Blood transfusions can be life-saving, however, adverse effects occur. Some adverse effects are attributed to cytokine accumulation in blood products during storage. Interleukin-8 (IL- 8), interleukin-1β (IL-1β), and tumour necrosis factor-α (TNF-α), are cytokines that have been shown to accumulate during storage in human packed red blood cells (PRBC). Leukoreduction removes leukocytes from blood products and thus prevents cytokine accumulation. A recent study in dogs showed that IL-8 accumulated in canine PRBC collected from non-anaesthetised dogs, and leukoreduction prevented this accumulation. At our institution, blood is collected from anaesthetised terminal donors in haemorrhagic shock. We were thus interested to investigate the effect of this method of blood collection on cytokine accumulation in stored canine PRBCs. Acute haemorrhage in rats, pigs and people has been shown to increase inflammatory cytokines. It is unknown if similar increases occur in dogs with haemorrhagic shock, or ...
Ischemia-reperfusion injury, a form of sterile inflammation, is the leading risk factor for both short-term mortality following pulmonary transplantation and chronic lung allograft dysfunction. While it is well recognized that neutrophils are critical mediators of acute lung injury, processes that guide their entry into pulmonary tissue are not well understood. Here, we found that CCR2+ classical monocytes are necessary and sufficient for mediating extravasation of neutrophils into pulmonary tissue during ischemia-reperfusion injury following hilar clamping or lung transplantation. The classical monocytes were mobilized from the host spleen, and splenectomy attenuated the recruitment of classical monocytes as well as the entry of neutrophils into injured lung tissue, which was associated with improved graft function. Neutrophil extravasation was mediated by MyD88-dependent IL-1β production by graft-infiltrating classical monocytes, which downregulated the expression of the tight ...

The finding that TRAP is especially pronounced in T cells is remarkable, because T cells do not express TLR75 and do not respond directly to R-848 in the cell culture (data not shown). Thus, although the general effect of PBL depletion by R-848 is similar to the action of a well-known immunosuppressant, FTY720, the mechanisms involved must be different. FTY720 has been shown recently to inhibit the emigration of lymphocytes from lymphoid tissue by down-regulating a sphingosine-1-phosphate receptor, which lymphocytes need to leave lymphatic tissues.45 Instead, our data suggest that the endothelium is important for mediating TRAP. This assumption is based on the following observations: (1) In the presence of selectin blockade adoptively transferred leukocytes remained attached to the major vascularized organs (also nonlymphatic) in mice receiving R-848 (Figure 3), while in PBS-treated mice the leukocytes showed general homing to the spleen, which is known to be independent of selectin function.46 ...
In 2004, prestorage delays and CA storage were compared for occurrence of disorders. Fruit were harvested at a starch index of 5.9. Fruit were exposed to either a 2- or 5-day prestorage delay at 17 °C; or placed immediately into cold storage (control) at 0.5 °C. An additional treatment was CA storage at 2.5 °C. In February, occurrence of soft scald, soggy breakdown, and bitter pit were measured on 40 fruit per replication. Fruit were from `Honeycrisp/M.26 trees planted in 1994. Treatments were replicated five times with four trees in each replication. Soft scald was very severe in this year, with 84% of control fruit being affected. Two-day prestorage delay reduced it to 48% and 5-day delay to 21%. Soggy breakdown was also severe with 14% of the fruit being affected. Two- and 5-day delays had no effect on occurrence of soggy breakdown, but CA storage increased it to 65%. Bitter pit was very rare and not affected by any of the treatments. These results demonstrate that in severe cases, ...

Krieg et al. used high-dimensional single-cell mass cytometry with optimized immune marker panels and several bioinformatics tools to identify differences between responders and non-responders to anti-PD-1 immunotherapy. Before treatment, metastatic melanoma patients who would ultimately benefit from anti-PD-1 had reduced levels of T cells and increased levels of myeloid cells in their peripheral blood. The frequency of classical CD14+CD16-HLA-DRhi monocytes was identified as a possible predictive biomarker of response to anti-PD-1.

