The host and tissue specificity of retrovirus infection is largely determined by specific cellular receptors that mediate virus entry. Genes encoding these receptors are widely distributed in the genome, and the receptors identified to date show no sequence similarity. We have identified the cellular receptor for amphotropic murine retroviruses, Ram-1, by screening a rat cDNA expression library introduced into amphotropic virus-resistant hamster cells. The 656-amino acid receptor is homologous to the gibbon ape leukemia virus receptor at both hydrophobic termini but is highly divergent in the central hydrophilic region. Both receptors appear to be integral membrane proteins having multiple membrane-spanning regions. Identification of this family of receptors will help define the evolutionary relationship between retroviruses and their cellular receptors.

There have been many examples of virus-glycoprotein pairs that do not efficiently produce infectious pseudotyped virus together. For example, HIV-1 does not pseudotype efficiently with Rous sarcoma virus (RSV) Env, influenza virus hemagglutinin, or lymphocytic choriomeningitis glycoprotein (4, 16). The high pathogenicity of the Ebola virus reflects multiple concurrent processes on infection. Among other important determinants, Ebola fusogenic glycoprotein (GP) has been associated with the detachment of infected cells and eventually leads to vascular leakage and haemorrhagic fever. Here we report that the membrane-anchored GP is sufficient to induce the detachment of adherent cells. Objective To investigate the clinical effect of sevoflurane general anesthesia in the treatment of sepsis patients. Methods 74 patients with sepsis who underwent surgical treatment in the Department of Surgery in our hospital from March 2016 to February 2017 were divided into two groups, according to the different ways of maintaining anesthesia. All patients were given general anesthesia. The control group was maintained with propofol during operation. The observation group was maintained with sevoflurane. The changes in vital signs, changes in respiratory function indicators, adverse reactions and surgical outcome were compared between the two groups. Results The mean arterial pressure and heart rate after anesthesia in the observation group were controlled more smoothly than those in the control group,and the difference was statistically significant(P 0.05). Inefficient gene delivery continues to be a primary hurdle facing gene therapy. Viruses offer the highest gene transfer capabilities but are not optimized as therapeutics. Applying directed evolution, we randomly mutated the entire genome of amphotropic murine leukemia virus (MLV) and selected for improved stability and infection at 37°C. After one round of mutagenesis and several rounds of selection, we isolated MLV variants with double the half-life of wild-type MLV. The improved stability of the mutant MLV leads to increased virus production, titer, and infection efficiency. Remarkably, a single mutation in the protease (PR), G119E, in the MLV gag-pro-pol is responsible for the enhanced stability.
