Acetyl-l-carnitine (ALC) is a naturally occurring substance that, when administered at supra-physiological concentration, is neuroprotective. It is involved in membrane stabilization and in enhancement of mitochondrial functions. It is a molecule of considerable interest for its clinical application in various neural disorders, including Alzheimers disease and painful neuropathies. ALC is known to improve the cognitive capability of aged animals chronically treated with the drug and, recently, it has been reported that it impairs forms of non-associative learning in the leech. In the present study the effects of ALC on gene expression have been analyzed in the leech Hirudo medicinalis. In the medicinal leech, Hirudo medicinalis, chemical stimuli such as blood are known to increase locomotor activity - swimming and crawling. Hirudotherapy has been complicated by infections caused by Aeromonas spp., which are considered endosymbionts of the leech. Leeches were treated by feeding them artificially with 100μg/ml ciprofloxacin using a 0.05 Mol arginine solution as a phagostimulant. Aeromonads were identified using the API 20NE system, and species determined by gyrB sequencing of two representative isolates. Aeromonas spp. were detected in 57 out of 80 control leeches (71.3%), but in none of the 56 leeches treated with ciprofloxacin. Treated leeches survived for up to four months. Tested weekly, leeches took human blood for at least 4 weeks after treatment and were all negative for Aeromonas spp. All water samples in which leeches were kept before treatment were contaminated with Aeromonas spp. but in none of the NaCl/arginine solutions that were used to feed antibiotic treated leeches. The study of synapses is a relevant topic for all living organisms, as our bodies are coordinated by billions of successful nerve connections. Multiple cells often synapse onto one target cell, begging the question of how specific circuits then operate. What allows the target cell the ability to utilize both innervations? The leech, Hirudo verbana, is a useful model to study these questions. The heart tubes are innervated by two neurons, the heart excitatory and heart accessory neurons. Heart excitatory neurons originate at segments 3 through 18 within the leech, while heart accessory neurons originate at ganglia 5 and 6 and must therefore project axons intersegmentally. By labeling the germinal plates of staged leech embryos with monoclonal antibodies to the immunoglobulin superfamily member Tractin, we have documented the distribution and initial development of peripheral neurons in a hirudinid leech. We find, in addition to sensillar and extrasensillar sensory neurons, that there are 21 identifiable peripheral neurons in each hemisegment. These neurons are found in highly stereotyped positions, and all but two of them are associated with the segmental nerves. We show that eight of the peripheral neurons have the characteristic morphology of stretch receptor neurons and that they form a circumferentially distributed grid aligned in such a way that each of five specialized longitudinal muscle fascicles are monitored by at least two stretch receptor cells covering ventral, lateral, and dorsal regions of the body wall. We have cloned two innexins, Hm-inx1 and Hm-inx2, from the leech Hirudo medicinalis and shown that these have different expression patterns in the CNS. Hm-inx1 is neuronal-specific, whereas Hm-inx2 is glial-specific. We demonstrate that each of these innexins can form functional homotypic gap junctions in the paired Xenopus oocyte assay and that they can cooperate to form heterotypic junctions. Each of these junctions demonstrates novel properties not previously seen in this family. P. papillifera is a blood-feeder, feeding on turtles (and sometimes alligators!). This leech was found under a rock in Savannah River in South Carolina. During the reattachment of severed fingers and ears, or of the detached scalp, the blood flow needs to be reestablished. This is achieved by reconnecting the major arteries and veins. Here we describe the draft genomes of the owl limpet (Lottia gigantea), a marine polychaete (Capitella teleta) and a freshwater leech (Helobdella robusta), and compare them with other animal genomes to investigate the origin and diversification of bilaterians from a genomic perspective. Invertebrate-derived DNA (iDNA) from terrestrial haematophagous leeches has recently been proposed as a powerful non-invasive tool with which to detect vertebrate species and thus to survey their populations. However, to date little attention has been given to whether and how this, or indeed any other iDNA-derived data, can be combined with state-of-the-art analytical tools to estimate wildlife abundances, population dynamics and distributions. In 1920 Leach and Hamada made the journey half-way around the world to set up a pottery at St Ives in Cornwall. St Ives had long been an artists colony, and the pottery was built with the aid of a grant from the Handicrafts Guild.. The aim of the study was to evaluate the use of leeches of the genus Erpobdella as a means of assessing polychlorinated biphenyl contamination of watercourses. The River Skalice, heavily contaminated with PCBs, was selected as a model. The source of contamination was a road gravel processing factory in Rožmitál pod Třemšínem from which an estimated 1 metric ton of PCBs leaked in 1986. Levels of PCB were measured in leeches collected between 1992 to 2003 from 11 sites covering about 50 km of the river (the first sampling site upstream to the source of contamination and 10 sites downstream). The PCB indicator congeners IUPA no. 28, 52, 101, 118, 138, 153, and 180 were measured. Levels were highest at the four sampling sites nearest the source of pollution. Last Friday (Earth Day), waking up to a house full of sleeping people who wanted to remain so, I took an early morning bike ride, ending up downtown at The Pagoda. A previous visit was made here early in the life of this blog, but the blogger goes there fairly frequently. The Pagoda was a gift from Evergreen Shipping Lines to the city of Norfolk (thanks Chris for the correction), and an Oriental-style garden has been planted around it. When I first came to Norfolk in the late 70s, there was no pagoda or garden, rather this was the site of a giant, black, 500,000 gallon, molasses tank. The Pagoda was built on top of the tanks foundations and pillars, and today there is little evidence of its previous incarnation. Non-synaptic sensory cells, non-serotonergic pre- and postsynaptic partners and serotonergic neurons that do not form chemical synapses with the P cell failed to alter its responses to 5-HT. PCB signatures can be used for source identification, exposure studies, age dating and bio-monitoring. This study uses comprehensive two-dimensional gas chromatography with time-of-flight mass spectrometry (GCxGC-ToFMS) to produce a PCB signature comprised of over 80 PCBs for individual Leachs storm petrels (Oceanodroma leucorhoa). The Leachs storm petrel is a relatively small, elusive, understudied pelagic bird, which only returns to remote islands under darkness during the breeding season. Samples were obtained from 25 Leachs storm petrels found dead in Canada and the UK following storm events in 2006 and 2009. Tissue samples were extracted and analysed by GCxGC-ToFMS and results showed that 83 PCB congeners were present in ,60% of samples. An assessment of the PCB signature in four different tissue types showed that it did not vary greatly in samples obtained from the gut, heart, liver and stomach. Infectious haematopoietic necrosis (IHN) virus was isolated from freshwater leeches Piscicola salmositica and copepods Salmincola sp. removed from the gills of spawning sockeye salmon, Oncorhynchus nerka. This is the first report of the isolation of IHN virus from an animal other than salmonid fishes. High levels of IHN virus were also found in leeches taken from the bottom gravel of the spawning area. The prevalence of IHN virus in samples of individual leeches was as high as 100% and the virus was isolated from 95% of pooled samples of copepod and 1.5 × 108 pfu/g in the leech. The level of virus in leeches removed from fish gills was sometimes higher than the level of virus in the gill tissue itself.