Downloadable! Social environments constitute a framework in which it is possible to study how groups of agents interact in a society. The framework is general enough to analyse both non-cooperative and cooperative games. We identify a number of shortcomings of existing solution concepts that are used for social environments and propose a new concept called social rationalizability. The concept aims to identify the consequences of common knowledge of rationality and farsightedness within the framework of social environments. The set of socially rationalizable outcomes is shown to be non-empty for all social environments and it can be computed by an iterative reduction procedure. We introduce a definition of coalitional rationality for social environments and show that it is satisfied by social rationalizability(This abstract was borrowed from another version of this item.)
The recent explosion of data set size, in number of records and attributes, has triggered the development of a number of big data platforms as well as parallel data analytics algorithms. At the same time though, it has pushed for usage of data dimensionality reduction procedures. Indeed, more is not always better. Large amounts of data might sometimes produce worse performances in data analytics applications.
Whole blood is blood that contains red blood cells, white blood cells, platelets and plasma. A whole blood donation is divided into components, such as red blood cells and platelets that are used to treat different types of patient needs. All whole blood donations undergo leukoreduction to remove white blood cells, which reduces the risk of serious immune reactions that recipients may have when exposed to donor white cells. The volume of a single whole blood donation is about 480 ml or less than 10 percent of a donors total blood volume.
A particle/cell separation device is described which is particularly adapted for neutrophil depletion from a preparation of whole blood or platelet-rich plasma. Also described are blood and platelet rich plasma compositions produced using the device which are neutrophil-depleted.

Second Law of Thermodynamics:. $$dS = \frac{\delta Q}{T}$$. First Law of Thermodynamics:. $$dU = \delta Q + \delta W = \delta Q - PdV_m$$. Definition of Enthalpy:. $$dH = dU + d(PV_m) = dU +PdV_m + V_mdP$$ Substitutions:. $$dH = \delta Q + PdV_m + V_mdP - PdV_m = \delta Q +V_mdP$$ $$\delta Q = dH - V_mdP$$. Perfect gas equation (notice that I am using molar volume $V_m$, so we will be calculating molar entropy change):. $$PV_m = RT$$ $$V_m = \frac{RT}{P}$$. At constant pressure (Yes, I know pressure changes, bear with me. Well put that back in).. $$dH = C_p dT$$. Substitute to get rid of $V_m$, since we dont have volume data.. $$dQ = C_p dT - \frac{RT}{P}dP$$. Substitute into the second law:. $$dS = C_p \frac{dT}{T} - R\frac{dP}{P}$$. Integrate (now we account for change in pressure:. $$\int_{S_1}^{S_2} dS = C_p \int_{T_1}^{T_2} \frac{dT}{T} - R \int_{P_1}^{P_2} \frac{dP}{P} $$. $$\Delta S = C_p \ln \left(\frac{T_2}{T_1}\right) - R \ln \left(\frac{P_2}{P_1} \right)$$. The type I interferons (IFN) are a family of cytokines with pleiothropic activities that include inhibition of viral replication, cell proliferation and activation of the immune system. These properties give the IFNs important physiological and pathological roles in infection and cancer and have led to their therapeutic use for many clinical conditions. In humans, the type I IFNs consist of 12 different IFNa subtypes as well as single IFNb, w and k subtypes. They all compete for binding to a common receptor, consisting of two subunits, IFNAR1 and IFNAR2. In almost all cell types proliferation is inhibited by IFNs as a consequence of the antiviral properties. However, previous studies on human peripheral B-lymphocytes have shown increased survival as well as proliferation upon IFN treatment. We established a purification system for extraction of B-lymphocytes from buffy-coat, utilizing density centrifugation in combination with anti-CD19 magnetic beads. A study done by Paul Chasan, M.D., FACS, and published in Aesthetic Plastic Surgery, indicates a procedure called reductive augmentation is an effective method for achieving a more round-appearing breast.. Even though advanced breast reduction techniques generally lead to a high level of patient satisfaction, limitations still exist. Over time, there is typically less roundness and fullness at the upper area of the breast. The reductive augmentation method helps maintain a full, round breast appearance.. Reductive augmentation (RA) is a one-stage procedure combining breast reduction with breast augmentation (implants). Though adding implants following a reduction may seem counterintuitive, Dr. Chasan points out that when tissue is removed without adding implants, breasts may eventually sag. The RA procedure can also correct breast asymmetry, and address drooping at the bottom of the breast following earlier augmentations.. For Chasans study, 35 women underwent RA; 12 were primary surgeries, and ...

Two main modifications of Holmess method are described. In both the amount of silver nitrate in the impregnating bath is increased, and larger quantities of higher pyridine derivatives are substituted for pyridine. The preferred method for general purposes employs 2.4% 2:6 lutidine at pH 8.4, and gives a bluish picture with improved selectivity of fibre staining. The use of 1.6% 2:4:6 collidine gives a reddish picture, with highly selective fibre staining, and sharp resolution of neurosecretory cells. Minor modifications of pre-treatment and reduction procedures are also described. ...
Q: Is it possible to have a vertical chin that points downward as a result of fleshy tissue and not bone? My chin is vertically too long for my face and projects downward especially when I smile. Id hate to have complications from having the bone burred down on the tip but would also hate to have titanium plates & screws in my chin from a wedge of bone being cut out to reduce the chin. I live in Texas and I dont know any surgeons here that are properly certified and experienced in chin reductions. Please let me know. Thanks.. A: A long vertical chin can be the result of too much bone, too much soft tissue, or a combination of both. In most cases, vertical chin excess is a combination of both. By definition, a chin that is too long because of bone must have extra soft tissue to go with it. Making the proper diagnosis helps one correct the right chin reduction procedure. Not properly addressing the right problem, or inadequately treating it, is the reason so many chin reductions have ...

