Expression of CD137, a member of the tumor necrosis factor receptor family, is inducible on vascular endothelial cells by proinflammatory stimuli. Its ligand is expressed as a transmembrane protein on the surface of monocytes, and transmits activating signals into monocytes (reverse signaling) inducing monocyte migration. These findings led to the hypothesis that CD137 expression on activated endothelial cells facilitates recruitment of monocytes into inflammatory tissues including atherosclerotic lesions. Data from this study demonstrate that CD137 expression is inducible on endothelial cells by TNF stimulation. Recombinant CD137 protein and CD137 expressed on activated endothelial cells enhance ICAM-1-mediated adhesion of monocytes under defined flow conditions in vitro. The velocity of rolling leukocytes provides important information about the inflammation process, which aids biomedical researchers in the development of anti-inflammatory medications. In the application, cells are detected in the first video frame and then tracked through subsequent frames. Detection is accomplished by computing for every pixel in the frame the maximal Gradient Inverse Coefficient of Variation (GICOV) score across a range of possible ellipses. The GICOV score for an ellipse is the mean gradient magnitude along the ellipse divided by the standard deviation of the gradient magnitude. The mechanisms of leukocyte recruitment have been studied extensively in vitro and have shed light on the basic molecular structure-function relationship of adhesion and signaling molecules involved in this essential immune response. This review will summarize how these in vitro observations extend to leukocyte behavior in inflamed blood vessels in the microcirculation. We highlight physiological results that might not have been predicted from in vitro systems. Special attention is placed on the physiology of rolling, adhesion, and intralumenal crawling in blood vessels. The importance of the glycocalyx, secondary tethers, shear, and the microenvironment are discussed. Selectin P ligand, also known as SELPLG or CD162 (cluster of differentiation 162), is a human gene. SELPLG codes for PSGL-1, the high affinity counter-receptor for P-selectin on myeloid cells and stimulated T lymphocytes. We tagged 82 lactating northern fur seals (Callorhinus ursinus) with tri-axial accelerometers and magnetometers on two eastern Bering Sea islands (Bogoslof and St. Paul) with contrasting population trajectories. Using depth data, accelerometer data and spectral analysis we classified time spent diving (30%), resting (~7%), shaking and grooming their pelage (9%), swimming in the prone position (~10%) and two types of previously undocumented rolling behavior (29%), with the remaining time (~15%) unspecified. The reason for the extensive rolling behavior is not known. We ground-truthed the accelerometry signals for shaking and grooming and rolling behaviors-and identified the acceleration signal for porpoising-by filming tagged northern fur seals in captivity. Speeds from GPS interpolated data indicated that animals traveled fastest while in the prone position, suggesting that this behavior is indicative of destination-based swimming. It is widely known that stress conditioning can protect microcirculation and induce the release of vasoactive factors for a period of several hours. Little, however, is known about the long-term effects of stress conditioning on microcirculation, especially on the microcirculation of the periosteum of the calvaria. For this reason, we used intravital fluorescence microscopy to investigate the effects of heat shock priming on the microcirculation of the periosteum over a period of several days. Fifty-two Lewis rats were randomized into eight groups. Six groups underwent heat shock priming of the periosteum of the calvaria at 42.5°C, two of them (n = 8) for 15 minutes, two (n = 8) for 25 minutes and two (n = 8) for 35 minutes. After 24 hours, a periosteal chamber was implanted into the heads of the animals of one of each of the two groups mentioned above. To distinguish which one is more suitable for the melt crystallized PA46, we prepared double oriented samples of PA46 by mechanical rolling at 180 °C followed by annealing at 260 °C. The samples showed the typical α phase diffractions. Combining the two-dimensional (2D) wide-angle X-ray diffraction (WAXD) patterns obtained with the X-ray beam perpendicular and parallel to the chain axis, we concluded that PA46 adopted the monoclinic structure that was determined by Atkins based on the electron diffraction of single crystals grown from solution [1].
