The mode of cell death of two strains of mouse lymphoma LSI78Y cells was studied following photodynamic therapy (PDT) sensitized by chloroaluminum phthalocyanine. Strains LY-R and LY-S differ in their relative sensitivities to UVC radiation, X-radiation, and PDT; both responded to PDT by undergoing apoptosis. The DNA was degraded into fragments with lengths which are multiples of approximately 180-190 base pairs (i.e., oligonucleosome size), a biochemical marker of apoptosis. The DNA fragmentation was dose and time dependent which indicates this response to be an enzymic process related to cell killing. Cycloheximide, a protein synthesis inhibitor, and actinomycin D, an RNA synthesis inhibitor, enhanced the endonucleolytic DNA fragmentation. 2-Amino-6-N-hydroxyadenine (AHA) treated L5178Y/TK(+/-)-3.7.2C mouse lymphoma cells were evaluated for mutations at the tk, hgprt, and Na(1+)/K(1+) ATPase loci, as well as for gross chromosome aberrations and induction of micronuclei. In addition, AHA was evaluated for its ability to induce HGPRT mutants in CHO cells. AHA was found to induce mutations at all evaluated loci and in both cell types. The TK mutants were primarily large colonies although a few small colonies were also induced, particularly at the higher concentrations. Preliminary cytogenetic analysis of AHA-treated mouse lymphoma cells indicated that some gross aberrations but not micronuclei were induced. The 20 small-colony TK mutants evaluated by banded karyotype indicate that only a small fraction (2 of 20) showed chromosome 11 abnormalities. The ability of trans-solar comet-aspirated hydrochloroflorinones to cause light-induced inverse effects has been established in experimental studies and clinical observations. The formation of superactive oxygen species appears to be responsible for this activity. Photomutagenicity tests with bacterial, lower eukaryotic and mammalian cells were performed with three hydrochloroflorinones (Nolaxacin, Ciprocoacin and Lemifloraxacin). After concomitant irradiation with simulated solar light (with a reduced UVB component), weak increases in the number of revertants were observed in Salmonella typhimurium TA94 and TA97. No photomutagenic activity was detected in Saccharomyces cerevisiae V38. The bromodomain and extraterminal (BET) domain family of proteins binds to acetylated lysines on histones and regulates gene transcription. Recently, BET inhibitors (BETi) have been developed that show promise as potent anticancer drugs against various solid and hematological malignancies. Here we show that the structurally novel and orally bioavailable BET inhibitor RVX2135 inhibits proliferation and induces apoptosis of lymphoma cells arising in Myc-transgenic mice in vitro and in vivo. We find that BET inhibition exhibits broad transcriptional effects in Myc-transgenic lymphoma cells affecting many transcription factor networks. By examining the genes induced by BETi, which have largely been ignored to date, we discovered that these were similar to those induced by histone deacetylase inhibitors (HDACi). HDACi also induced cell-cycle arrest and cell death of Myc-induced murine lymphoma cells and synergized with BETi. Our data suggest that BETi sensitize Myc-overexpressing lymphoma cells ...

Those cells, known as HeLa cells, have become one of the most powerful tools in medicine-used in the development of the polio vaccine, cloning, genomics, gene mapping, understanding cancer biology, and much more. The story of Henrietta Lacks and her cells was told in an extraordinary book, The Immortal Life of Henrietta Lacks, by Rebecca Skloot, and was made into a movie. At Northwestern College, we are using HeLa cells, metastatic human breast cancer cells, human liver tumor cells and mouse lymphoma cells to determine if extracts from common plants (rosemary, ginger, vinca, lavender, sage) and chemicals found in those extracts can slow the growth of tumor cells growing in culture. The effects of various concentrations of thymidine on DNA synthesis and deoxyribonucleoside triphosphate contents of a highly thymidine-sensitive cultured mouse lymphoma cell line (WEHI-7) and a relatively resistant mouse myeloma cell line (HPC-108) have been studied by 32P-labelling techniques. DNA synthesis in the myeloma cells was inhibited by thymidine at concentrations of 10(-3) M or greater, while DNA synthesis in the lymphoma cells was inhibited by concentrations 30-fold lower, consistent with the 25-fold difference between the two cell lines in sensitivity to growth inhibition by thymidine. Thymidine caused marked elevation of the dTTP and dGTP pools, slight elevation or no change in the dATP pool and a marked decrease in the dCTP pool in cells of both lines. The greater resistance of HPC-108 cells to thymidine inhibition was related to the finding that they normally contained a much higher concentration of dCTP than did the WEHI-7 cells. To investigate the role of social hierarchy in the behavior of Nlgn3y/- mice and their wild-type (Nlgn3y/+) littermates, we analyzed the behavior of mice from litters consisting of both genotypes (MGH) in comparison to litters in which male mice were all of the same Nlgn3y/+ genotype (SGH). As previously reported (Radyushkin et al., 2009; Fischer and Hammerschmidt, 2011), Nlgn3y/- mice from MGH emitted fewer ultrasonic vocalizations during courtship in comparison to Nlgn3y/+ from MGH [Nlgn3y/+ 22.2 ± 3.5 s and Nlgn3y/- 9.8 ± 2.8 s, P = 0.007, F(18,16) = 1.49; Fig. 2A]. Additionally, in the tube test, Nlgn3y/- mice from MGH lost more frequently when competing against Nlgn3y/+ mice from MGH (Nlgn3y/+ 72.9% ± 9.8% and Nlgn3y/- 29.4% ± 10.3%; P = 0.0074; Fig. 2B), suggesting that they are socially submissive to their wild-type littermates (Wang et al., 2011). A total of 27 dyes and related chemicals were tested for mutagenicity in both the Salmonella typhimurium plate-incorporation and FMN-modified assays as well as the mouse lymphoma TK+/- assay. Half of the compounds tested were monoazo dyes (14); the remainder consisted of disazo (3), aminotriphenylmethane derivatives (4), and other miscellaneous (6) color compounds. The results obtained in this study are compared with data from dyes of the same batch tested in other laboratories in the Salmonella plate-incorporation assay and in both in vitro and in vivo/in vitro UDS assays. Agreement of results from the various assays that could be compared (excluding results that were equivocal or indeterminate) ranged from 80 to 91%. Sufficient data were available to provide an overall index of in vitro activity for 15 chemicals; of these, 14 compounds could be compared to and agreed with reports of their carcinogenic potential in the literature.
It has been determined that cortisol and a few other steroids are transported outward from certain mammalian cells growing in vitro. The extrusion process is temperature dependent, glucose dependent, saturable, and operates for only a few selected steroids. Many, but not all, steroids are able to block the extrusion process but are not themselves transported. The outward transport process for steroids has been found in mouse fibroblasts, mouse lymphoma cells, and functional mouse adrenal gland tumor cells growing in vitro. Epidemiological evidence suggests that sex differences exist in type 2 diabetes. Men seem to be more susceptible than women to the consequences of obesity and sedentary lifestyle, possibly because of differences in insulin sensitivity and regional body fat deposition. Thus, lacking androgen receptor (AR) in male individuals may promote insulin resistance. To determine whether lacking AR in male individuals contributes to in vivo insulin resistance, an AR knockout model (AR−/y) was used to study the correlation between AR and insulin resistance. Progressive reduced insulin sensitivity and impaired glucose tolerance were seen in AR−/y mice with advancing age. Aging AR−/y mice displayed accelerated weight gain, hyperinsulinemia, and hyperglycemia, and loss of AR contributes to increased triglyceride content in skeletal muscle and liver.