Patrolling Monocytes Promote the Pathogenesis of Early Lupus-Like Glomerulonephritis Researchers found that glomerulonephritis was driven by TLRs but, remarkably, proceeded independently of immune complexes. Rather, disease in three different mouse models and patients with systemic lupus erythematosus was characterized by glomerular accumulation of patrolling monocytes, a cell type with an emerging key function in vascular inflammation. In this study, we focused on the role of neutrophils in the recruitment of DCs to the site of L. major inoculation, one main question addressed being whether neutrophils could transiently and locally contribute to the Langherans cell/dermal dendritic cell trafficking as well as to the recruitment of monocytes, the latter being capable to be programmed to either macrophages or monocyte-derived DCs. Our results demonstrate a previously unappreciated role of primary neutrophil extravasation, and of the CCL3 released by neutrophils in the rapid recruitment and trafficking of LC/dDC as well as of monocyte-derived DC to the site of L. major inoculation. We report here for the first time that the selective secretion of CCL3 by neutrophils is critical in vivo for the recruitment of DCs to the site of Leishmania inoculation, as revealed by markedly reduced recruitment of DCs in mice with pharmacological neutralization or absence of CCL3, and in neutrophil-depleted mice. This decrease was restored in ...

Background and objectives: Platelet storage lesions (PSL) are the sum of deleterious changes that platelets undergo during storage, affecting platelet concentrate viability for transfusion. In veterinary medicine, transfusion medicine has gained popularity and recent advances have been made. There is a continuous effort to prolong the shelf life and improve the quality of platelet concentrates. Removal of leukocytes has been proven to reduce febrile post-transfusion reactions and decrease platelet premature activation and apoptosis in humans. The present study objectives were to determine if leukoreduced canine platelet concentrates have fewer and delayed onset of in vitro storage lesions compared with non-leukoreduced concentrates and to determine which variable best predicts in vitro platelet viability. Methods: Platelet concentrates from twelve dogs were stored between 22°C - 24°C under continuous agitation for 7 days. Each sample was equally divided into leukoreduced (LR), leukoreduced ...

Light-evoked intracellular voltage noise records have been obtained from Limulus eccentric cells, from threshold light intensity to an intensity .10(5) times threshold. These data are analyzed in terms of a simple adapting-bump noise model. It is shown how the model yields a data reduction procedure that slightly generalizes the familiar use of Campbells theorem for Poisson shot noise: the correlative effect of adaptation amends Campbells theorem by a single multiplicative factor, which may be estimated directly from the power spectrum of the noise data. The model also permits direct estimation of the bump shape from the power spectrum. The bump shape estimated from noise at dim light is in excellent agreement with the average shape of bumps observed directly in the dark. The data yield a bump rate that is linear with light up through about 50 times threshold intensity but that falls short of linearity by a factor of 35 at the brightest light. The bump height decreases as the -0.4 power of ...

The link between cancer progression and inflammation is now well established (1). Numerous lines of investigation have shown that inflammatory cells are key participants in cancer progression and metastasis (42). This study is, to our knowledge, the first to identify a functional role for neutrophils in the adhesion of CTCs in the liver. Neutrophil depletion resulted in a significant decrease in both gross metastases using an intrasplenic model of liver metastasis. Activated neutrophil infusion 10 minutes before the administration of cancer cells resulted in increased adhesion of H-59 cells in another model of metastasis using intravital microscopy. Again at 72 hours postinjection, neutrophil depletion caused a profound reduction in microscopic liver metastases, which was rescued when activated peripheral blood neutrophils were admixed to the cancer cell inoculum. These results indicate that early interactions between neutrophils and cancer cells within sinusoids of the liver promote cancer ...
I think I am going to devote an entire blog series to the topic of ideas I had from a pin. Many times I find a pin that sparks an idea. The pin itself may not have been exactly what I would replicate… but the idea takes on a path and journey of its own.. So, in honor of all of the teachers and educators who follow this blog, I thought I would share and adapt a pin that I found two days ago. A truly sentimental and adorable idea a photographer mother shared. Her daughter was turning 8 and the class used the SMART board to list one adjective that described the birthday girl. I can only imagine how special and uplifting this was for the child as well as touching for the mother. The final product was the picture of the child with all of the adjectives floating around her. Truly, what an amazing keepsake!. ...
