showed that G:C to A:T transitions and |2 bp deletions/insertions were significantly induced by both 208-MeV (12)C(5+) and gamma-rays. -1 and -2 frameshift mutations were characteristic in the gamma-ray irradiated group. For low LET radiation, accumulation of the same dose over weeks or months, however, is expected to reduce the lifetime risk appreciably, possibly by a factor of 2 or more. The Committees estimated risks for males and females are similar. The risk from exposure during childhood is estimated to be about twice as large as the risk for adults, but such estimates of lifetime risk are still highly uncertain due to the limited follow-up of this age group. . . . . . However, not all patients respond to treatment and most patients experience hematological toxicity. α-emitters, however, have a shorter pathlength and higher linear energy transfer allowing them to be more potent agents. There were tons of bands playing extremely metallic hardcore, but other than 18 Visions, Earth Crisis, Bloodlet and a handful of others, they were pretty rough around the edges. Thats not necessarily a bad thing, but VOD were about a zillion times more polished and "professional" than anybody else in the hardcore scene, and they turned a lot of heads with this one.. Now here I am again wondering why me? Why another difficult baby?
